    Invasive alien pests threaten the carbon stored in Europe’s forests

    Forests mitigate climate change by sequestering large amounts of carbon (C). However, forest C storage is not permanent, and large pulses of tree mortality can thwart climate mitigation efforts. Forest pests are increasingly redistributed around the globe. Yet, the potential future impact of invasive alien pests on the forest C cycle remains uncertain. Here we show that large parts of Europe could be invaded by five detrimental alien pests already under current climate. Climate change increases the potential range of alien pests particularly in Northern and Eastern Europe. We estimate the live C at risk from a potential future invasion as 1027 Tg C (10% of the European total), with a C recovery time of 34 years. We show that the impact of introduced pests could be as severe as the current natural disturbance regime in Europe, calling for increased efforts to halt the introduction and spread of invasive alien species.© The Author(s) 201

    Identification of a novel human deoxynivalenol metabolite enhancing proliferation of intestinal and urinary bladder cells

    The mycotoxin deoxynivalenol (DON) is an abundant contaminant of cereal based food and a severe issue for global food safety. We report the discovery of DON-3-sulfate as a novel human metabolite and potential new biomarker of DON exposure. The conjugate was detectable in 70% of urine samples obtained from pregnant women in Croatia. For the measurement of urinary metabolites, a highly sensitive and selective LC-MS/MS method was developed and validated. The method was also used to investigate samples from a duplicate diet survey for studying the toxicokinetics of DON-3-sulfate. To get a preliminary insight into the biological relevance of the newly discovered DON-sulfates, in vitroexperiments were performed. In contrast to DON, sulfate conjugates lacked potency to suppress protein translation. However, surprisingly we found that DON-sulfates enhanced proliferation of human HT-29 colon carcinoma cells, primary human colon epithelial cells (HCEC-1CT) and, to some extent, also T24 bladder cancer cells. A proliferative stimulus, especially in tumorigenic cells raises concern on the potential impact of DON-sulfates on consumer health. Thus, a further characterization of their toxicological relevance should be of high priority

    Deoxynivalenol (DON) is a protein synthesis inhibitor produced by the Fusarium species, which frequently contaminates grains used for human or animal consumption. We treated a wheat suspension culture with DON or one of its acetylated derivatives, 3-acetyl-DON (3-ADON), 15-acetyl-DON (15-ADON) and 3,15-diacetyl-DON (3,15-diADON), and monitored the metabolization over a course of 96 h. Supernatant and cell extract samples were analyzed using a tailored LC-MS/MS method for the quantification of DON metabolites. We report the formation of tentatively identified DON-15-O-β-D-glucoside (D15G) and of 15-acetyl-DON-3-sulfate (15-ADON3S) as novel deoxynivalenol metabolites in wheat. Furthermore, we found that the recently identified 15-acetyl-DON-3-O-β-D-glucoside (15-ADON3G) is the major metabolite produced after 15-ADON challenge. 3-ADON treatment led to a higher intracellular content of toxic metabolites after six hours compared to all other treatments. 3-ADON was exclusively metabolized into DON before phase II reactions occurred. In contrast, we found that 15-ADON was directly converted into 15-ADON3G and 15-ADON3S in addition to metabolization into deoxynivalenol-3-O-β-D-glucoside (D3G).     Implications from palaeoseismological investigations at the Markgrafneusiedl Fault (Vienna Basin, Austria) for seismic hazard assessment

    Intraplate regions characterized by low rates of seismicity are challenging for seismic hazard assessment, mainly for two reasons. Firstly, evaluation of historic earthquake catalogues may not reveal all active faults that contribute to regional seismic hazard. Secondly, slip rate determination is limited by sparse geomorphic preservation of slowly moving faults. In the Vienna Basin (Austria), moderate historical seismicity (Imax,obs/Mmax,obs=8/5.2) concentrates along the left-lateral strike-slip Vienna Basin Transfer Fault (VBTF). In contrast, several normal faults branching out from the VBTF show neither historical nor instrumental earthquake records, although geomorphological data indicate Quaternary displacement along those faults. Here, located about 15 km outside of Vienna, the Austrian capital, we present a palaeoseismological dataset of three trenches that cross one of these splay faults, the Markgrafneusiedl Fault (MF), in order to evaluate its seismic potential. Comparing the observations of the different trenches, we found evidence for five to six surface-breaking earthquakes during the last 120 kyr, with the youngest event occurring at around 14 ka. The derived surface displacements lead to magnitude estimates ranging between 6.2±0.5 and 6.8±0.4. Data can be interpreted by two possible slip models, with slip model 1 showing more regular recurrence intervals of about 20–25 kyr between the earthquakes with M≥6.5 and slip model 2 indicating that such earthquakes cluster in two time intervals in the last 120 kyr. Direct correlation between trenches favours slip model 2 as the more plausible option. Trench observations also show that structural and sedimentological records of strong earthquakes with small surface offset have only low preservation potential. Therefore, the earthquake frequency for magnitudes between 6 and 6.5 cannot be constrained by the trenching records. Vertical slip rates of 0.02–0.05 mm a−1 derived from the trenches compare well to geomorphically derived slip rates of 0.02–0.09 mm a−1. Magnitude estimates from fault dimensions suggest that the largest earthquakes observed in the trenches activated the entire fault surface of the MF including the basal detachment that links the normal fault with the VBTF. The most important implications of these palaeoseismological results for seismic hazard assessment are as follows. (1) The MF is an active seismic source, capable of rupturing the surface despite the lack of historical earthquakes. (2) The MF is kinematically and geologically equivalent to a number of other splay faults of the VBTF. It is reasonable to assume that these faults are potential sources of large earthquakes as well. The frequency of strong earthquakes near Vienna is therefore expected to be significantly higher than the earthquake frequency reconstructed for the MF alone. (3) Although rare events, the potential for earthquake magnitudes equal or greater than M=7.0 in the Vienna Basin should be considered in seismic hazard studies.© Author(s) 201

    Quicklime application instantly increases soil aggregate stability

    Agricultural intensification, especially enhanced mechanisation of soil management, can lead to the deterioration of soil structure and to compaction. A possible amelioration strategy is the application of (structural) lime. In this study, we tested the effect of two different liming materials, ie limestone (CaCO3) and quicklime (CaO), on soil aggregate stability in a 3-month greenhouse pot experiment with three agricultural soils. The liming materials were applied in the form of pulverised additives at a rate of 2 000 kg ha−1. Our results show a significant and instantaneous increase of stable aggregates after quicklime application whereas no effects were observed for limestone. Quicklime application seems to improve aggregate stability more efficiently in soils with high clay content and cation exchange capacity. In conclusion, quicklime application may be a feasible strategy for rapid improvement of aggregate stability of fine textured agricultural soils

    FI-ICP-TOFMS for quantification of biologically essential trace elements in cerebrospinal fluid – high-throughput at low sample volume

    In this work, we introduce a high-throughput quantitative multi-element method for biological fluids enabled by omitting sample preparation and an analysis time of a few seconds per sample. For the first time, flow injection of an undiluted cerebrospinal fluid (CSF) was combined to state-of-the-art ICP-TOFMS detection for multi-element analysis. Owing to the low sample volume and trace element concentrations of the CSF, flow injection methods with only 5 μL sample intake were used in combination with an icpTOF 2R TOF-based ICP-MS instrument. Due to the lack of certified reference materials for CSF analysis, a validated method employing open vessel digestion of the CSF material in combination with ICP-sectorfield-MS analysis was carried out and used as a reference. Additionally, the performance of the flow injection ICP-TOFMS was cross-validated by flow injection quadrupole-based ICP-MS/MS analysis using both external calibration and isotope dilution strategies. In the latter case, the sample had to be injected several times because of the need for tailored gas conditions for different elements. Overall, flow injection of biological fluids delivered quantitative values, which were in excellent agreement with the gold standard established by ICP-SFMS demonstrating the capability of ICP-TOFMS analysis in terms of resolution and sensitivity for the accurate quantification of trace elements in biological samples

    INO80 represses osmostress induced gene expression by resetting promoter proximal nucleosomes

    The conserved INO80 chromatin remodeling complex is involved in regulation of DNA damage repair, replication and transcription. It is commonly recruited to the transcription start region and contributes to the establishment of promoter-proximal nucleosomes. We find a substantial influence of INO80 on nucleosome dynamics and gene expression during stress induced transcription. Transcription induced by osmotic stress leads to genome-wide remodeling of promoter proximal nucleosomes. INO80 function is required for timely return of evicted nucleosomes to the 5΄ end of induced genes. Reduced INO80 function in Arp8-deficient cells leads to correlated prolonged transcription and nucleosome eviction. INO80 and the related complex SWR1 regulate incorporation of the H2A.Z isoform at promoter proximal nucleosomes. However, H2A.Z seems not to influence osmotic stress induced gene regulation. Furthermore, we show that high rates of transcription promote INO80 recruitment to promoter regions, suggesting a connection between active transcription and promoter proximal nucleosome remodeling. In addition, we find that absence of INO80 enhances bidirectional promoter activity at highly induced genes and expression of a number of stress induced transcripts. We suggest that INO80 has a direct repressive role via promoter proximal nucleosome remodeling to limit high levels of transcription in yeast.© The Author(s) 201

    Effect of Duration of High-Grain Feeding on Chewing, Feeding Behavior, and Salivary Composition in Cows with or without a Phytogenic Feed Supplement

    Switching diets from forage to a high-grain (HG) diet increases the risk of rumen fermentation disorders in cattle. However, the effects of the duration of the HG feeding, after the diet switch, on animal behavior and health have received considerably less attention. This experiment primarily aimed to assess the effects of the duration of an HG diet on the chewing, eating, and lying behavior and salivation dynamics in a control group (CON) and a group of cows receiving a phytogenic feed supplement (TRT) at 0.04% (DM basis), which included L-menthol, thymol, eugenol, mint oil, and cloves powder. The experiment was a crossover design with nine non-lactating cows, and two experimental periods with an intermediate washout of four weeks. In each period, the cows were first fed a forage diet for a week to collect baseline measurements representing week 0; then, the diet was switched over a week to HG (65% concentrate), which was fed for four continuous weeks (week 1, week 2, week 3, and week 4 on an HG diet, respectively). The cows were divided in two groups of four and five animals and were randomly allocated to CON or TRT. The data analysis revealed that at the start of the HG feeding, the dry matter intake and the cows\u27 number of lying bouts increased, but the eating time, rumination time, and meal frequency decreased, resulting in a greater eating rate. We also found that an advanced duration on an HG diet further decreased the rumination time, total chewing time, chewing index, and sorting in favor of short feed particles, with the lowest values in week 4. The feed bolus size increased but feed the ensalivation decreased in week 4 compared to week 0. The dietary switch increased salivary lysozyme activity, and the advanced duration on the HG diet increased salivary pH, but salivary phosphate decreased in weeks 1 and 2 on the HG diet. Supplementation with TRT increased sorting in favor of physically effective NDF (peNDF) in week 2 and increased salivary pH in week 4 on an HG diet. Overall, the negative effects of the HG diet in cattle are more pronounced during the initial stage of the HG feeding. However, several detrimental effects were exacerbated with the cows\u27 advanced duration on feed, with host adaptive changes still observed after 3 and 4 weeks following the diet switch. The TRT mitigated some of the negative effects through the temporal improvement of the salivary properties and the intake of peNDF, which are known to modulate rumen fermentation

    A meta-analysis of shrinking cities in Europe and Japan. Towards an integrative research agenda

    Empirical research on urban shrinkage is being conducted around the globe, since many countries are confronted with the phenomenon of shrinking cities. So far, the research on urban shrinkage has focused strongly on case studies, which is why we can benefit from a diverse and empirically rich knowledge base on the phenomenon and its regional manifestations. By bridging and comparing the European and Japanese academic discourse, we aim to identify the different recurring theories and key issues discussed under the umbrella term ‘urban shrinkage’ and strive to uncover blind spots of the debate. For this purpose, we conduct a qualitative meta-analysis of 100 empirical cases that are documented in the literature dealing with shrinking cities in the EU and Japan. This meta-analysis is based on comparative qualitative content analysis. It reveals a regionally differentiated pattern of various causes, effects and responses documented for shrinking cities in Western, Mediterranean and post-socialist EU countries and in Japan. Based on these findings, we offer an agenda for future research by suggesting an integrative perspective on the context-specific dynamics of urban shrinkage. We argue for an integrative understanding of shrinking cities in order to develop a valid knowledge base for evidence-based policy recommendations.© 2019 The Author(s
