    Aligned Across Difference: Structural Injustice, Sex Work, and Human Trafficking

    Feminist scholars and activists engage in meaningful, contentious debates about the relationships among sex, gender, power, and society. One of the most recent iterations of these arguments reinscribes the pleasure of sex positivity and danger of patriarchal exploitation onto new subjects: sex work and human trafficking. This paper brings together two separate empirically based research projects, one working with sex workers and the other working with members of the anti-trafficking community. As scholars working across these topics, we provide new normative propositions that may bridge these different approaches to resilience, survival, danger, and risk. We find that the real threat identified by our participants was the wide reach of the carceral state onto migrating, working, and trafficked bodies. Our projects find unexpected commonality in shared perceptions of pleasure, agency, and danger among sex workers, human trafficking survivors, and service providers working with trafficked persons. Current debates ignore the lived experiences of our participants, who attempt to find pleasure in context-specific agency and survival, and who locate danger in the looming forces of the security state, criminality, and structural inequalities

    Augmentation of response to nab-paclitaxel by inhibition of insulin-like growth factor (IGF) signaling in preclinical pancreatic cancer models

    Nab-paclitaxel has recently shown greater efficacy in pancreatic ductal adenocarcinoma (PDAC). Insulin like growth factor (IGF) signaling proteins are frequently overexpressed in PDAC and correlate with aggressive tumor phenotype and poor prognosis. We evaluated the improvement in nab-paclitaxel response by addition of BMS-754807, a small molecule inhibitor of IGF-1R/IR signaling, in preclinical PDAC models. In subcutaneous xenografts using AsPC-1 cells, average net tumor growth in different therapy groups was 248.3 mm3 in controls, 42.4 mm3 after nab-paclitaxel (p = 0.002), 93.3 mm3 after BMS-754807 (p = 0.01) and 1.9 mm3 after nab-paclitaxel plus BMS-754807 (p = 0.0002). In subcutaneous xenografts using Panc-1 cells, average net tumor growth in different therapy groups was: 294.3 mm3 in controls, 23.1 mm3 after nab-paclitaxel (p = 0.002), 118.2 mm3 after BMS-754807 (p = 0.02) and -87.4 mm3 (tumor regression) after nab-paclitaxel plus BMS-754807 (p = 0.0001). In peritoneal dissemination model using AsPC-1 cells, median animal survival was increased compared to controls (21 days) after therapy with nab-paclitaxel (40 days, a 90% increase, p = 0.002), BMS-754807 (27 days, a 29% increase, p = 0.01) and nab-paclitaxel plus BMS-754807 (47 days, a 124% increase, p = 0.005), respectively. Decrease in proliferation and increase in apoptosis by nab-paclitaxel and BMS-754807 therapy correlated with their in vivo antitumor activity. In vitro analysis revealed that the addition of IC25 dose of BMS-754807 decreased the nab-paclitaxel IC50 of PDAC cell lines. BMS-754807 therapy decreased phospho-IGF-1R/IR and phospho-AKT expression, and increased cleavage of caspase-3 and PARP-1. These results support the potential of BMS-754807 in combination with nab-paclitaxel as an effective targeting option for pancreatic cancer therapy

    Semantically-Enhanced Information Extraction

    Information Extraction using Natural Language Processing (NLP) produces entities along with some of the relationships that may exist among them. To be semantically useful, however, such discrete extractions must be put into context through some form of intelligent analysis. This paper1,2 offers a two-part architecture that employs the statistical methods of traditional NLP to extract discrete information elements in a relatively domain-agnostic manner, which are then injected into an inference-enabled environment where they can be semantically analyzed. Within this semantic environment, extractions are woven into the contextual fabric of a user-provided, domain-centric ontology where users together with user-provided logic can analyze these extractions within a more contextually complete picture. Our demonstration system infers the possibility of a terrorist plot by extracting key events and relationships from a collection of news articles and intelligence reports

    Enhancing the Connectedness Between Undergraduate Students, Faculty, and the DPT Program Within the Same Institution

    Background Minimal data exists on optimizing the retention of undergraduate students to the DPT program within the same academic institution. Many DPT programs provide information and resources for applicants to explain admission requirements and overview the program curriculum. Purpose Identify and understand undergraduate student and faculty member’s knowledge, perceptions and resources regarding the DPT program within the same institution. Develop resources and tools to enhance the connectedness within the undergraduate and graduate DPT program in the same institution and fill identified gaps. Participants 41 undergraduate Science and Kinesiology students 6 Kinesiology faculty members 5 Science faculty members 4 active DPT students who attended CSP as undergraduates 18 active DPT students Methods Qualitative data collection via surveys and focus groups Triangulation of data, coding and development of themes Action research strategies used to create tangible products and resources Results Majority of undergraduate students choose their major in high school or freshman year of college. Undergraduate students expressed a lack of resources and understanding of the DPT program within their institution (Figure 1). Undergraduate students expressed desire for admission requirements, informational meetings and mentorship (Figure 2). Faculty echoed lack of resources and provided ideas for potential products. Conclusion Themes emerged that students and faculty were provided with little information regarding the DPT program within their home institution. Analysis of student and staff focus groups, questionnaires, and interviews, determined the need for a variety of products including: Program resource guide, Mentorship program, Social media (Facebook & Instagram), On campus marketing, and informational events. Implications Designing and implementing products may help bridge the gap between the undergraduate and graduate populations in the same institution. Future research will examine the impact of products and to also determine if graduate applications within institution increased. Future phases may include admissions, undergraduate student advisors and other CSP undergraduate departments

    Generation and analysis of transcriptomic resources for a model system on the rise: the sea anemone Aiptasia pallida and its dinoflagellate endosymbiont

    <p>Abstract</p> <p>Background</p> <p>The most diverse marine ecosystems, coral reefs, depend upon a functional symbiosis between cnidarian hosts and unicellular dinoflagellate algae. The molecular mechanisms underlying the establishment, maintenance, and breakdown of the symbiotic partnership are, however, not well understood. Efforts to dissect these questions have been slow, as corals are notoriously difficult to work with. In order to expedite this field of research, we generated and analyzed a collection of expressed sequence tags (ESTs) from the sea anemone <it>Aiptasia pallida </it>and its dinoflagellate symbiont (<it>Symbiodinium </it>sp.), a system that is gaining popularity as a model to study cellular, molecular, and genomic questions related to cnidarian-dinoflagellate symbioses.</p> <p>Results</p> <p>A set of 4,925 unique sequences (UniSeqs) comprising 1,427 clusters of 2 or more ESTs (contigs) and 3,498 unclustered ESTs (singletons) was generated by analyzing 10,285 high-quality ESTs from a mixed host/symbiont cDNA library. Using a BLAST-based approach to predict which unique sequences derived from the host versus symbiont genomes, we found that the contribution of the symbiont genome to the transcriptome was surprisingly small (1.6–6.4%). This may reflect low levels of gene expression in the symbionts, low coverage of alveolate genes in the sequence databases, a small number of symbiont cells relative to the total cellular content of the anemones, or failure to adequately lyse symbiont cells. Furthermore, we were able to identify groups of genes that are known or likely to play a role in cnidarian-dinoflagellate symbioses, including oxidative stress pathways that emerged as a prominent biological feature of this transcriptome. All ESTs and UniSeqs along with annotation results and other tools have been made accessible through the implementation of a publicly accessible database named AiptasiaBase.</p> <p>Conclusion</p> <p>We have established the first large-scale transcriptomic resource for <it>Aiptasia pallida </it>and its dinoflagellate symbiont. These data provide researchers with tools to study questions related to cnidarian-dinoflagellate symbioses on a molecular, cellular, and genomic level. This groundwork represents a crucial step towards the establishment of a tractable model system that can be utilized to better understand cnidarian-dinoflagellate symbioses. With the advent of next-generation sequencing methods, the transcriptomic inventory of <it>A. pallida </it>and its symbiont, and thus the extent of AiptasiaBase, should expand dramatically in the near future.</p

    Recommendations to Optimize the Use of Volumetric MRI in Huntington's Disease Clinical Trials

    Volumetric magnetic resonance imaging (vMRI) has been widely studied in Huntington's disease (HD) and is commonly used to assess treatment effects on brain atrophy in interventional trials. Global and regional trajectories of brain atrophy in HD, with early involvement of striatal regions, are becoming increasingly understood. However, there remains heterogeneity in the methods used and a lack of widely-accessible multisite, longitudinal, normative datasets in HD. Consensus for standardized practices for data acquisition, analysis, sharing, and reporting will strengthen the interpretation of vMRI results and facilitate their adoption as part of a pathobiological disease staging system. The Huntington's Disease Regulatory Science Consortium (HD-RSC) currently comprises 37 member organizations and is dedicated to building a regulatory science strategy to expedite the approval of HD therapeutics. Here, we propose four recommendations to address vMRI standardization in HD research: (1) a checklist of standardized practices for the use of vMRI in clinical research and for reporting results; (2) targeted research projects to evaluate advanced vMRI methodologies in HD; (3) the definition of standard MRI-based anatomical boundaries for key brain structures in HD, plus the creation of a standard reference dataset to benchmark vMRI data analysis methods; and (4) broad access to raw images and derived data from both observational studies and interventional trials, coded to protect participant identity. In concert, these recommendations will enable a better understanding of disease progression and increase confidence in the use of vMRI for drug development

    U-Pb geochronology and paleogeography of the Valanginian–Hauterivian Neuquén Basin: Implications for Gondwana-scale source areas

    Sedimentary basins located at the margins of continents act as the final base level for con­tinental-scale catchments that are sometimes located thousands of kilometers away from the basin, and this condition of exceptionally long sediment transfer zones is probably reinforced in supercontinents, such as Gondwana. One of the most prominent marine basins in southwestern Gondwana during the Jurassic and Early Cretaceous was the Neuquén Basin (west-central Argentina), but its role as a sediment repository of far-flung source areas has not been extensively considered. This contribution provides the first detailed detrital-zircon U-Pb geochronology of the Valanginian–Hauterivian Pilmatué Member of the Agrio Formation, which is combined with sedimentology and paleogeographic reconstructions of the unit within the Neuquén Basin for a better understanding of the fluvial delivery systems. Our detrital-zircon signatures suggest that Triassic–Permian zircon populations were probably sourced from the adjacent western sector of the North Patagonian Massif, whereas Early Jurassic, Cambrian, Ordovician, and Proterozoic grains were most likely derived from farther east, in the eastern sector of the North Patagonian Massif, as well as presently remote terranes such as the Saldania Belt in southern Africa. We thus propose a Valanginian–Hauterivian longitudinal delivery system that, starting in the mid-continent region of southwestern Gondwana and by effective sorting, was bringing fine-grained or finer caliber sand to the Neuquén Basin shoreline. This delivery system was probably active (though not necessarily continuously) from Early Jurassic to Early Cretaceous until finally coming to an end during the opening of the South Atlantic Ocean in the latest Early Cretaceous.Centro de Investigaciones Geológica

    A Unifying Gravity Framework for Dispersal

    Most organisms disperse at some life-history stage, but different research traditions to study dispersal have evolved in botany, zoology, and epidemiology. In this paper, we synthesize concepts, principles, patterns, and processes in dispersal across organisms. We suggest a consistent conceptual framework for dispersal, which utilizes generalized gravity models. This framework will facilitate communication among research traditions, guide the development of dispersal models for theoretical and applied ecology, and enable common representation across taxonomic groups, encapsulating processes at the source and destination of movement, as well as during the intervening relocation process, while allowing each of these stages in the dispersal process to be addressed separately and in relevant detail. For different research traditions, certain parts of the dispersal process are less studied than others (e.g., seed release processes in plants and termination of dispersal in terrestrial and aquatic animals). The generalized gravity model can serve as a unifying framework for such processes, because it captures the general conceptual and formal components of any dispersal process, no matter what the relevant biological timescale involved. We illustrate the use of the framework with examples of passive (a plant), active (an animal), and vectored (a fungus) dispersal, and point out promising applications, including studies of dispersal mechanisms, total dispersal kernels, and spatial population dynamics
