Immunomodulatory properties of recombinant human granulocyte-macrophage colony-stimulating factor
Granulocyte-macrophage colony stimulating factor (GM-CSF) is a myelopoietic growth factor that exerts pleiotropic effect not only on the differentiation of immature progenitor cells into polymorphonuclear neutrophils, monocytes/macrophages and dendritic cells, but also controls the functioning of differentiated cells. GM-CSF is currently being investigated in clinical trials as an immunomodulator and adjuvant. However, a wide range of biological activities and, sometimes, paradoxical effects of this cytokine require more thorough studies of its action, in order to predict its efficacy under different conditions of immunotherapy. In this work, we have studied the effect of recombinant human GM-CSF on metabolic activity of mouse peritoneal exudate cells in primary cell cultures. Metabolic (redox) activity of the cells was assessed by their ability to reduce nitroblue tetrazolium (NBT) in the course of MF- and Fc-dependent phagocytosis triggered by addition of opsonized zymosan, or sheep erythrocytes to the culture medium. We have shown the dose-dependent stimulatory effect of GM-CSF on the oxidative metabolism of phagocytic peritoneal macrophages and neutrophils. Upon culturing the pepton-elicited cells at wide range of GM-CSF concentrations (5 to 40,000 ng/mL) for 2 and 24 hours, a more pronounced effect of the substance was observed for neutrophils. The GM-CSF preparation caused a significant increase (by 13-17%) in the redox activity of neutrophils induced by opsonized zymosan that persisted at a low dose range, and was retained after 24 hours. The stimulatory effect of GM-CSF on macrophages with NBT index increase by 16% was observed in the short-term cultures. In general, the elicited cells of both types showed a more pronounced response to lower concentrations of GM-CSF (5-125 ng/mL), and weaker effect at higher doses of the preparation. A similar dependence was found when studying the resident macrophages. Culturing of resident cells with GM-CSF at the doses of 5,000 to 40,000 ng/mL for 24 hours caused a significantly increased redox activity of the cells induced by zymosan, or sheep erythrocytes (by 33-52%). In both cases, the maximal response was detected at a dose of 5,000 ng/mL and decreased with increasing dose. The stimulatory effect of GM-CSF upon resident macrophages was more pronounced as compared to elicited cells, which was characterized by the prolonged period of cell activation (up to 24 hours of culture). The data obtained are of interest, in view of prospective usage of GM-CSF as a component of immunomodulatory and adjuvant therapy for various infectious diseases
Features of model coronaviruses distribution in feline organs and tissues in the context of COVID-19 pathogenesis study
To date, the reisreason to believe that, unlike classical a cuterespiratory virusinfections caused by adenoviruses, rhinoviruses, orthomyxoviruses, COVID-19 behaves completely differently. Firstly, the pathological process esare likely to be immune-mediated and the immun esystem quites lowly ensurest heelimination of the virus from the organism. Secondly, the dynamics of the diseases ymptom development and the duration of intestinal virusshedding after recovery give reason to believe that theSARS-CoV-2 infection is mainly localizedin the intestine. A possible reason isthat in the presence of proteolyticenzymes, viral particlesmature, hydrophilic aminoacids are removed from the surface of the virion, making it more hydrophobic and able to ad here to cells due to hydrophobic interactions. The presence of the ACE2 recept or mainly in the enterocytes of the ileumdoes not exclude the accumulation of coronavirusin lymphocytes, given that there are more lymphocytes in the gastrointestinal tract than anywhere else, this fact can beconsidered as another justification for the predominant accumulation of coronaviruses, including SARS-CoV-2 in the intestine. A distinctive feature of feline coronavirus infection and, in particular, infectious feline peritonitis, from human COVID-19 infection was considered to be the presence of effusion peritonitis as the main complication leading to death, while respiratory and card iovascular in sufficiency is more characteristic for humans. Never the less, cases of serous peritonitis in humans infected with COVID-19 have already been described. In the context of the analyzed model, theclinical case describedin the study allows principal possibility of exacerbation of chronic coronavirus infection in caseof re-infection (superinfection) and development of apredominantlylocal infection
Comparison of physicochemical properties of tooth paste with peptiids and simple paste without peptides
Based on experimental data, a comparative analysis of a number of physicochemicalparameters of toothpastes with peptides and similar in composition pastes without peptides was made. The effect of the peptide component on surfactants and the buffer capacity of aqueous toothpaste suspensions are shown. The influence of the physicochemical properties of toothpastes on the health of the oral cavity is described.На основании экспериментальных данных сделан сравнительный анализ ряда физико-химических показателей зубных паст с пептидами и аналогичных по составу паст без пептидов. Показано влияние пептидного компонента на поверхностно-активные свойства и буферную емкость водных суспензий зубных паст. Use of the Clinpro tm XT Varnish protective coating as a remineralizing therapy for increased tooth abrasion
The article provides an analysis of experimental data obtained in the study of the drug “Clinpro TM XT Varnish” (3M ESPE), which is used as remineralizing therapy for increased destruction of the surface of hard tooth tissues. Before the study, the chemical composition of the surface layers of tooth enamel was determined by scanning electron microscopy with a Jeol JSM-6390LV apparatus. To increase the concentration of the mineral component of enamel, remgel was applied by application.
