Study of endometrium by trans-vaginal sonography and itβs correlation with histopathology in perimenopausal women with abnormal uterine bleeding
Background: Objectives of the study were to study the endometrial patterns in cases of abnormal uterine bleeding (AUB) and anatomical (structural) lesions of uterus using transvaginal sonography (TVS) and endometrial histopathology and to determine the efficacy of TVS.Methods: Cross-section study of 50 perimenopausal age group with AUB in TNMC BYL Nair hospital from Nov 2017 to Nov 2018.Results: AUB was seen 40 to 45 years multiparous women TVS endometrial thickness (ET) T 6-10 mm (46%), ET 11-15 (22%), ET>15 mm (14%), <5 mm in (18%), showed 21 (42%) patients with endometrial hyperplasia on TVS, 12 (24%) simple hyperplasia 4 (8%) complex hyperplasia on histopathology fibroid 8%, adenomyosis 2%. endometrial polyp 6%.Conclusions: Endometrial lining exceeds 10 mm dilation and curettage to be done r/o endometrial hyperplasia, to study the endometrial patterns in cases of abnormal uterine bleeding and anatomical (structural) lesions of uterus using transvaginal sonography and endometrial histopathology
Structure and function relationship among the peptidyl prolyl cis/trans isomerases
Proteins exist in two conformers. The trans conformation is favored by the most of the amino acids. The proline residue due to its unique geometry has a high probability of being in the cis conformation. Thus the cis/trans isomerisation of the peptide bond preceding the proline residue becomes a rate limiting step in the folding and unfolding of the proteins. The enzymes which catalyze this rate limiting step were discovered by Fischer in porcine kidney and called as peptidyl prolyl cis trans isomerases (PPIases). There are four families of the PPIases. They are the parvulins, cyclophilins, FKBPs and trigger factors. All the four families catalyze a common reaction and the give rise to a stable trans product. We therefore wanted to analyse if cross complementation exists across the PPIase families. Our analysis has shown that the prokaryotic and the PPIase domain of the eukaryotic parvulins show a high structural similarity. The catalytic residues were found to be conserved across the genera. Our study has shown that a single domain 92 amino acid long prokaryotic parvulin PpiC from E.coli could complement for the function of Ess1 in Saacharomyces cerevisiae. We have also shown that under conditions of over expression the carboxy terminus of NifM from Azotobacter vinelandii could functionally replace Ess1 in S. cerevisiae. However the complete nifM was unable to do so. We have shown that the amino terminus of NifM acts as a regulatory unit not only for the PPIase activity of its carboxy terminus domain but also for the PPIase activity of PpiC and human Pin1. Using random mutagenesis we have identified the potential docking sites on amino terminus of NifM. These sites are defined as the residues which are responsible for the regulatory activity of NifM. Further we have found that FKBPs which show a high similarity with human Pin1 was unable to isomerise substrate specific to the parvulins. Our analysis has shown, the substrate binding pocket in FKBP is large due to its aromatic nature. Hence it is unable FKBPs to complement for the function of the parvulins

When I left my country of birth, India, to come to the united states in 1990, it was in search of a place where women enjoyed much greater freedom, power, and esteem than those of us from my part of the world. While I have found this to be somewhat true, I also see women struggling for recognition and representation in every aspect of life here, much as their counterparts allover the world. Does it matter that women are so underrepresented? Do women really make a difference? According to a 1992 study conducted by the center for American Woman and Politics, the Norwegian Prime Minister, Gro Brundtland, noted that in her first term as Prime Minister she appointed women to 20 percent of cabinet positions. In her second term, she appointed women to half of the cabinet roles. When asked, Do women make a difference? her response was, Absolutely, women change the agenda. India, probably the largest democracy in the world (population of nearly 100 million), and a country considered less developed in many senses, was one of the first few countries to elect a woman Prime Minister, Indira Gandhi, in 1975. The country can also boast two women governors, two women Chief Ministers of states, one woman ambassador and a fair number of women representatives in the two houses of the parliament. India\u27s immediate neighbor, Pakistan, has recently elected a woman Prime Minister, Benazir Bhutto. In 1993, more than two centuries since the birth of the country and almost 75 years since women won the right to vote, women still hold a tiny portion of political power in the united states. While the practice of democratic elections in the united states may not be unique, the role of the press and its complete freedom to discuss not only the race but the personal, past and present history of candidates, is certainly unique. with this perspective I approached this study, not only for my own interest but also hoping it would contribute to an understanding of the press and the women candidates for public office in the united states. Enhanced transmission by a grating composed of left-handed materials
We present a detailed theoretical analysis about the influence of surface polaritons on the transmission properties of electromagnetic waves at the periodically corrugated interface between the vacuum and left-handed material by using nonlinear boundary condition approach. The principle behind this approach is to match the wave fields across the grating interface by using a set of linear wave equation with nonlinear boundary conditions. The resonant transmission of the incident electromagnetic radiation in this structure is feasible within a certain frequency band, where there is a range of frequency over which both the electric permittivity and the magnetic permeability are simultaneously negative. The enhanced transmission is attributed to the coupling of the incident electromagnetic wave with the excited surface polaritons on grating interface. Effectiveness of neuromuscular exercises (NEMEX) in knee osteoarthritis: a systematic review with metaanalysis
The main objective was to evaluate the effectiveness of Neuromuscular exercises (NEMEX) on pain and function in patients with knee osteoarthritis. Pubmed, Cochrane and Pedro and Google Scholar were accessed with eligibility criteria Randomized controlled trials, single blinded controlled trial and Controlled before and after study comparing and assessing the effectiveness of NEMEX in knee osteoarthritis, articles published in English language till 2020 with NEMEX either alone or in conjunction with other interventions (drugs, educational packages) in patients with Knee osteoarthritis. Outcome measures used for meta-analysis were visual analog scale (VAS) for pain and Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) for function and the other measures Knee injury osteoarthritis outcome score (KOOS), Knee Index and Knee Adduction Moment (KAM) and Hip Disability and osteoarthritis outcome score (HOOS). The result of the metaanalysis for the included two studies showed statistically significant reduction in pain (VAS) in the NEMEX group as compared to the other group Z = 0.64, p = 0.03, I2 = 79% and no significant reduction in WOMAC scores Z = 0.70, P = 0.64.The study concludes that there was statistically significant improvement in pain (VAS) but no functional improvement (WOMAC) with the NEMEX whether used in isolation or in conjunction with other interventions was seen in the patients with knee OA. However, large numbers of studies are required to generalize the effectiveness of NEMEX in knee osteoarthritis. Implications of the study includes NEMEX can be used as a potential intervention in reducing pain, improving strength and function in knee osteoarthritis patients
Distinct subunit pairing criteria within the heterodimeric IL-12 cytokine family.
The heterodimeric IL-12 cytokine family is characterized by the sharing of three Ξ± (p19, p28, p35) and two Ξ² (p40 and Ebi3) subunits, and includes IL-12 (p35/p40), IL-23 (p19/p40), IL-27 (p28/Ebi3) and IL-35 (p35/Ebi3). In this study, the dimerization interfaces of IL-12 family members were characterized, with emphasis on IL-35. Ebi3 and p35 subunits from human and mouse paired effectively with each other, indicating there is no species barrier to IL-35 dimerization and suggesting a conserved dimerization interface. Specific p35 residues that contribute to formation of the IL-12 interface were assessed for their contribution to the IL-35 interface, and candidate Ebi3 residues were screened for their contribution to both IL-27 and IL-35 interfaces. Several residues were identified as critical to the IL-12 or IL-27 interfaces. Conversely, no single mutation was identified that completely disrupts p35/Ebi3 pairing. Linear alanine scanning mutagenesis on both p35 and Ebi3 subunits was performed, focusing on residues that are conserved between the mouse and human proteins. Additionally, a structure-based alanine-scanning approach in which mutations were clustered based on proximitiy was performed on the p35 subunit. Both approaches suggest that IL-35 has distinct criteria for subunit pairing and is remarkabley less sensitive to structural perturbation than IL-12 and IL-27. Additionally, studies using a panel of anti-p35 and anti-Ebi3 antibodies indicate differential availability of epitopes within IL-12 family members that share these subunits, suggesting that IL-35 has distinct structural features, relative to IL-12 and IL-27. These results may be useful in future directed therapeutic targeting of IL-12 family members
