    Testing the Underlying Chemical Principles of the Biotic Ligand Model (BLM) to Marine Copper Systems: Measuring Copper Speciation Using Fluorescence Quenching

    Speciation of copper in marine systems strongly influences the ability of copper to cause toxicity. Natural organic matter (NOM) contains many binding sites which provides a protective effect on copper toxicity. The purpose of this study was to characterize copper binding with NOM using fluorescence quenching techniques. Fluorescence quenching of NOM with copper was performed on nine sea water samples. The resulting stability con- stants and binding capacities were consistent with literature values of marine NOM, show- ing strong binding with log K values from 7.64 to 10.2 and binding capacities ranging from 15 to 3110 nmole mg C −1 . Free copper concentrations estimated at total dissolved copper concentrations corresponding to previously published rotifer effect concentrations, in the same nine samples, were statistically the same as the range of free copper calculated for the effect concentration in NOM-free artificial seawater. These data confirms the applicability of fluorescence spectroscopy techniques for NOM and copper speciation characterization in sea water and demonstrates that such measured speciation is consistent with the chemical principles underlying the Biotic Ligand Model (BLM) approach for bioavailability-based metals risk assessment

    Surface-slip equations for multicomponent, nonequilibrium air flow

    Equations are presented for the surface slip (or jump) values of species concentration, pressure, velocity, and temperature in the low-Reynolds-number, high-altitude flight regime of a space vehicle. These are obtained from closed-form solutions of the mass, momentum, and energy flux equations using the Chapman-Enskog velocity distribution function. This function represents a solution of the Boltzmann equation in the Navier-Stokes approximation. The analysis, obtained for nonequilibrium multicomponent air flow, includes the finite-rate surface catalytic recombination and changes in the internal energy during reflection from the surface. Expressions for the various slip quantities have been obtained in a form which can readily be employed in flow-field computations. A consistent set of equations is provided for multicomponent, binary, and single species mixtures. Expression is also provided for the finite-rate species-concentration boundary condition for a multicomponent mixture in absence of slip

    A debris/ice Thermal Protection System (TPS) assessment and photographic analysis was conducted for Space Station Mission STS-43. Debris inspections of the flight elements and launch pad were performed before and after launch. Ice/frost conditions on the External Tank (ET) were assessed by the use of computer programs, nomographs, and infrared scanner data during cryogenic loading of the vehicle followed by on-pad visual inspection. High speed photography was analyzed after launch to identify ice/debris sources and to evaluate potential vehicle damage and/or in-flight anomalies

    Upwind MacCormack Euler solver with non-equilibrium chemistry

    A computer code, designated UMPIRE, is currently under development to solve the Euler equations in two dimensions with non-equilibrium chemistry. UMPIRE employs an explicit MacCormack algorithm with dissipation introduced via Roe's flux-difference split upwind method. The code also has the capability to employ a point-implicit methodology for flows where stiffness is introduced through the chemical source term. A technique consisting of diagonal sweeps across the computational domain from each corner is presented, which is used to reduce storage and execution requirements. Results depicting one dimensional shock tube flow for both calorically perfect gas and thermally perfect, dissociating nitrogen are presented to verify current capabilities of the program. Also, computational results from a chemical reactor vessel with no fluid dynamic effects are presented to check the chemistry capability and to verify the point implicit strategy

    Developing the Quantitative Histopathology Image Ontology : A case study using the hot spot detection problem

    Interoperability across data sets is a key challenge for quantitative histopathological imaging. There is a need for an ontology that can support effective merging of pathological image data with associated clinical and demographic data. To foster organized, cross-disciplinary, information-driven collaborations in the pathological imaging field, we propose to develop an ontology to represent imaging data and methods used in pathological imaging and analysis, and call it Quantitative Histopathological Imaging Ontology – QHIO. We apply QHIO to breast cancer hot-spot detection with the goal of enhancing reliability of detection by promoting the sharing of data between image analysts

    Precision cosmology defeats void models for acceleration

    The suggestion that we occupy a privileged position near the centre of a large, nonlinear, and nearly spherical void has recently attracted much attention as an alternative to dark energy. Putting aside the philosophical problems with this scenario, we perform the most complete and up-to-date comparison with cosmological data. We use supernovae and the full cosmic microwave background spectrum as the basis of our analysis. We also include constraints from radial baryonic acoustic oscillations, the local Hubble rate, age, big bang nucleosynthesis, the Compton y-distortion, and for the first time include the local amplitude of matter fluctuations, \sigma_8. These all paint a consistent picture in which voids are in severe tension with the data. In particular, void models predict a very low local Hubble rate, suffer from an "old age problem", and predict much less local structure than is observed.Comment: 22 pages, 12 figures; v2 adds models in closed backgrounds; conclusions strengthened; version accepted to Phys.     Can we avoid dark energy?

    The idea that we live near the centre of a large, nonlinear void has attracted attention recently as an alternative to dark energy or modified gravity. We show that an appropriate void profile can fit both the latest cosmic microwave background and supernova data. However, this requires either a fine-tuned primordial spectrum or a Hubble rate so low as to rule these models out. We also show that measurements of the radial baryon acoustic scale can provide very strong constraints.     Molecular Basis for Kir6.2 Channel Inhibition by Adenine Nucleotides

    AbstractKATP channels are comprised of a pore-forming protein, Kir6.x, and the sulfonylurea receptor, SURx. Interaction of adenine nucleotides with Kir6.2 positively charged amino acids such as K185 and R201 on the C-terminus causes channel closure. Substitution of these amino acids with other positively charged residues had small effects on inhibition by adenine nucleotide, while substitution with neutral or negative residues had major effects, suggesting electrostatic interactions between Kir6.2 positive charges and adenine nucleotide negative phosphate groups. Furthermore, R201 mutation decreased channel sensitivity to ATP, ADP, and AMP to a similar extent, but K185 mutation decreased primarily ATP and ADP sensitivity, leaving the AMP sensitivity relatively unaffected. Thus, channel inhibition by ATP may involve interaction of the α-phosphate with R201 and interaction of the β-phosphate with K185. In addition, decreased open probability due to rundown or sulfonylureas caused an increase in ATP sensitivity in the K185 mutant, but not in the R201 mutant. Thus, the β-phosphate may bind in a state-independent fashion to K185 to destabilize channel openings, while R201 interacts with the α-phosphate to stabilize a channel closed configuration. Substitution of R192 on the C-terminus and R50 on the N-terminus with different charged residues also affected ATP sensitivity.
