    The purpose of this project was to validate Tetrahymena as a test organism for ecotoxicological risk assessment. The first step in this validation procedure was to select suitable candidate tests with regard to sensitivity, standardization and ecological relevance. 5 aniline derivatives were tested for their effects on 11 different physiological, biochemical and behavioural endpoints. The three testsystems proliferation rate, 48h population density and the chemosensory response were selected and further validated using a heterogeneous set of 45 existing chemicals randomly chosen from EINECS. In this second step of the validation procedure almost identical results were obtained with both growth parameters as cytotoxicological endpoints. No significant correlation exists between the growth assays and the chemosensory response, indicating a different underlying mechanism of toxic interactions. The combination of the common cytotoxic endpoint growth with a behavioural characteristic comprises a complementary set of sublethal endpoints for ecotoxicological screening purposes. Data from recommended standard aquatic toxicity tests cannot predict hazards for Tetrahymena. However a highly significant correlation was found between the proliferation inhibition of mammalian cell cultures (CHO) and Tetrahymena.     Indikation oekotoxischer Wirkungen von Umweltchemikalien durch Tetrahymena im Rahmen eines Ringtests

    This project was concerned with the selection of suitable test protocols for the indication of the effects of environmental chemicals on Tetrahymena. Based on its wide acceptance and high degree of standardisation, the growth test was chosen for intercalibrating exercises on 6 reference chemicals involving 5 praticipating laboratories from Denmark, The Netherlands, the USA and Germany. The aim of this internationa research effort was to work out a harmonized proposal for an OECD growth-test protocol with protozoa. Results indicate that the ciliate species Tetrahymena pyrifomis and T.thermophila are equally suited as test organisms for exotoxicological purposes. The growth assay reveals predominantly higher sensitivities for the detection of chemical pollution than the vitality or oxygen consumption tests, or even than the chemosensory response, a behavioural parameter. Both species can be tested in axenic or monoxenic culture. Effects on proliferation-rate and stationary cell-density are almost identical. The highest effect potential of the test chemicals was found in synthetic medium. Growth inhibition in axenic, particle-free cultures with synthetic or the more complex PPY-medium can be measured not only by direct cell counting, but also photometrically. Tetrahymena growth tests can be miniaturezed to microplate scale. Semi-static and flow-through tests can also be performed. The marginal deviation of the ring-test results and the comparison of EC-data with the OECD standard tests reveal the Tetrahymena growth to be a highly reproducible and sensitive parameter. The inclusion of a protozoan test protocol would be an ecologically relevent supplement to the existing test battery in aquatic toxicity testing.     Continuing the investigations concerning the ecological relevance of packaging materials the Federal Environmental Agency commissioned life cycle assessments on packing systems for non-alcoholic drinks and wine. The investigation aims at compiling information on environmentally relevant material and energy flows of those packaging systems with a significant importance in the specified beverage market segments, on the basis of representative and average framework conditions (reference year 1996) and comparison of their potential ecological impacts. The life cycle assessment was conducted in accordance with the ISO-standards 14040 to 14043. The Critical Review was performed corresponding to ISO 14040 chapter 7.3.3. The analysed packaging systems were chosen on basis of their market relevance (in general more than 5% market share). For the life cycle steps filling and distribution a comprehensive data collection has been carried out. The unit processes of the life cycle have been described in the report with some characteristic features and more detailed in the separate volume. An impact assessment and a conclusive interpretation of the results are further parts of the investigation.
