Glucocorticoid Receptor and Adipocyte Biology.
Glucocorticoids are steroid hormones that play a key role in metabolic adaptations during stress, such as fasting and starvation, in order to maintain plasma glucose levels. Excess and chronic glucocorticoid exposure, however, causes metabolic syndrome including insulin resistance, dyslipidemia, and hyperglycemia. Studies in animal models of metabolic disorders frequently demonstrate that suppressing glucocorticoid signaling improves insulin sensitivity and metabolic profiles. Glucocorticoids convey their signals through an intracellular glucocorticoid receptor (GR), which is a transcriptional regulator. The adipocyte is one cell type that contributes to whole body metabolic homeostasis under the influence of GR. Glucocorticoids' functions on adipose tissues are complex. Depending on various physiological or pathophysiological states as well as distinct fat depots, glucocorticoids can either increase or decrease lipid storage in adipose tissues. In rodents, glucocorticoids have been shown to reduce the thermogenic activity of brown adipocytes. However, in human acute glucocorticoid exposure, glucocorticoids act to promote thermogenesis. In this article, we will review the recent studies on the mechanisms underlying the complex metabolic functions of GR in adipocytes. These include studies of the metabolic outcomes of adipocyte specific GR knockout mice and identification of novel GR primary target genes that mediate glucocorticoid action in adipocytes
A Basic Course for Sunday School Teacher Training
Teaching has never been easy. There is no formula for instant teaching success. It takes time, energy, patience, work and prayer. But, teaching can be a exciting and satisfying experience, if the teacher knows what to teach and how to do it effectively.
There are many specific steps and techniques which teachers can employ that will lead to more effective teaching. This project is a attempt to identify and systematize some of the basic elements of teaching and learning in the church. We can not cover everything, but we can uncover some of the essential inqurdients to effective teaching.
Teacher training has long been beyond the reach of Taiwan\u27s church, for lacking of adequate teaching personnel and material. Now here is a tool which designed for use on the local level. This material can be used with good success by Pastor, Sunday school Superintendent, and others willing to spend a reasonable amount of time and effert in preparation. One of its greatest uses will be to challenge prospective teachers, enlist them in the work of the Sunday school and train them for that most important ministry
How Do Immigrant Older Adults in the United States Fare in Later Life? Examining Differences in Immigrant Status and Life-course Timing of Migration on Late-life Disablement and Mortality
This dissertation focuses on immigrant older adults in the United States and their experiences with late-life disablement. Specifically, this dissertation examines immigrant status differences in late-life disability among immigrants compared to non-immigrants and investigates how life-course timing of immigration shapes late-life disability among immigrant older adults using data from the National Health and Aging Trends Study (NHATS) 2011-2016. Drawing from life course and segmented assimilation theoretical frameworks, this research is guided by two broad research aims: 1) To determine if the immigrant health and mortality advantage extends to late-life disablement, 2) To examine how life-course timing of migration affects late-life disablement for immigrants. Results from Markov transition models show that compared to non-immigrant older adults, immigrants have greater risk of decline and lower risk of recovery from late-life disablement, yet have lower mortality risk. Among immigrant older adults, life-course timing of migration is an important predictor of late-life disability and mortality risk, as immigrants who migrated later-in-life have increased disability risk coupled with lower mortality risk relative to their counterparts who immigrated earlier in life. Moreover, Sullivan based life table calculations indicate longer life expectancies for immigrant older adults compared to their non-immigrant counterparts, but with a smaller proportion of disability-free years. This research highlights key areas where future research and policy makers can work towards reducing health disparities in late-life disablement and mortality for immigrant older adults in the United States
Sunday Friends: The Working Alternative to Charity
Sunday Friends is a non-profit organization in San José, California, that provides multiple activities for families who are in need of financial support. Given the particular location of the program, most families are Latino and bilingual. Participants and program volunteers form a community at an elementary school on a couple of scheduled Sundays each month. When family members participate in activities designed to educate, improve skills, and to give back to the larger community, they earn tickets that they can redeem for items that they need and want from the Sunday Friends store. Activities include healthy cooking projects, “Thank You Letter” writing, English-as-a-Second-Language programs, crafts, and education in nutrition and financial literacy. The program’s central focus is to empower families to break out of poverty. A specific guiding principal is the developmental assets approach promoted by the Search Institute in Minneapolis (http://www.search-institute.org/). This approach encourages individuals and organizations to work together toward a common goal of supporting the healthy development of all children and youth. Healthy development is conceptualized as consisting of the development of external assets (i.e., support, empowerment, boundaries and expectations, constructive activities) and internal assets (i.e., commitment to learning, positive values, social competencies, and positive identity). The 2009 evaluation’s primary focus was on whether Sunday Friends was succeeding at fostering developmental assets for children. Other relevant indicators of success were perceptions of program effectiveness, regard for program activities, and nutrition and healthy eating habits. Data were gathered from family members (adults and minors) during program activities using written surveys administered by volunteers to the research team (most of whom were bilingual). The families were recruited in person by program staff onsite. The questionnaires were written in English and Spanish (with the choice made by respondents). Sunday Friends volunteers were recruited to the study via email by Sunday Friends staff. They completed questionnaires through an online survey platform. In all, 74 parents or guardians, 67 children or youth, and 45 volunteers participated in the data collection. Across the three sets of surveys (parents or guardians, children or youth, and volunteers), there are fairly consistent results. The families and volunteers in the Sunday Friends program believe that Sunday Friends is effectively engaging them and meeting its goals. Overall, children and youth report positive experiences at Sunday Friends. On every dimension of psychological well-being, connectedness with others, and self-efficacy, the answers provided by children and youth indicate that Sunday Friends is promoting developmental assets. In addition, parents and guardians report that Sunday Friends has positive influences on their lives. The majority of measures, whether questions about self-efficacy, the effect of Sunday Friends on their children, or improved nutrition for their family, indicate that Sunday Friends is meeting its goals. Finally, volunteers agree that the program makes an important contribution to the lives of both children and adults. They also feel that Sunday Friends enhances their awareness of the community and contributes to making their personal lives more meaningful. The survey results presented here should be interpreted with caution because all persons surveyed were continuing participants in the program, and the cross-sectional data could not detect changes over time. Despite this caution, each group’s data and triangulation across family and volunteer surveys reveal that milestones are accomplished at Sunday Friends. It is safe to conclude that Sunday Friends’ community-based approach to empowering parents and youths is achieving its goals
Enhanced anticancer activity of a combination of docetaxel and Aneustat (OMN54) in a patient-derived, advanced prostate cancer tissue xenograft model.
The current first-line treatment for advanced metastatic prostate cancer, i.e. docetaxel-based therapy, is only marginally effective. The aim of the present study was to determine whether such therapy can be improved by combining docetaxel with Aneustat (OMN54), a multivalent botanical drug candidate shown to have anti-prostate cancer activity in preliminary in vitro experiments, which is currently undergoing a Phase-I Clinical Trial. Human metastatic, androgen-independent C4-2 prostate cancer cells and NOD-SCID mice bearing PTEN-deficient, metastatic and PSA-secreting, patient-derived subrenal capsule LTL-313H prostate cancer tissue xenografts were treated with docetaxel and Aneustat, alone and in combination. In vitro, Aneustat markedly inhibited C4-2 cell replication in a dose-dependent manner. When Aneustat was combined with docetaxel, the growth inhibitions of the drugs were essentially additive. In vivo, however, the combination of docetaxel and Aneustat enhanced anti-tumor activity synergistically and very markedly, without inducing major host toxicity. Complete growth inhibition and shrinkage of the xenografts could be obtained with the combined drugs as distinct from the drugs on their own. Analysis of the gene expression of the xenografts using microarray indicated that docetaxel + Aneustat led to expanded anticancer activity, in particular to targeting of cancer hallmarks that were not affected by the single drugs. Our findings, obtained with a highly clinically relevant prostate cancer model, suggest, for the first time, that docetaxel-based therapy of advanced human prostate cancer may be improved by combining docetaxel with Aneustat
Constraints on Muon Decay Parameters from Neutrino Mass
We derive model-independent constraints on chirality-changing terms in the
muon decay Lagrangian using limits on neutrino mass. We consider all
dimension-six operators invariant under the gauge symmetry of the Standard
Model which contribute to either a Dirac neutrino mass or muon decay. Taking an
upper limit on neutrino mass of 1 eV, we derive limits on the contributions of
chirality-changing operators to the Michel parameters four orders of magnitude
tighter than the current experimental constraints. We also identify two
operators which, due to their flavor structure, are not constrained by neutrino
mass. If near-future experiments find contributions to muon decay from these
operators, it could indicate interesting flavor structure in physics beyond the
