    Opioid Exposed Mothers and Infants in Delaware: Clinical and Legal Considerations

    Drug use is on the rise in Delaware, as demonstrated by the continued increase in infants born with neonatal abstinence syndrome. Thoughful, evidence based, and coordinated approaches are necessary to impact this problem. There is solid evidence that mothers and infants who remain together have improved outcomes. Professional medical and nursing societies are unanimous in support of non-punitive approaches to care. Medical Professionals, legislators, and society in general would benefit from ongoing education on the addiction disease process in order to best care for the increasing number of mother/baby dyads with neonatal abstinence syndrome

    The Turkey Ig-like receptor family: identification, expression and function.

    The chicken leukocyte receptor complex located on microchromosome 31 encodes the chicken Ig-like receptors (CHIR), a vastly expanded gene family which can be further divided into three subgroups: activating CHIR-A, bifunctional CHIR-AB and inhibitory CHIR-B. Here, we investigated the presence of CHIR homologues in other bird species. The available genome databases of turkey, duck and zebra finch were screened with different strategies including BLAST searches employing various CHIR sequences, and keyword searches. We could not identify CHIR homologues in the distantly related zebra finch and duck, however, several partial and complete sequences of CHIR homologues were identified on chromosome 3 of the turkey genome. They were designated as turkey Ig-like receptors (TILR). Using cDNA derived from turkey blood and spleen RNA, six full length TILR could be amplified and further divided according to the typical sequence features into one activating TILR-A, one inhibitory TILR-B and four bifunctional TILR-AB. Since the TILR-AB sequences all displayed the critical residues shown to be involved in binding to IgY, we next confirmed the IgY binding using a soluble TILR-AB1-huIg fusion protein. This fusion protein reacted with IgY derived from various gallinaceous birds, but not with IgY from other bird species. Finally, we tested various mab directed against CHIR for their crossreactivity with either turkey or duck leukocytes. Whereas no staining was detectable with duck cells, the CHIR-AB1 specific mab 8D12 and the CHIR-A2 specific mab 13E2 both reacted with a leukocyte subpopulation that was further identified as thrombocytes by double immunofluorescence employing B-cell, T-cell and thrombocyte specific reagents. In summary, although the turkey harbors similar LRC genes as the chicken, their distribution seems to be distinct with predominance on thrombocytes rather than lymphocytes

    Response of chickens to oral vaccination with Newcastle disease virus vaccine strain I2 coated on maize offal

    Thermostable Newcastle disease (ND) vaccine virus strain I2 was investigated for its efficacy as foodborne vaccine, using maize offal as the vehicle. Immune response to vaccination and resistance tochallenge were assessed by standard methods. Results showed that following primary vaccination, 40 (64.5%) out of the 62 birds produced detectable haemagglutination inhibiting (HI) antibody, but only 4(6.5%) produced HI (log2) antibody titre 3.0 regarded as protective with a geometric mean titre (GMT) of 3.1. After a booster dose, 49 (79.0%) seroconverted and 20 (32.3%) had HI (log2) titres 3.0 with GMT of 4.9. When challenged all vaccinated birds survived while all control (unvaccinated) birds died. Prechallenge HI antibody titre of 50 vaccinated birds selected for challenge showed that 13 (26.0%) had titres 3.0 and GMT = 4.5, while post-challenge, 31 (62.0%) had HI (log2) 3.0 with GMT of 7.2. Using Student t test analysis of significance, the birds were observed to show 70% HI antibody production at a P 0.3 and 3 degree of freedom (df), and 70% secondary immune response on challenge at 4df. It is therefore concluded that the vaccine could be effective for protection of village chickens as food-borne vaccine provided the carrier foods are adequately treated

    High speed boat traffic: A risk to crocodilian populations

    Injuries related to boat traffic have been documented as a major source of human-related injuries and deaths in many aquatic species but have not been documented in crocodilians. We examined the proportion of boat-related injuries in Spectacled Caiman (Caiman crocodilus fuscus) in the Tortuguero region of Costa Rica within waterways with enforced speed limits and those without. No boat injuries were documented in waterways with speed limits; however, in waterways without speed limits, 36.6% of caimans we captured retained old scars or fresh lacerations related to boat propellers, including two mortalities. Presence of enforced speed zones was positively associated with lower rates of collisions, boat propeller related injuries, and mortalities. Slower boat speed may provide sufficient time for caimans to avoid oncoming boats. Increasing boat traffic in the region due to increasing ecotourism and a rapidly expanding local community may increase the vulnerability of this crocodilian species to boat related injuries and population eduction. Imposing enforced speed limits within non-essential transportation routes is a reasonable mitigation measure to help protect this species

    Chicken TREM-B1, an Inhibitory Ig-Like Receptor Expressed on Chicken Thrombocytes

    Triggering receptors expressed on myeloid cells (TREM) form a multigene family of immunoregulatory Ig-like receptors and play important roles in the regulation of innate and adaptive immunity. In chickens, three members of the TREM family have been identified on chromosome 26. One of them is TREM-B1 which possesses two V-set Ig-domains, an uncharged transmembrane region and a long cytoplasmic tail with one ITSM and two ITIMs indicating an inhibitory function. We generated specific monoclonal antibodies by immunizing a Balb/c mouse with a TREM-B1-FLAG transfected BWZ.36 cell line and tested the hybridoma supernatants on TREM-B1-FLAG transfected 2D8 cells. We obtained two different antibodies specific for TREM-B1, mab 7E8 (mouse IgG1) and mab 1E9 (mouse IgG2a) which were used for cell surface staining. Single and double staining of different tissues, including whole blood preparations, revealed expression on thrombocytes. Next we investigated the biochemical properties of TREM-B1 by using the specific mab 1E9 for immunoprecipitation of either lysates of surface biotinylated peripheral blood cells or stably transfected 2D8 cells. Staining with streptavidin coupled horse radish peroxidase revealed a glycosylated monomeric protein of about 50 kDa. Furthermore we used the stably transfected 2D8 cell line for analyzing the cytoplasmic tyrosine based signaling motifs. After pervanadate treatment, we detected phosphorylation of the tyrosine residues and subsequent recruitment of the tyrosine specific protein phosphatase SHP-2, indicating an inhibitory potential for TREM-B1. We also showed the inhibitory effect of TREM-B1 in chicken thrombocytes using a CD107 degranulation assay. Crosslinking of TREM-B1 on activated primary thrombocytes resulted in decreased CD107 surface expression of about 50-70%

    Hepatitis C cross-genotype immunity and implications for vaccine development.

    While about a quarter of individuals clear their primary hepatitis C (HCV) infections spontaneously, clearance (spontaneous or treatment-induced) does not confer sterilizing immunity against a future infection. Since successful treatment does not prevent future infections either, an effective vaccine is highly desirable in preventing HCV (re)infection. However, development of an effective vaccine has been complicated by the diversity of HCV genotypes, and complexities in HCV immunological responses. Smaller studies on humans and chimpanzees reported seemingly opposing results regarding cross-neutralizing antibodies. We report a lack of cross-genotype immunity in the largest cohort of people to date. In the adjusted Cox proportional hazards model, reinfection with a heterologous HCV genotype (adjusted Hazard Ratio [aHR]: 0.45, 95% CI: 0.25-0.84) was associated with a 55% lower likelihood of re-clearance. Among those who cleared their first infection spontaneously, the likelihood of re-clearance was 49% lower (aHR: 0.51, 95% CI: 0.27-0.94) when reinfected with a heterologous HCV genotype. These findings indicate that immunity against a particular HCV genotype does not offer expanded immunity to protect against subsequent infections with a different HCV genotype. A prophylactic HCV vaccine boosted with multiple HCV genotype may offer a broader and more effective protection

    Delivery of thermostable Newcastle disease (ND) vaccine to chickens with broken millet grains as the vehicle

    The efficacy of treated broken millet grains as a carrier for delivery of thermostable Newcastle disease (ND) vaccine HRV4 to free-range chickens in three locations was assessed by haemagglutinationinhibition (HI) test and challenge experiment. Out of 256 birds fed with first dose of the vaccine, 130 (50.8%) produced detectable HI antibody but only 16 (6.3%) attained serum antibody level of log2 3.0adjudged protective. From the locations, Igumale (74 birds), Kuru (88 birds) Riyom (94 birds), only 1 (1.4%), 8 (9.1%) and 7 (7.4%) attained log2 titre 3.0, respectively. A booster vaccine dose 2 weeks lateron 236 of the birds led to 126 (53.4%) attaining HI (log2) titre 3.0 [Igumale (63 birds), Kuru (86 birds) Riyom (87 birds) had 33 (53.5%), 46 (53.5%) and 42 (48.3%) respectively attaining log2 titres of 3.0]. Out of a total of 70 buy-back chickens challenged, 49 (70.0%) survived. The break down showed that from Igumale (20 birds), Kuru (25 birds) Riyom (25 birds), the survivors were 13 (65.0%), 19 (76.0%) and 17 (68.0%), respectively. Out of 28 unvaccinated control birds challenged, only 4 survived. The overall results showed that millet, if properly treated, could be a good vaccine carrier and that the method of vaccination was relatively efficacious.
