    Effects of food restriction on growth, body composition and gene expression related in regulation of lipid metabolism and food intake in grass carp

    It is well known that most fish would prefer to use body lipid stores for energy expenditure when receiving a long-term food restriction. However, the mechanism of this is still not clear. In the present study, a growth experiment was carried out to investigate the effects of food restriction on growth performance, gene expression related in regulation of lipid metabolism and food ingestion in grass carp (Ctenopharyngodon idellus). Four rations, satiation (S), 80% S, 60% S and 40% S, were adopted in this study. Each treatment was randomly assigned to triplicate net cages of 15 fish (177.3 +/- 3.3 g) per cage. The experiment lasted for 49 days at 30.0 +/- 3.0 degrees C. The experimental results showed that a significant increase in feeding rate and weight gain was found in grass carp with the increased ration level. The body lipid and energy content of the grass carp exhibited a significant decrease when receiving food restriction. The transcriptional levels of the genes involved in lipogenesis (srebp-1c, fas, ppar gamma) were down-regulated at the rations of food restriction. The relative expression of hepatic fas (fatty acid synthetase) and srebp-1c (sterol regulatory element-binding protein 1c) in the fish at satiation were significantly higher than the restricted-fed groups. Similarly, the expressions of hepatic ppar. (peroxisome proliferator-activated receptor-gamma) in the fish at the ration of satiation and 80% S were significantly higher than the group at the low ration of 40% S. However, the expression of hepatic cpt-1a (carnitine palmitoyl transferase I) involved in fatty acid beta-oxidation in fish was significantly up-regulated when receiving food restriction. Other hepatic lipolysis genes of ppar alpha (peroxisome proliferators-activated receptor alpha) and hl (hepatic lipase) didn&#39;t show any significant changes in restricted-fed fish. The transcriptional levels of hepatic leptin and hypothalamus pomc (proopiomelanocortin) were significantly down-regulated in fish fed with restricted rations. But the hypothalamus npy (neuropeptide Y) and lepr (leptin receptor) had no change. The present results indicated that a long-term food restriction could cause less accumulation of lipid and could be through a way of down-regulating lipogenesis genes and up-regulating lipolysis genes. Long-term restriction could also activate the appetite of grass carp by down-regulating some anorexigenic genes.     Glycyrrhizin Attenuates Salmonella enterica Serovar Typhimurium Infection: New Insights Into Its Protective Mechanism

    Glycyrrhizin (GL), a triterpenoid glycoside, serves important functions in various biological activities, including antiviral and antitumor immune responses. However, the anti-inflammatory effects of GL on Salmonella enterica serovar Typhimurium (ST)-induced injury in mice and the mechanisms underlying the protection of GL are poorly understood. Here, we investigated the effects of GL on host immune responses against ST infection in mice. A phenotypic analysis using hematoxylin and eosin (H&amp;E) staining and transmission electron microscopy showed that GL relieved ST-induced weight loss and intestinal mucosal injury. A colonization assay showed that GL significantly reduced ST colonization in the ileum and colon and translocation to the liver and spleen. An antibacterial activity assay and real-time PCR revealed that GL had no direct inhibitory impact on ST growth or virulence gene expression. ELISA showed that GL pretreatment significantly decreased proinflammatory cytokine (IFN-γ, TNF-α, IL-6) secretion and increased anti-inflammatory cytokine (IL-10) secretion in the ileum, colon and serum of ST-infected mice. Moreover, flora analysis showed that GL reduced Akkermansia, Sutterella, Prevotella and Coprococcus but enriched Parabacteroides and Anaerotruncus in the cecum of ST-infected mice. These results suggest that GL promotes the secretion of immune factors and modulates intestinal flora to prevent further ST infection. We also analyzed the effect of GL on immunocytes and found that GL promoted the phenotypic and functional maturation of murine bone marrow-derived dendritic cells (BMDCs). Flow cytometry and western blotting demonstrated that NF-κB, ERK, and p38 MAPK were required for GL-induced BMDC maturation. The above findings indicate that GL attenuates ST infection by modulating immune function and intestinal flora. This study enriches our current knowledge of GL-mediated immunological function and provides a new perspective on the prevention of Salmonella infection in animals and humans

    NF-κB Induced the Donor Liver Cold Preservation Related Acute Lung Injury in Rat Liver Transplantation Model

    We have observed at our clinical work that acute lung injury (ALI) often occurs in patients transplanted with donor livers persevered for long time. So, we conducted this study to investigate the influence of cold preservation time (CPT) of donor liver on ALI induced by liver transplantation (LT), and further study the role of nuclear factor-κB (NF-κB) in the process.Wistar rats were used as donors and recipients to establish orthotopic rat liver transplantation models. Donor livers were preserved at 4°C for different lengths of time. The effect of NF-κB inhibitor, ammonium pyrrolidinedithiocarbamate (PDTC), on ALI was detected. All samples were harvested after 3 h reperfusion. The severity of liver injury was evaluated first. The expressions of tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) in liver tissue and liver outflow serum were measured respectively. The severity indexes of ALI, the activity of NF-κB and inhibitor-κBα (I-κBα) in lung/liver were measured accordingly.With the prolonged liver CPT, the liver damage associated indexes and ALI-related indexes all increased significantly. TNF-α and IL-1β in liver outflow serum increased accordingly, and the activity of NF-κB in liver/lung increased correspondingly. All these ALI-associated indexes could be partially reversed by the use of PDTC.Extended CPT aggravates the damage of donor liver and induces the expressions of TNF-α and IL-1β in liver. These inflammatory factors migrate to lung via liver outflow blood and activate NF-κB in lung, inducing ALI finally. NF-κB may play a critical role in LT-related ALI. Patients with or at risk of ALI may benefit from acute anti-inflammatory treatment with PDTC

    Wafer-Level Packaging Method for RF MEMS Applications Using Pre-Patterned BCB Polymer

    A radio-frequency micro-electro-mechanical system (RF MEMS) wafer-level packaging (WLP) method using pre-patterned benzo-cyclo-butene (BCB) polymers with a high-resistivity silicon cap is proposed to achieve high bonding quality and excellent RF performance. In this process, the BCB polymer was pre-defined to form the sealing ring and bonding layer by the spin-coating and patterning of photosensitive BCB before the cavity formation. During anisotropic wet etching of the silicon wafer to generate the housing cavity, the BCB sealing ring was protected by a sputtered Cr/Au (chromium/gold) layer. The average measured thickness of the BCB layer was 5.9 μm. In contrast to the conventional methods of spin-coating BCB after fabricating cavities, the pre-patterned BCB method presented BCB bonding layers with better quality on severe topography surfaces in terms of increased uniformity of thickness and better surface flatness. The observation of the bonded layer showed that no void or gap formed on the protruding coplanar waveguide (CPW) lines. A shear strength test was experimentally implemented as a function of the BCB widths in the range of 100–400 μm. The average shear strength of the packaged device was higher than 21.58 MPa. A RF MEMS switch was successfully packaged using this process with a negligible impact on the microwave characteristics and a significant improvement in the lifetime from below 10 million to over 1 billion. The measured insertion loss of the packaged RF MEMS switch was 0.779 dB and the insertion loss deterioration caused by the package structure was less than 0.2 dB at 30 GHz

    Comparison of the preventive effect of urethral cleaning versus disinfection for catheter-associated urinary tract infections in adults: A network meta-analysis

    Background: Catheter-associated urinary tract infections (CAUTIs) are the main cause of infectious complications in patients with indwelling urinary catheters (IDCs). However, the best cleaning methods for the prevention of CAUTIs have not been evaluated clearly in previous studies. Methods: An electronic database search was performed, from inception to December 2017. Randomized controlled trials and quasi-experimental trials using different methods of urethral cleaning versus disinfection to prevent CAUTIs were considered. The study selection and data collection were performed independently by two reviewers. The risk of bias assessment was performed using the Cochrane risk of bias scale. The primary outcome was the incidence rates of CAUTIs. A network meta-analysis was conducted to compare the effect among the different methods of urethral cleaning versus disinfection to prevent CAUTIs. Results: Thirty-three studies (6490 patients) with seven different methods of urethral cleaning versus disinfection were eligible for inclusion, and the data were summarized in the network meta-analysis. No evidence of heterogeneity (P > 0.05) was observed among the studies. The network meta-analysis showed that there was no difference in the incidence of CAUTIs when comparing the different urethral cleaning methods versus disinfection (P > 0.05 for all). However, chlorhexidine ranked first in the results of the Bayesian analysis and is recommended for preventing CAUTIs. Conclusions: Current evidence suggests that there are no significant differences among different urethral cleaning versus disinfection methods with regard to CAUTI incidence rates.     Value Coefficient of Polyethylene Fiber Soil Embankment Slope Based on Response Surface Analysis

    The utilization of polymers can strengthen soil, but at a high price. In this study, value coefficients were proposed to evaluate the cost-effectiveness of fiber-reinforced roadbeds, and the effects of embankment-slope-influencing factors on the value coefficients were analyzed by response surface methodology. Ultrahigh-molecular-weight polyethylene fiber (UPEF) was used as the reinforcement material for soil. First, the shear strength parameters of fiber soil with different fiber diameters were obtained from the direct shear tests to set the parameters of the finite element models. Second, three factors, namely filling height, slope angle, and fiber diameter, were selected as input parameters based on the Box&ndash;Behnken Design (BBD) experimental design method, and their effects on the value coefficient of the fiber soil embankment slope were investigated. Finally, the design parameters at the maximum value coefficient of the fiber soil embankment slope were determined based on the results of the response surface analysis. The results indicated that the addition of UPEF could effectively improve the cohesion of the soil; the interaction between the filling height and fiber diameter is most obvious.
