Histological evaluation of the exposure to 3,4-dichloroaniline in the estuarine mysid Mesopodopsis slabberi, under experimental conditions
This work presents an experimental approach to test Mesopodopsis slabberi as a potential indicator of pollution. The toxic effects of 3,4-dichloroaniline (DCA) on the histology of this estuarine mysid were studied. After an acclimation period of two days, the mysids were exposed to different sublethal 3,4-DCA concentrations (0.10, 0.30, 0.50, 0.90, 1.00, 1.10, 1.20, 1.30 and 1.40 mg/L), for a period of 48 h. Each concentration had seven replicates, and one control. After the exposure period, organisms were sacrificed and submitted to a standard histological procedure with some modifications. Histological effects were analyzed in several tissues and damages were found in organisms exposed to concentrations higher then 0.30 mg/L. Muscular tissue, cuticular lens and gonads were clearly affected by 3,4-DCA, presenting accumulations of this toxic substance and lesions on the structures
Guarding net effects on landings and discards in Mediterranean trammel net fishery: Case analysis of Egadi Islands Marine Protected Area (Central Mediterranean Sea, Italy)
Discards remain among the main negative impacts of fishing activities, and their reductions are strengthened by the European Common Fisheries Policy (European Regulation 1380/2013). Trammel net fisheries appear more sustainable compared with other fishing techniques, especially from an ecological viewpoint. Despite this, reports show that trammel net fisheries deliver discard quantities between 10% and 43% of the total catch biomass. To supplement existing information, this current work attempts to address the discard reduction using guarding net in the small-scale fisheries of Egadi Islands MPA (Western Sicily, Central Mediterranean Sea). To assess the reduction of unwanted catches, 48 experimental fishing trials were conducted within a 6-month period. The experimental fishing trial employed a trammel net made up of 20 panels alternated with two different net configurations. The control panels (CN) held a large outer (180 mm) and small inner (31.25 mm) meshes. The test panels (GN) with guarding net constituted a three-mesh-high (50-mm mesh size) net placed between trammel net panels and a lead line. A total of 3,310 individuals belonging to 106 taxa and nine phyla were caught. Crustaceans were the most abundant unwanted catches in the control panels, whereas bioconstructions occurred in the guarding net panels. The discard ratios of CN and GN panels were statistically different (t-value = –2.55; p< 0.05). The analysis of catch per unit effort showed higher catches of CN panels for both commercial and discard fractions (p< 0.05). Moreover, the guarding net panels caught the main discarded species at 20% lower compared with the control. The overall value of the catch at the CN panels (€ 3,366.90) was higher than the total income (€ 2,043.70) generated using the GN panels, which suggests a significant commercial loss of 40% (p< 0.05)
Moisture effect on the separation of CO2/CH4 mixtures with amine-functionalised porous silicas
The effect of minor amounts of water on the CO2 and CH4 adsorption on primary and secondary amine-functionalised mesoporous silicas (APTES@SBA-15 and DEAPTES@SBA15, respectively) was studied with a combination of high-pressure gas adsorption, solid state NMR of labelled 13CO2 and density functional theory (DFT) calculations. Known amounts of water were pre-adsorbed on the materials (0.047 to 0.157 mmol∙g−1) and the adsorption performance for CO2 and CH4 was compared to the performance of the dry samples. We observed that even when only minor amounts of water are present, the tertiary amine-functionalised material revealed a significant enhancement of the selectivity for CO2 (from ca. 5.8 to 208) while the one with primary amine maintained the same adsorption properties. This is related to the change in the adsorption mechanism in DEAPTES@SBA15 when water is present since water participates in the reaction of the tertiary amine with CO2 to produce bicarbonate (confirmed by NMR and DFT results). Such species was not observed in APTES@SBA-15 with water. From a broader perspective, the results presented in this work are relevant to confirm the suitability of this type of hybrid adsorbents in industrial applications related with CO2 adsorption, where minor amounts of water may be present in the process streams, such as during bio, landfill, or natural gas upgrading, or also in carbon capture applications.publishe
An innovative technique to improve safety of volatile anesthetics suction from the cardiopulmonary bypass circuit
Context: Myocardial injury during cardiac surgery on cardiopulmonary bypass (CPB) is a major determinant of morbidity and mortality. Preclinical and clinical evidence of dose- and time-related cardioprotective effects of volatile anesthetic drugs exist and their use during the whole surgery duration could improve perioperative cardiac protection. Even if administering volatile agents during CPB are relatively easy, technical problems, such as waste gas scavenging, may prevent safe and manageable administration of halogenated vapors during CPB. Aims: The aim of this study is to improve the safe administration of volatile anesthesia during CPB. Settings and Design: Tertiary teaching hospital. Subjects and Methods: We describe an original device that collects and disposes of any volatile anesthetic vapors present in the exit stream of the oxygenator, hence preventing its dispersal into the operating theatre environment and adaptively regulates pressure of oxygenator chamber in the CPB circuit. Results: We have so far applied a prototype of this device in more than 1300 adult cardiac surgery patients who received volatile anesthetics during the CPB phase. Conclusions: Widespread implementation of scavenging system like the one we designed may facilitate the perfusionist and the anesthesiologist in delivering these cardioprotective drugs with beneficial impact on patients' outcome without compromising on safety

How is artificial lighting affecting the catches in deep water rose shrimp trawl fishery of the Central Mediterranean Sea?
The effect of artificial lights mounted on the headrope trawl net on the catch of deep water rose shrimp (Parapenaeus longirostris), European hake (Merluccius merluccius), and Atlantic horse mackerel (Trachurus trachurus) was tested in a survey carried out on-board a commercial trawler off the SW Sicilian coast. A total of 18 repeated nocturnal hauls, alternating without (control) and with (test) LED lights (10 green and 10 white) according to the fishers’ setup, were conducted. Overall, the test net catch rates were not significantly higher than those of the control net (Kruskal-Wallis test, p > 0.05), except for P. longirostris (p < 0.05). Conversely, the two-tailed Kolmogorov–Smirnov test revealed statistical differences in the size structure of P. longirostris, M. merluccius, and T. trachurus between the test and control nets (p < 0.05). Using generalised linear mixed models, the test net was found to yield higher catches of undersized individuals of the three species and adults of P. longirostris than the control net. Our study results are discussed in the context of the exploitation and management of Mediterranean trawl fisheries
High shear stress enhances endothelial permeability in the presence of the risk haplotype at 9p21.3
Single nucleotide polymorphisms (SNPs) are exceedingly common in non-coding loci, and while they are significantly associated with a myriad of diseases, their specific impact on cellular dysfunction remains unclear. Here, we show that when exposed to external stressors, the presence of risk SNPs in the 9p21.3 coronary artery disease (CAD) risk locus increases endothelial monolayer and microvessel dysfunction. Endothelial cells (ECs) derived from induced pluripotent stem cells of patients carrying the risk haplotype (R/R WT) differentiated similarly to their non-risk and isogenic knockout (R/R KO) counterparts. Monolayers exhibited greater permeability and reactive oxygen species signaling when the risk haplotype was present. Addition of the inflammatory cytokine TNF alpha further enhanced EC monolayer permeability but independent of risk haplotype; TNF alpha also did not substantially alter haplotype transcriptomes. Conversely, when wall shear stress was applied to ECs in a microfluidic vessel, R/R WT vessels were more permeable at lower shear stresses than R/R KO vessels. Transcriptomes of sheared cells clustered more by risk haplotype than by patient or clone, resulting in significant differential regulation of EC adhesion and extracellular matrix genes vs static conditions. A subset of previously identified CAD risk genes invert expression patterns in the presence of high shear concomitant with altered cell adhesion genes, vessel permeability, and endothelial erosion in the presence of the risk haplotype, suggesting that shear stress could be a regulator of non-coding loci with a key impact on CAD
Fertility-sparing treatment for endometrial cancer and atypical endometrial hyperplasia in patients with Lynch Syndrome: Molecular diagnosis after immunohistochemistry of MMR proteins
Introduction: Lynch Syndrome (LS) represents the hereditary condition that is most frequently associated with endometrial cancer (EC). The aim of this study is to assess the presence of Lynch Syndrome (LS) in young women with mismatch repair (MMR)-deficient atypical endometrial hyperplasia (AEH) and non-myoinvasive FIGO G1 endometrioid EC and its possible impact on the outcome of conservative treatment. Methods: Six MMR-deficient cases identified from a previous cohort of 69 conservatively treated patients were selected to be screened for germline mutations in MMR genes. In each patient, the outcomes of conservative treatment for AEH and EEC, including response, relapse, progression, and pregnancy, were assessed. Results: Five out of 6 patients underwent genetic test for LS. Three out of these 5 patients showed a positive genetic test. Patient 1 showed the c.942 + 2 T>A heterozygous variant of MSH2 mutation; after 12 months of complete response, she had relapse and progression of disease. Patient 4 showed the c.2459-1G>C variant of MSH2 mutation; after complete response, she failed to achieve pregnancy; she had relapse after 24 months and underwent hysterectomy. Patient 6 showed the c.803 + 1 heterozygous variant of PMS2 mutation; she had relapse of disease after 18 months from the first complete response and then underwent hysterectomy. Conclusions: In this series, 3 out of 6 women with MMR-deficiency had LS. None of the patients achieved pregnancy, and those who responded to treatment had subsequent relapse of disease. Patients undergoing fertility-sparing treatment for atypical endometrial hyperplasia and endometrial cancer should perform MMR immunohistochemical analysis in order to screen LS
