    Multicenter Evaluation of the Idylla GeneFusion in Non-Small-Cell Lung Cancer

    Targeted therapy in lung cancer requires the assessment of multiple oncogenic driver alterations, including fusion genes. This retrospective study evaluated the Idylla GeneFusion prototype, an automated and ease-of-use (<2 minutes) test, with a short turnaround time (3 hours) to detect fusions involving ALK, ROS1, RET, and NTRK1/2/3 genes and MET exon 14 skipping. This multicenter study (18 centers) included 313 tissue samples from lung cancer patients with 97 ALK, 44 ROS1, 20 RET, and 5 NTRKs fusions, 32 MET exon 14 skipping, and 115 wild-type samples, previously identified with reference methods (RNA-based next generation sequencing/fluorescence in situ hybridization/quantitative PCR). Valid results were obtained for 306 cases (98%), overall concordance between Idylla and the reference methods was 89% (273/306); overall sensitivity and specificity were 85% (165/193) and 96% (108/113), respectively. Discordances were observed in 28 samples, where Idylla did not detect the alteration identified by the reference methods; and 5 samples where Idylla identified an alteration not detected by the reference methods. All of the ALK-, ROS1-, and RET-specific fusions and MET exon 14 skipping identified by Idylla GeneFusion were confirmed by reference method. To conclude, Idylla GeneFusion is a clinically valuable test that does not require a specific infrastructure, allowing a rapid result. The absence of alteration or the detection of expression imbalance only requires additional testing by orthogonal methods

Due to the development of modern technology, these treatments can be offered to a wider group of patients. For many years, aortic root surgery consisted of mechanical conduit implantation and, therefore, necessitated life-long anticoagulation. At present, in patients with aortic root aneurysm and significant aortic valve regurgitation, it is possible to perform minimal-access valve sparing surgical procedures. The current paper is a brief description of the surgical technique for aortic root aneurysm surgery with preservation of the patientÔÇÖs own valve using the David procedure

    Increased brain 1H-MRS glutamate and lactate signals following maximal aerobic capacity exercise in young healthy males: an exploratory study

    Physical exercise involves increased neuronal activity of many brain structures, but 1H-MRS investigations on the effects of human brain glutamate (Glu) concentrations on acute exercise have been sparse. Previous studies consistently found increases in brain lactate (Lac) concentrations following graded exercise up to 85% of the predicted maximal heart rate. However, the reported effects on brain concentrations of glutamine and glutamate were not consistent. This study aimed to determine the effect of acute intense graded maximal exercise on 1H-MRS signals related to concentrations of Glu, glutamate+glutamine (Glx), and Lac. Young adult males were randomly divided into two groups and subjected to 1H-MRS when resting (NE) or shortly after cessation of the intense graded exercise intended to pass the anaerobic threshold (E). 1H-MRS spectra were acquired from the large voxel encompassing the occipito-parietal cortex only once. Estimates of Glu, Glx, and Lac concentrations were calculated in institutional units by normalizing to a spectroscopic signal originating from creatine-containing compounds (Cr). Concentrations of Glu, Glx, and Lac were respectively 11%, 12.6%, and 48.5% higher in E than in NE (p < 0.001). The increased brain Lac signal in the exercising group indicated that in our experiment, vigorous exercise resulted in passing the anaerobic threshold and lactate apparently entered the brain. Concomitantly glutamate-related resonance signals from the vicinity of the occipito-parietal cortex were significantly increased; physiological mechanisms underlying these phenomena require further study. Future studies should evaluate whether the normalization rate of these concentrations is a marker of general physical fitness

    Context. In 2020, the initial version of the Stellar Potential Perturbers Database (StePPeD) was presented with the aim to deliver up-to-date information on the stars and stellar systems that may perturb a long-period comet motion. We used the minimal distance between a star and the Sun as a selecting tool when compiling a list of interesting objects with close encounters with the Solar System, and our selection for that study was based on G╬▒i╬▒ DR2 data. Aims. When the Gaia EDR3 data release was published, it became necessary to update this database. Additionally, we performed Monte Carlo simulations to obtain uncertainties on the parameters of the closest approach to the Sun of each object. Methods. We recalculated the close approach parameters of all stars in the previous StePPeD release, which resulted in removing approximately one-third of the total. Then we searched for new candidates in the whole Gaia EDR3 catalogue. We also take into account the duplicity of the found stars and additionally searched for double stars passing near the Sun which had been overlooked in previous papers. We also found the necessary mass estimates for new objects and updated this information for previously selected stars. Results. After a careful checking of all the collected data, we composed a new list of 155 potential stellar perturbers of the long-period comet motion. We applied a new threshold of 2 pc for the minimum star-Sun distance. This list consists of 146 single stars and nine multiple systems. For each object, we also estimated the uncertainty of the parameters of their closest approach to the Sun. Among these stars, we found a new potential strong past perturber, HD 7977, and confirmed the plausibility of a similar action on the part of Gliese 710 in the future

    Catalytic Removal of NOx on Ceramic Foam-Supported ZnO and TiO2 Nanorods Ornamented with W and V Oxides

    Energy consumption steadily increases and energy production is associated with many environmental risks, e.g., generating the largest share of greenhouse gas emissions. The primary gas pollution concern is CO2, CH4, and nitrogen oxides (NOx). Environmental catalysis plays a pivotal role in NOx mitigation (DeNOx). This study investigated, for the first time, a collection of ceramic foams as potential catalyst support for selective catalytic NOx reduction (SCR). Ceramic foams could be an attractive support option for NOx removal. However, we should functionalize the surface of raw foams for such applications. A library of ceramic SiC, Al2O3, and ZrO2 foams ornamented with nanorod ZnO and TiO2 asWand V oxide support was obtained for the first time. We characterized the surface layer coating structure using the XPS, XRF and SEM, and TEM microscopy to optimize the W to V molar ratio and examine NO2 mitigation as the SCR model, which was tested only very rarely. Comparing TiO2 and ZnO systems reveals that the SCR conversion on ZnO appeared superior vs. the conversion on TiO2, while the SiC-supported catalysts were less efficient than Al2O3 and ZrO2-supported catalysts.
