    The Neuro-endocrinological Role of Microbial Glutamate and GABA Signaling.

    Gut microbiota provides the host with multiple functions (e.g., by contributing to food digestion, vitamin supplementation and defense against pathogenic strains) and interacts with the host organism through both direct contact (e.g., through surface antigens) and soluble molecules, which are produced by the microbial metabolism. The existence of the so-called gut-brain axis of bi-directional communication between the gastrointestinal tract and the central nervous system also supports a communication pathway between the gut microbiota and neural circuits of the host, including the central nervous system. An increasing body of evidence has shown that gut microbiota is able to modulate gut and brain functions, including the mood, cognitive functions and behavior of humans. Nonetheless, given the extreme complexity of this communication network, its comprehension is still at its early stage. The present contribution will attempt to provide a state-of-the art description of the mechanisms by which gut microbiota can affect the gut-brain axis and the multiple cellular and molecular communication circuits (i.e., neural, immune and humoral). In this context, special attention will be paid to the microbial strains that produce bioactive compounds and display ascertained or potential probiotic activity. Several neuroactive molecules (e.g., catecholamines, histamine, serotonin and trace amines,) will be considered, with special focus on Glu and GABA circuits, receptors and signaling. From the basic science viewpoint, microbial endocrinology deals with those theories in which neurochemicals, produced by both multicellular organisms and prokaryotes (e.g., serotonin, GABA, glutamate), are considered as a common shared language that enables interkingdom communication. With regards to its application, research in this area opens the way toward the possibility of the future use of neuroactive molecule-producing probiotics as therapeutic agents for the treatment of neurogastroenteric and/or psychiatric disorders

    Wearable sensors system for an improved analysis of freezing of gait in Parkinson's disease using electromyography and inertial signals

    We propose a wearable sensor system for automatic, continuous and ubiquitous analysis of Freezing of Gait (FOG), in patients affected by Parkinson's disease. FOG is an unpredictable gait disorder with different clinical manifestations, as the trembling and the shuffling-like phenotypes, whose underlying pathophysiology is not fully understood yet. Typical trembling-like subtype features are lack of postural adaptation and abrupt trunk inclination, which in general can increase the fall probability. The targets of this work are detecting the FOG episodes, distinguishing the phenotype and analyzing the muscle activity during and outside FOG, toward a deeper insight in the disorder pathophysiology and the assessment of the fall risk associated to the FOG subtype. To this aim, gyroscopes and surface electromyography integrated in wearable devices sense simultaneously movements and action potentials of antagonist leg muscles. Dedicated algorithms allow the timely detection of the FOG episode and, for the first time, the automatic distinction of the FOG phenotypes, which can enable associating a fall risk to the subtype. Thanks to the possibility of detecting muscles contractions and stretching exactly during FOG, a deeper insight into the pathophysiological underpinnings of the different phenotypes can be achieved, which is an innovative approach with respect to the state of art

    The Death of Oakes: Time for a Rights-Specific Approach?

    In this paper, the authors develop their the sis that the time has come to abandon the universal framework for the application of section 1 of the Canadian Charter of Rights and Freedoms set out in R. v. Oakes. The paper describes how the Oakes test has been eroded, diluted and, in some cases, abandoned. The authors review the history of the application of the Oakes test, as well as recent developments in Supreme Court jurisprudence. The paper concludes that the Supreme Court’s struggles with Oakes are inherent in any attempt to apply a universal approach to section 1 analysis, and conclude that a rights-specific approach to section 1 balancing should be adopted
