    Evaluation of acetylcholinesterase and butyrylcholinesterase inhibitory activity of Huperzine-A; in silico and in vitro studies

    The present study is focused on exploring the Acetylcholinesterase and Butyrylcholinesterase inhibitory activity of Huperzine-A in silico and in vitro. In this study, Huperzine-A-A was docked with Acetylcholinesterase and Butyrylcholinesterase. Docking studies revealed the excellent interaction of Huperzine-A-A with these targets. The result of present study provides insight for the in vitro studies. The in vitro studies the enzyme kinetics of Huperzine-A-A via Lineweaver brooks plot revealed the kinetics and non-competitive inhibitory nature of the later. Further studies on Huperzine-A-A are necessary to develop and establish its role on brain cholinergic system and cognitive deficits which may serve a stepping stone in CNS medication

    Biodeterioration of Polyethylene High Density by Aspergillus versicolor and Aspergillus terreus

    The present article investigates the biodeterioration ability of polyethylene high density by Aspergillus versicolor and Aspergillus terreus. Shake flask incubation for 30 days was done for the purpose of biodeterioration of polyethylene high density. The observations were recorded for initial and final dry weight of polyethylene high density before and after incubation in the culture medium were compared the percentage of biodeterioration of polyethylene. The strongest ability was observed by Aspergillus terreus with 7.59% biodegradability followed by Aspergillus versicolor with 3.79% of biodegradability. In conclusion, Aspergillus terreus was found to be efficient organism that degraded the polyethylene high density with maximum biodegradability as compared to Aspergillus versicolor

    Comparative study of various tocolytics in preterm labour

    Background: Preterm birth is one of the major factors responsible for neonatal mortality and morbidity. In this study, we have to see the role of various tocolytics in preventing preterm labour.Methods: The observational study was conducted on 200 patients admitted in Upper India Sugar Exchange Maternity Hospital, Kanpur.Results: In our study calcium channel blocker were associated with statistically and clinically significantly better neonatal outcome and fewer maternal side effects than any other tocolytic. So calcium channel blocker would be preferred first line tocolytic with regard to several important outcomes.Conclusions: Preterm birth continued to be problem for obstetrician. Administration of short term tocolytic agents can prolong pregnancy for women in pre-term labor. Prolonging pregnancy may have benefits because it allows for other proven interventions, such as antenatal corticosteroid administration to be accomplished. The reviews proven an overview currently utilize tocolytic agents and the evidence of demonstrating their efficacy for prolonging pregnancy by at least 48 hours

    Hyperhomocysteinemia: a risk factor in unexplained infertility

    Background: To study the role of hyperhomocysteinemia in unexplained infertility and the impact of its correction with vitamin and mineral supplementation.Methods: Total of 60 patients were included with 30 patients in case and 30 patients in control groups. Cases included patients with history of inability to conceive with frequent regular unprotected intercourse for at least 1 year. Controls included age matched parous females with at least one live birth and no history of abortions. Among the patients of unexplained infertility with hyperhomocysteinemia, homocysteine lowering agents were given and outcome studied in the form of lowering of homocysteine levels and number of conceptions.Results: The mean age was 28.1 years in study and 29.5 years in the control group .Mean level of serum homocysteine was significantly higher in study group than normal fertile women i.e.20.5µmol/l and 10.9 µmol/l respectively. Among the patients of unexplained infertility, 22(73.3%) were found to have range above the normal healthy levels. In these patients homocysteine lowering agents were given for 6 weeks and lowering of mean homocysteine levels was observed which was 10.4 µmol/l.  Six (27.3%) patients conceived spontaneously during the follow up period.Conclusions: Serum homocysteine levels are inversely correlated with infertility. Homocysteine lowering agents have a favourable impact on the outcome of infertility and their use is suggested in cases of unexplained infertility associated with hyperhomocysteinemia.

    Umbilical cord cyst: a diagnostic dilemma

    Umbilical cord cyst refers to any cystic lesion associated with the umbilical cord. Cord cysts can be defined as true or false cysts and may occur in any location along the cord. They are irregular in shape and are located between the cord vessels. Authors are reporting the case of an infant with an umbilical cord tumor which had twice been misdiagnosed previously as a hemangioma, based on ultrasound image of its cystic and solid component with good vascular supply. The ultrasound image most likely suggestive of a hemangioma as a differential diagnosis led to caesarean section in our patient (based on large size of the lesion and fear of rupture of same during process of labour). The definitive diagnosis was made only after birth of the baby. Final diagnosis of true umbilical cord cyst was made after histopathological examination. Thus, there can be confusion in the diagnosis between umbilical cord hemangiomas and umbilical cord cysts based on ultrasound

    Synergistic effect of folic acid and galantamine against experimentally induced oxidative stress in IMR 32 cells

    286-292Galantamine is an active constituent obtained from Galanthus nivalis L., a traditional herb known for its pharmacological properties, particularly nootropic effect. Folic acid is a dietary supplement that enhances neuronal activity. Effect of galantamine and folic acid on human neuronal cells is well known. In the present study, we explored the protective effect of galantamine and folic acid, both independently as well as in combination, over antioxidant defence system and nootropic effects on human neuroblastoma cells IMR-32. The treatment galantamine, folic acid and their combination was given for 24 h and cytotoxicity study was carried out by trypan blue dye exclusion assay. Apoptosis and necrosis were observed using Propidium iodide (PI) and Hoechst double staining method. Biochemical assays viz. total protein, protein carbonyl, lipid peroxidation and glutathione were analyzed along with super oxide dismutase and catalase. Result of cytotoxicity showed dose dependent increase in percent viability and significant decrease was observed in apoptosis and necrosis. Moreover, exposure to Galantamine, Folic acid and their combination significantly decreased lipid peroxidation and protein carbonyl formation along with the enhancement in antioxidant defence mechanism. Findings of these dose reliant toxicity study of Galantamine , Folic acid and their combination suggest that these has higher potency when given together and shows synergistic effect. They also causes repair of human neuronal cells IMR-32 cells enhancing the cell viability and consumption of Galantamine and Folic acid together will help in prevention of CNS disorders and neurodegeneration
