    RGD-conjugated gold nanorods induce radiosensitization in melanoma cancer cells by downregulating αvβ3 expression

    Background: Melanoma is known to be radioresistant and traditional treatments have been intractable. Therefore, novel approaches are required to improve the therapeutic efficacy of melanoma treatment. In our study, gold nanorods conjugated with Arg-Gly-Asp peptides (RGD-GNRs) were used as a sensitizer to enhance the response of melanoma cells to 6 mV radiation. Methods and materials: A375 melanoma cells were treated by gold nanorods or RGD-GNRs with or without irradiation. The antiproliferative impact of the treatments was measured by MTT assay. Radiosensitizing effects were determined by colony formation assay. Apoptosis and cell cycle data were measured by flow cytometry. Integrin alpha(v)beta(3) expression was also investigated by flow cytometry. Results: Addition of RGD-GNRs enhanced the radiosensitivity of A375 cells with a dose-modifying factor of 1.35, and enhanced radiation-induced apoptosis. DNA flow cytometric analysis indicated that RGD-GNRs plus irradiation induced significant G2/M phase arrest in A375 cells. Both spontaneous and radiation-induced expressions of integrin alpha(v)beta(3) were downregulated by RGD-GNRs. Conclusion: Our study indicated that RGD-GNRs could sensitize melanoma A375 cells to irradiation. It was hypothesized that this was mainly through downregulation of radiation-induced alpha(v)beta(3), in addition to induction of a higher proportion of cells within the G2/M phase.     Characterizing large-scale weak interlayer shear zones using conditional random field theory

    The shear behavior of large-scale weak intercalation shear zones (WISZs) often governs the stability of foundations, rock slopes, and underground structures. However, due to their wide distribution, undulating morphology, complex fabrics, and varying degrees of contact states, characterizing the shear behavior of natural and complex large-scale WISZs precisely is challenging. This study proposes an analytical method to address this issue, based on geological fieldwork and relevant experimental results. The analytical method utilizes the random field theory and Kriging interpolation technique to simplify the spatial uncertainties of the structural and fabric features for WISZs into the spatial correlation and variability of their mechanical parameters. The Kriging conditional random field of the friction angle of WISZs is embedded in the discrete element software 3DEC, enabling activation analysis of WISZ C2 in the underground caverns of the Baihetan hydropower station. The results indicate that the activation scope of WISZ C2 induced by the excavation of underground caverns is approximately 0.5–1 times the main powerhouse span, showing local activation. Furthermore, the overall safety factor of WISZ C2 follows a normal distribution with an average value of 3.697

    Molecular Identification of Multidrug-Resistant Campylobacter Species From Diarrheal Patients and Poultry Meat in Shanghai, China

    Emerging resistance to the antimicrobial agents of choice for treatment of thermophilic Campylobacter infections is becoming a serious threat to public health. In this study, 548 Campylobacter (372 C. jejuni and 176 C. coli) isolates from diarrheal patients and poultry meat were subjected for antibiotic susceptibility analysis to ciprofloxacin, tetracycline, gentamicin, erythromycin and clindamycin. Among them, 151 Campylobacter (32 C. jejuni and 119 C. coli) were identified as multidrug resistant isolates. PFGE analysis was performed on the 151 multidrug resistant isolates to determine their genetic relatedness, and 103 PFGE genotypes were determined. Some isolates from both human and chicken belonged to identical genotypes, indicating these clones might be able to spread between human and chicken. Antibiotic resistant genes of the 151 isolates were identified. The numbers of isolates carried tet (O), aadE, ermB, and aadE-sat4-aphA were 148 (98%), 89 (58.9%), 31 (20.5%), and 10 (6.6%), respectively. Almost all (n = 150, 99.3%) had gyrA mutation at codon 86. And the 23s rRNA A2075G point mutation was found in 56 (37.1%) isolates. Gene mutations at the cmeR-cmeABC intergenic region may lead to the activation of CmeABC multidrug efflux pump, and in this study novel sequence types of the intergenic region were identified in both C. jejuni and C. coli.     Diagnosis of mixed infection and a primary immunodeficiency disease using next-generation sequencing: a case report

    Major Histocompatibility Complex Class II (MHC II) deficiency is a rare primary immunodeficiency disorder (PID) with autosomal recessive inheritance pattern. The outcome is almost fatal owing to delayed diagnosis and lacking of effective therapy. Therefore, prompt diagnosis, timely and effective treatment are critical. Here, we report a 117-day-old boy with diarrhea, cough, cyanosis and tachypnea who was failed to be cured by empiric antimicrobial therapy initially and progressed to severe pneumonia and respiratory failure. The patient was admitted to the pediatric intensive care unit (PICU) immediately and underwent a series of tests. Blood examination revealed elevated levels of inflammatory markers and cytomegalovirus DNA. Imaging findings showed signs of severe infection of lungs. Finally, the diagnosis was obtained mainly through next-generation sequencing (NGS). We found out what pathogenic microorganism he was infected via repeated conventional detection methods and metagenomic next-generation sequencing (mNGS) of sputum and bronchoalveolar lavage fluid (BALF). And his whole exome sequencing (WES) examination suggested that CIITA gene was heterozygous mutation, a kind of MHC II deficiency diseases. After aggressive respiratory support and repeated adjustment of antimicrobial regimens, the patient was weaned from ventilator on the 56th day of admission and transferred to the immunology ward on the 60th day. The patient was successful discharged after hospitalizing for 91 days, taking antimicrobials orally to prevent infections post-discharge and waiting for stem cell transplantation. This case highlights the potential importance of NGS in providing better diagnostic testing for unexplained infection and illness. Furthermore, pathogens would be identified more accurately if conventional detection techniques were combined with mNGS

    Dual channel dual focus optical coherence tomography for imaging accommodation of the eye

    A dual channel dual focus spectral-domain optical coherence tomography was developed for imaging the accommodation of the eye in real time. The system can provide simultaneous cross-sectional imaging of all the surfaces of the anterior segment of the eye including the cornea, anterior chamber, anterior and posterior surfaces of the crystalline lens. Thus, the modification of the curvatures of the anterior and posterior surfaces of the crystalline lens and the dimensions of the anterior segment of the eye with accommodation can be calculated. The system was successfully tested in imaging accommodation. The preliminary results demonstrated the feasibility of this novel approach

    Analysis of mechanical behavior of soft rocks and stability control in deep tunnels

    Due to the weakness in mechanical properties of chlorite schist and the high in situ stress in Jinping II hydropower station, the rock mass surrounding the diversion tunnels located in chlorite schist was observed with extremely large deformations. This may significantly increase the risk of tunnel instability during excavation. In order to assess the stability of the diversion tunnels laboratory tests were carried out in association with the petrophysical properties, mechanical behaviors and water-weakening properties of chlorite schist. The continuous deformation of surrounding rock mass, the destruction of the support structure and a large-scale collapse induced by the weak chlorite schist and high in situ stress were analyzed. The distributions of compressive deformation in the excavation zone with large deformations were also studied. In this regard, two reinforcement schemes for the excavation of diversion tunnel bottom section were proposed accordingly. This study could offer theoretical basis for deep tunnel construction in similar geological conditions

    Mechanical Anisotropy and Failure Characteristics of Columnar Jointed Rock Masses (CJRM) in Baihetan Hydropower Station: Structural Considerations Based on Digital Image Processing Technology

    The columnar joints in Baihetan hydropower station are primary tensile joints since they were formed during the process of lava condensation. Understanding the influence of columnar jointed rock mass (CJRM) on the mechanical response and failure modes is the basis for designing of associated engineering works. Hence, the structural characteristics of Baihetan CJRM were analyzed by carrying out a geological survey at first. Three groups of numerical models capable of reflecting the structural characteristics of CJRM were then established to analyze the mechanical and failure characteristics. The results in this study showed that: (1) Irregularity of columnar basalt restricted crack propagation on columnar joints and also led to stress concentration in the distorted parts, and thus, damage of basalt columns; (2) when the included angle between direction of concentrated defect structures in CJRM and uniaxial stress was large, the defect structures can prevent crack propagation on columnar joints, and the failure of defect structure can cause the overall failure of the rock mass; and (3) under the condition of same columnar structure and included angle, the peak strength of models with microcracks and structural plane was low and the irregular shape of columnar joints decreased the anisotropy of mechanical parameters
