    Electron paramagnetic resonance study of tumor affected bone marrow

    To study the mechanism of interaction of tumor cells with bone marrow cells continuous wave electron paramagnetic resonance (EPR) experiments at 9 GHz including a spin trapping of superoxide were carried out. The common features of the EPR spectra in healthy and tumor affected tissues of donors and rats as well as their difference are presented and discussed. It is proposed that labile iron pool plays a significant role in mechanisms of tumor invasion. We hope that the observed EPR features could be used to study the mechanisms of invasion and progression of tumor in different organs. © 2013 Springer Science+Business Media Dordrecht

    Superoxide- and NO-dependent mechanisms of the reprogramming of bone marrow cells by tumor cells

Electron paramagnetic resonance (EPR) experiments in vitro; spin trapping of the reactive oxygen/nitrogen species (superoxide radicals and nitric oxide, NO); gel zymography measurements in the tumor tissues, in the healthy and tumor-affected bone marrow (BM) samples of rats are carried out. The superoxide and NO generation rates are derived; matrix metalloproteinases (MMP-2 and MMP- 9) concentrations are measured. Their changes after the incubation of BM samples with Guérin carcinoma cells at 37 °C are defined. It is shown that the impact of tumor cells on BM manifests in the metabolic disorder, increased concentrations of active forms of MMP-2 and MMP-9, increased production of superoxide and NO radicals. Correlation between the appearance and intensity of the broad EPR signal at g = 2.2–2.4 with the concentrations of active forms of MMP-2 and MMP-9, NO and superoxide radicals’ rates is observed. The obtained spatial and temporal changes of the measured parameters demonstrate the usefulness of the potential application of EPR imaging to study the mechanisms of tumor invasion. The EPR signal may indicate the presence of distant metastases, may become a part of diagnostics and used for the estimation of the therapeutic treatments in the preclinical studies. It is proposed that labile iron pool is responsible for the appearance of the EPR signal in tumor and BM samples

ROS can activate matrix metalloproteinases (MMP), damage DNA and RNA. Therefore, the levels of MMP, ROS and RNS can serve as additional prognostic markers and for the estimation of the effectiveness of tumor therapy. Concerning gastric cancer, the prognostic role of MMP, its connection with the cancer staging remains controversial and correlations between the activity of MMP with the ROS and RNS levels are insufficiently confirmed. Superoxide generation rates, nitric oxide (NO) levels, concentrations of active forms of matrix metalloproteinases MMP-2 and MMP-9 in tumor and adjacent tissues of patients with stomach cancer at different disease stages were measured by electron spin resonance (ESR) including spin-trapping and polyacrylamide gel zymography. It is shown that the activity of MMP-2 and MMP-9 in tumor tissue correlate with the superoxide radicals generation rate and NO levels (r = 0.48÷0.67, p < 0.05). The activity of MMP-2 and MMP-9 in tumor tissues and superoxide radical generation rates correlate positively with the stage of regional dissemination (r = 0.45 and 0.37, correspondingly, p < 0.05), but MMP-2 and MMP-9 activity inversely depends on distant metastatic degree of stomach cancer (r = 0.58; p < 0.05). Additionally, the feasibility of ESR to locally determine oxidative stress is demonstrated

    Modulation of ROS/NO production by murine peritoneal macrophages in response to bacterial CPG DNA stimulation

    Aim: To investigate the features of metabolic activation induced by bacterial CpG DNA (bCpG DNA) in peritoneal macrophages (PMjs). Methods: Electron paramagnetic resonance spin-trapping technique using respective spin traps was applied to study the generation rate of reactive oxygen species and NO production by PMjs of BALB/c mice. Results: For the first time the capability of bCpG DNA isolated from Bacillus subtilis GP1-807-03 culture medium to elevate activity of NADP·H oxidase and inducible NO synthase in PMjs of normal and tumor-bearing mice have been demonstrated. The main differences in superoxide anion generation rate and production of NO by PMjs of normal mice and mice with transplanted solid Ehrlich carcinoma were showed. The effects of bCpG DNA stimulation in vitro on ROS and NO production by PMjs depended on concentration and time exposure with bCpG DNA. Furthermore, response of PMjs from tumor-bearing mice on bCpG DNA stimulation was delayed as compared to PMjs of normal mice. Conclusion: The present findings suggest that bCpG DNA have modulatory effect on ROS/NO production by PMjs from normal and tumor-bearing animals.Цель: изучить особенности метаболической активации перитонеальных макрофагов (ПМФ) в ответ на воздействие, вызванное CpG ДНК бактериального происхождения. Методы: для исследования скорости генерирования радикальных форм кислорода (РФК) и продукции оксида азота (NO) ПМФ мышей линии BALB/c был применен метод электронного парамагнитного резонанса с использованием спиновых ловушек. Результаты: впервые в эксперименте продемонстрирована способность бактериальной CpG ДНК, выделенной из культуральной жидкости Bacillus subtilis GP1-807-03, повышать активность НАДФ·Н-оксидазы и индуцибельной NO-синтазы ПМФ у мышей с солидной карциномой Эрлиха и интактных мышей. Отмечены различия в скорости генерирования супероксидных радикал-анионов и уровне продуцирования NO ПМФ у животных с карциномой Эрлиха по сравнению с контрольными животными, что выражалось в замедлении ответа на воздействие бактериальной CpG ДНК. Выявлена концентрационная и временная зависимость влияния бактериальной CpG ДНК in vitro на продукцию РФК и NO ПМФ. Выводы: полученные данные свидетельствуют о том, что бактериальная CpG ДНК обладает модулирующим влиянием на продукцию РФК и NO ПМФ in vitro и in vivo
