    The existence of an insulin-stimulated glucose and non-essential but not essential amino acid substrate interaction in diabetic pigs

    <p>Abstract</p> <p>Background</p> <p>The generation of energy from glucose is impaired in diabetes and can be compensated by other substrates like fatty acids (Randle cycle). Little information is available on amino acids (AA) as alternative energy-source in diabetes. To study the interaction between insulin-stimulated glucose and AA utilization in normal and diabetic subjects, intraportal hyperinsulinaemic euglycaemic euaminoacidaemic clamp studies were performed in normal (n = 8) and streptozotocin (120 mg/kg) induced diabetic (n = 7) pigs of ~40-45 kg.</p> <p>Results</p> <p>Diabetic vs normal pigs showed basal hyperglycaemia (19.0 ± 2.0 vs 4.7 ± 0.1 mmol/L, <it>P </it>< .001) and at the level of individual AA, basal concentrations of valine and histidine were increased (<it>P </it>< .05) whereas tyrosine, alanine, asparagine, glutamine, glutamate, glycine and serine were decreased (<it>P </it>< .05). During the clamp, diabetic vs normal pigs showed reduced insulin-stimulated glucose clearance (4.4 ± 1.6 vs 16.0 ± 3.0 mL/kg·min, <it>P </it>< .001) but increased AA clearance (166 ± 22 vs 110 ± 13 mL/kg· min, <it>P </it>< .05) at matched arterial euglycaemia (5-7 mmol/L) and euaminoacidaemia (2.8-3.5 mmol/L). The increase in AA clearance was mainly caused by an increase in non-essential AA clearance (93.6 ± 13.8 vs 46.6 ± 5.4 mL/kg·min, <it>P </it>< .01), in particular alanine (14.2 ± 2.4 vs 3.2 ± 0.4 mL/kg·min, <it>P </it>< .001)<b/>. Essential AA clearance was largely unchanged (72.9 ± 8.5 vs 63.3 ± 8.5 mL/kg· min), however clearances of threonine (<it>P </it>< .05) and tyrosine (<it>P </it>< .01) were increased in diabetic vs normal pigs (8.1 ± 1.3 vs 5.2 ± 0.5, and 14.3 ± 2.5 vs 6.4 ± 0.7 mL/kg· min, respectively).</p> <p>Conclusions</p> <p>The ratio of insulin-stimulated glucose versus AA clearance was decreased 5.4-fold in diabetic pigs, which was caused by a 3.6-fold decrease in glucose clearance and a 2.0-fold increase in non-essential AA clearance. In parallel with the Randle concept (glucose - fatty acid cycle), the present data suggest the existence of a glucose and non-essential AA substrate interaction in diabetic pigs whereby reduced insulin-stimulated glucose clearance seems to be partly compensated by an increase in non-essential AA clearance whereas essential AA are preferentially spared from an increase in clearance.</p

    A unified picture of nucleon antinucleon annihilation into pions emerges from a classical description of the pion wave produced in annihilation and the subsequent quantization of that wave as a coherent state.     Antibody persistence and booster responses to split-virion H5N1 avian influenza vaccine in young and elderly adults

    Avian influenza continues to circulate and remains a global health threat not least because of the associated high mortality. In this study antibody persistence, booster vaccine response and cross-clade immune response between two influenza A(H5N1) vaccines were compared. Participants aged over 18-years who had previously been immunized with a clade 1, A/Vietnam vaccine were re-immunized at 6-months with 7.5 mu g of the homologous strain or at 22-months with a clade 2, alum-adjuvanted, A/Indonesia vaccine. Blood sampled at 6, 15 and 22-months after the primary course was used to assess antibody persistence. Antibody concentrations 6-months after primary immunisation with either A/Vietnam vaccine 30 mu g alum-adjuvanted vaccine or 7.5 mu g dose vaccine were lower than 21days after the primary course and waned further with time. Re-immunization with the clade 2, 30 mu g alum-adjuvanted vaccine confirmed cross-clade reactogenicity. Antibody crossreactivity between A(H5N1) clades suggests that in principle a prime-boost vaccination strategy may provide both early protection at the start of a pandemic and improved antibody responses to specific vaccination once available

    Study of the pitch change of carbon coils during their growth

    AbstractIn the present paper, carbon coils (CCs) were prepared by CVD. Their morphology, particularly pitch changes of the carbon coils prepared in different conditions were observed. It was found that the carbon source flow plays an important role in carbon coil growth and its morphology evolution. The appropriate atmosphere and flow rate is beneficial to the steady reactivity of catalyst particles. As such each carbon coil can grow well and have an exact growth rate. When the carbon supply is sufficient, the CCs exhibit close spiral and small coil diameter. When carbon supply decreases, small carbon supply leads to large pitch and coil diameter. CCs can be synthesized with different coil pitch under different carbon supply. This may be of great significance for the controllable preparation of carbon coil and its application

    This report involves a 74-year-old-male who developed a thoracic epidural hematoma with paraparesis on the second postoperative day in conjunction with thoracic epidural anesthesia established before surgery for acute abdominal aortic dissection. The finding indicates that laminectomy can be performed successfully as late as three days after diagnosis of the hematoma, with a complete restitution of neurological function. High-dose steroid treatment may have been a contributing factor for the positive outcome

    Consensus on Aquatic Primary Productivity Field Protocols for Satellite Validation and Model Synthesis

    The NASA PACE project, in conjunction with the IOCCG, EUMETSAT, and JAXA, have initiated an Aquatic Primary Productivity working group, with the aim to develop community consensus on multiple methods for measuring aquatic primary productivity used for satellite validation and model synthesis. A workshop to commence the working group efforts was held December 05-07, 2018 at the University Space Research Association headquarters in Columbia, MD U.S.A., bringing together 26 active researchers from 16 institutions. The group discussed the primary differences, nuances, scales, uncertainties, definitions, and best practices for measurements of primary productivity derived from in situ/on-deck/laboratory radio/stable isotope incubations, dissolved oxygen concentrations (from incubations or autonomous platforms such as floats or gliders), oxygen-argon ratios, triple oxygen isotope, natural fluorescence, and FRRF/ETR/kinetic analysis. These discussions highlighted the necessity to move the community forward towards the establishment of climate-quality primary productivity measurements that follow uniform protocols, which is imperative to ensure that existing and future measurements can be compared, assimilated, and their uncertainties determined for model development and validation. The specific deliverable resulting from of this activity will be a protocol document, published in coordination with the IOCCG. This presentation will discuss the findings of the meeting, and address future activities of the working group

    Non-cytotoxic 1,2,3-triazole tethered fused heterocyclic ring derivatives display Tax protein inhibition and impair HTLV-1 infected cells

    Human T cell lymphotropic virus type 1 (HTLV-1) is a human retrovirus that infects approximately 10–20 million people worldwide and causes an aggressive neoplasia (adult T-cell leukemia/lymphoma - ATL). Therapeutic approaches for the treatment of ATL have variable effectiveness and poor prognosis, thus requiring strategies to identify novel compounds with activity on infected cells. In this sense, we initially screened a small series of 25 1,2,3-triazole derivatives to discover cell proliferation inhibitors and apoptosis inducers in HTLV-1-infected T-cell line (MT-2) for further assessment of their effect on viral tax activity through inducible-tax reporter cell line (Jurkat LTR-GFP). Eight promising compounds (02, 05, 06, 13, 15, 21, 22 and 25) with activity ≥70% were initially selected, based on a suitable cell-based assay using resazurin reduction method, and evaluated towards cell cycle, apoptosis and Tax/GFP expression analyses through flow cytometry. Compound 02 induced S phase cell cycle arrest and compounds 05, 06, 22 and 25 promoted apoptosis. Remarkably, compounds 22 and 25 also reduced GFP expression in an inducible-tax reporter cell, which suggests an effect on Tax viral protein. More importantly, compounds 02, 22 and 25 were not cytotoxic in human hepatoma cell line (Huh-7). Therefore, the discovery of 3 active and non-cytotoxic compounds against HTLV-1-infected cells can potentially contribute, as an initial promising strategy, to the development process of new drugs against ATL
