    Human hair follicles are self-renewing structures, which undergo three phases of hair cycle; anagen, catagen, and telogen. It is well known that Ξ²-catenin, a transducer of Wnt signaling, is critical in development, growth, and regeneration of hair follicle. In the absence of Wnt signal, Ξ²-catenin is ordinarily phosphorylated in the cytoplasm by glycogen synthase kinase (GSK)-3 and then degraded by proteasome. Therefore, inhibition of GSK-3 is able to increase Ξ²-catenin and would be able to induce growth of hairs. Valproic acid (VPA) is an anticonvulsant and mood-stabilizing drug used for decades and is known to inhibit the GSK-3Ξ² in cancer cell line and neuronal cells. However, the effect of VPA on hairs has not been studied yet. In this study, VPA enhanced the proliferation of human dermal papilla (DP) cells and outer root sheath (ORS) cells in vitro. In addition, hair follicles treated with VPA showed more rapid elongation of hairs and remained longer in anagen stage. In DP cells treated with VPA, Ξ²-catenin protein was found to be increased and accumulated in the nucleus, but the mRNA level of Ξ²-catenin was not increased. Instead, GSK-3Ξ² was phosphorylated at Ser9 by VPA, which means inactivation of GSK-3Ξ². These findings suggest that VPA increases Ξ²-catenin by preventing its degradation, not by promoting the synthesis. Anagen hair induction using mice model showed accelerated regeneration of new hair coat, higher anagen induction score and thicker back skin in VPA-treated mice. VPA also enhanced the de novo neogenesis after wounding, which has been known as Wnt-dependent process. In conclusion, the results from this study suggest that VPA stimulates growth and regeneration of hairs in both of in vitro and animal model.
