    Integrated aquaculture contributes to the transfer of mcr-1 between animals and humans via the aquaculture supply chain

    Background Since its discovery in 2015, the mobile colistin resistance gene mcr-1 has been reported in bacteria from > 50 countries. Although aquaculture-associated bacteria may act as a significant reservoir for colistin resistance, systematic investigations of mcr-1 in the aquaculture supply chain are scarce. Objectives We investigated the presence of colistin resistance determinants in the aquaculture supply chain in south China and determined their characteristics and relationships. Methods A total of 250 samples were collected from a duck-fish integrated fishery, slaughter house, and market in Guangdong Province, China, in July 2017. Colistin-resistant bacteria were isolated on colistin-supplemented CHROMagar Orientation plates, and the species were identified by matrix-assisted laser desorption/ionization time-of-flight assay. The presence of mcr genes was confirmed by polymerase chain reaction analysis. We examined the minimum inhibitory concentrations (MICs) of 16 antimicrobial agents against the isolates using agar diffusion and broth microdilution methods. Whole-genome sequencing (WGS) was used to explore the molecular characteristics and relationships of mcr-1-positive Escherichia coli (MCRPEC). Results Overall, 143 (57.2%) colistin-resistant bacteria were isolated, of which, 56 (22.4%, including 54 Escherichia coli and two Klebsiella pneumoniae) and four Aeromonas species were positive for mcr-1 and mcr-3, respectively. The animal-derived MCRPEC were significantly more prevalent in integrated fishery samples (40.0%) than those in market (4.8%, P 90%) but were susceptible to carbapenems and tigecycline. WGS analysis suggested that mcr-1 was mainly contained on plasmids, including IncHI2 (29.6%), IncI2 (27.8%), IncX4 (14.8%), and IncP (11.1%). Genomic analysis suggested mcr-1 transmission via the aquatic food chain. Conclusions MCRPEC were highly prevalent in the aquaculture supply chain, with the isolates showing resistance to most antibiotics. The data suggested mcr-1 could be transferred to humans via the aquatic food chain. Taking the “One Health” perspective, aquaculture should be incorporated into systematic surveillance programs with animal, human, and environmental monitoring

    Associations between plasma metal mixture exposure and risk of hypertension: A cross-sectional study among adults in Shenzhen, China

    BackgroundMetal exposure affects human health. Current studies mainly focus on the individual health effect of metal exposure on hypertension (HTN), and the results remain controversial. Moreover, the studies assessing overall effect of metal mixtures on hypertension risk are limited.MethodsA cross-sectional study was conducted by recruiting 1,546 Chinese adults who attended routine medical check-ups at the Eighth Affiliated Hospital of Sun Yat-sen University, Shenzhen. The plasma levels of 13 metals were measured using inductively coupled plasma mass spectrometry. Multivariate logistic regression model, restricted cubic spline (RCS) model and the Bayesian Kernel Machine Regression (BKMR) model were applied to explore the single and combined effect of metals on the risk of HTN.ResultsA total of 642 (41.5%) participants were diagnosed with HTN. In the logistic regression model, the adjusted odds ratios (ORs) were 0.71 (0.52, 0.97) for cobalt, 1.40 (1.04, 1.89) for calcium, 0.66 (0.48, 0.90), and 0.60 (0.43, 0.83) for aluminum in the second and third quartile, respectively. The RCS analysis showed a V-shaped or an inverse V-shaped dose-response relationship between metals (aluminum or calcium, respectively) and the risk of HTN (P for non-linearity was 0.017 or 0.009, respectively). However, no combined effect was found between metal mixture and the risk of hypertension.ConclusionsPlasma levels of cobalt, aluminum and calcium were found to be associated with the risk of HTN. Further studies are needed to confirm our findings and their potential mechanisms with prospective studies and experimental study designs

    Role of Essential Amino Acids in Age-Induced Bone Loss

    Age-induced osteoporosis is a global problem. Essential amino acids (EAAs) work as an energy source and a molecular pathway modulator in bone, but their functions have not been systematically reviewed in aging bone. This study aimed to discuss the contribution of EAAs on aging bone from in vitro, in vivo, and human investigations. In aged people with osteoporosis, serum EAAs were detected changing up and down, without a well-established conclusion. The supply of EAAs in aged people either rescued or did not affect bone mineral density (BMD) and bone volume. In most signaling studies, EAAs were proven to increase bone mass. Lysine, threonine, methionine, tryptophan, and isoleucine can increase osteoblast proliferation, activation, and differentiation, and decrease osteoclast activity. Oxidized L-tryptophan promotes bone marrow stem cells (BMSCs) differentiating into osteoblasts. However, the oxidation product of tryptophan called kynurenine increases osteoclast activity, and enhances the differentiation of adipocytes from BMSCs. Taken together, in terms of bone minerals and volume, more views consider EAAs to have a positive effect on aging bone, but the function of EAAs in bone metabolism has not been fully demonstrated and more studies are needed in this area in the future

    SIIS-SLAM: A Vision SLAM Based on Sequential Image Instance Segmentation

    Simultaneous localization and mapping (SLAM) is a fundamental function of intelligent robots. To reduce the influence of dynamic objects on SLAM in dynamic environments, this study pro-poses a visual SLAM based on sequential image segmentation, referred to as SIIS-SLAM. Based on ORB-SLAM3, SIIS-SLAM integrates the sequential image instance segmentation and optical flow dynamic detection module. The sequential image segmentation module is designed to eliminate the effectiveness of dynamic objects in the estimation of relative pose between sequential frames. Specifically, based on the coarse relative pose estimated by ORB-SLAM3 and the box coordinates of instances detected by Mask R-CNN, the sequential image segmentation module effectively improves the speed and accuracy of instance segmentation. Dynamic objects can be effectively detected by combining the instance segmentation results and optical flow module. Filtering the feature points in dynamic objects can improve the accuracy and robustness of SLAM. Experimental results demonstrate that SIIS-SLAM achieves the better accuracy in dynamic environments compared to ORB SLAM3 and other advanced methods

    Improved Particle Filter Using Clustering Similarity of the State Trajectory with Application to Nonlinear Estimation: Theory, Modeling, and Applications

    A clustering similarity particle filter based on state trajectory consistency is presented for the mathematical modeling, performance estimation, and smart sensing of nonlinear systems. Starting from an information fusion model based on the consistency principle of the spatial state trajectory, the predicted observation information of the current particle filter (original trajectory) and future multistage Gaussian particle filter (modified trajectory) are selected as the state trajectories of the sampling particles. Clustering similarity methods are used to measure the state trajectories of the sampling particles and the actual system (reference trajectory). The importance weight of a first-order Markov model is updated with the measurement results. By integrating the targeted compensation scheme of the latest measurement information into the sequential importance sampling process, the adverse effects of the particle degradation phenomenon are effectively reduced. The convergence theorems of the improved particle filter are proposed and proved. The improved filter is applied to practical cases of nonlinear process estimation, economic statistical prediction, and battery health assessment, and the simulation results show that the improved particle filter is superior to traditional filters in estimation accuracy, efficiency, and robustness

    Dietary Factors of blaNDM Carriage in Health Community Population: A Cross-Sectional Study

    Aim: There is an ongoing debate as to what extent antimicrobial resistance (AMR) can be transmitted from dietary to humans via the consumption of food products. We investigated this association between dietary and global spreading carbapenem-resistant gene blaNDM Methods: We did a cross-sectional study to assess the risk factors for carrier of blaNDM in health community. Healthy adults were recruited from the residents attending Community Healthcare Service in Shenzhen City (Guangdong Province, China), through 1February 2018 to 31December 2019, and 718 pre-participants were included in this study. Questionnaire were obtained and the qualitative food frequency questionnaire (Q-FFQ) were used to assess dietary intake. qPCR was applied to confirm the carrier of blaNDM in participants’fecal samples. Multivariable logistic regression was used to estimate the odds ratio (OR) and 95% confidence interval (95% CI) of each outcome according to each dietary factor before and after prosperity score matching (PSM). Results: we showed that a high intake of coarse grain (OR 1.003; 95% CI 1.001–1.005, p < 0.01) and root and tuber crops (OR 1.003; 95% CI 1.001–1.004, p < 0.05) were independent risk factor for blaNDM carrier in health communities, suggesting a possible transfer of AMRbetweendietary andhumans. Surprisingly, we also showed an association between a higher intake of poultry as a protective, which may be explained by the beneficial effects on the gut microbiota. Conclusion: Dietary factors such as intake of coarse grain, root and tuber crops and poultry were associated with blaNDM carrier in health communities. The influence of dietary factorson blaNDM carrier in the present study provides insights for the tangible dietary advice with guidelines to the routine of people with the risk of blaNDM carrier. This demonstrates the role of dietary intake in the prevention of blaNDM carrier, since prevention is the best way to control modifiable risk factors. A lower carrier rate of blaNDM is helpful to reduce the possibility of transmission and pathogenicity. Further studies on food, microbiota and antimicrobial resistance are necessary to confirm this possible association and unravel underlying mechanisms

    Enhancing sustainable potato production—a case study in northern china

    Potato is the fourth most important staple crop in China. To meet the increased demand and environmental objectives, potato production should be enhanced by sustainable practices that aim to maximize yield and resource use efficiencies and minimize environmental impacts. Most experiments so far have taken place on experimental stations, while on-farm experimentation is needed to evaluate, demonstrate and scale more sustainable practices. On-farm field experiments were conducted in two years (2017, 2018) in northern China to investigate and demonstrate the effects of different nitrogen (N) fertiliser and irrigation input levels on potato yield, quality, resource use efficiency and environmental impacts. The 2017 experimental results on one farmer’s field showed that under irrigated conditions, adding N fertiliser (from 0 to 267 kg ha−1) did not increase yield due to the high soil N supply, rather it reduced tuber quality. The 2018 experimental results, in which four additional farmers were involved, indicated that, under irrigated conditions, reducing N fertiliser from the current rates (189–252 kg ha−1) to lower levels (109–181 kg ha−1) did not affect yield nor quality; while further reducing N fertiliser inputs (to 9–117 kg ha−1) resulted in a yield reduction (18% on average) in some fields. In both years, irrigation improved tuber yield and quality compared to that under rainfed conditions. The nitrogen use efficiency was improved and N surplus was reduced by applying irrigation and reducing N fertiliser input. Farmers expressed they were willing to reduce N fertiliser input by 10–20%, and indicated that a widespread adaptation of drip irrigation is hindered by the high costs and labor requirements. Site-specific recommendations on optimum N fertiliser and irrigation management must be provided, which should preferably be based on regular quantitative monitoring of soil N supply and soil moisture content

    Curcumin Inhibits Acute Vascular Inflammation through the Activation of Heme Oxygenase-1

    Curcumin has several therapeutic properties such as anti-inflammatory effect. Heme oxygenase-1 (HO-1) has been showed to have cytoprotective effects in some pathological conditions. However, the role of HO-1 in anti-inflammatory effect of curcumin is unknown. In this study, we investigate whether the anti-inflammatory effect of curcumin in vascular may be involved in the activation of HO-1. New Zealand white rabbits were fed regular control diet or control diet added with 0.3% curcumin (wt/wt) for four weeks. Acute vascular inflammation of rabbits was induced by putting a collar on the left common carotid artery for 24 hours. HO-1 inhibitor and siRNA were used to investigate the role of HO-1 in the anti-inflammatory effect of curcumin in collared vascular. We also explored the mechanism of curcumin-induced activation of HO-1 in vitro. The serum bilirubin and vascular, liver, and spleen HO-1 mRNA levels were significantly increased in curcumin-treated rabbits. The vascular inflammation was significantly decreased in the curcumin-treated animals compared with the control. Treatment of the rabbits with an inhibitor of HO or HO-1 siRNA to knock down the carotid artery HO-1 abolished the ability of curcumin to inhibit vascular inflammation. Treatment of cultured human artery endothelial cells with curcumin induced the HO-1 expression through the activation of nuclear factor-E2-related factor 2 (Nrf2) and an antioxidant responsive element via the p38 MAPK signalling pathway. In conclusion, curcumin inhibits vascular inflammation in vivo and in vitro through the activation of HO-1

    Short‐Term Exposure to Ambient Air Pollution and Hospital Admissions for Sequelae of Stroke in Chinese Older Adults

    Abstract Extensive evidence suggests that ambient air pollution contributes to a higher risk of hospital admissions for cerebrovascular diseases; however, its association with admissions for sequelae of stroke remains unclear. A time‐stratified case‐crossover study was conducted among 31,810 older adults who were admitted to hospital for sequelae of stroke in Guangzhou, China during 2016–2019. For each subject, daily residential exposure to fine particulate matter (PM2.5), inhalable particulate matter (PM10), sulfur dioxide (SO2), nitrogen dioxide (NO2), carbon monoxide (CO), and ozone (O3) was extracted from a validated grid data set. Conditional logistic regression models were used for exposure‐response analyses. In single‐pollutant models, each interquartile range (IQR) increase of lag 04‐day exposure to CO (IQR: 0.25 mg/m3) and lag 3‐day exposure to O3 (69.6 μg/m3) was significantly associated with a 4.53% (95% confidence interval: 1.67%, 7.47%) and 5.63% (1.92%, 9.48%) increase in odds of hospital admissions for sequelae of stroke, respectively. These associations did not significantly vary across age or sex. With further adjustment for each of the other pollutants in 2‐pollutant models, the association for CO did not change significantly, while the association for O3 disappeared. We estimated that 7.72% of the hospital admissions were attributable to CO exposures. No significant or consistent association was observed for exposure to PM2.5, PM10, SO2, or NO2. In conclusion, short‐term exposure to ambient CO, even at levels below the WHO air quality guideline, was significantly associated with an increased odds of hospital admissions for sequelae of stroke, which may lead to considerable excess hospital admissions
