    Early Interfaced Neural Activity from Chronic Amputated Nerves

    Direct interfacing of transected peripheral nerves with advanced robotic prosthetic devices has been proposed as a strategy for achieving natural motor control and sensory perception of such bionic substitutes, thus fully functionally replacing missing limbs in amputees. Multi-electrode arrays placed in the brain and peripheral nerves have been used successfully to convey neural control of prosthetic devices to the user. However, reactive gliosis, micro hemorrhages, axonopathy and excessive inflammation currently limit their long-term use. Here we demonstrate that enticement of peripheral nerve regeneration through a non-obstructive multi-electrode array, after either acute or chronic nerve amputation, offers a viable alternative to obtain early neural recordings and to enhance long-term interfacing of nerve activity. Non-restrictive electrode arrays placed in the path of regenerating nerve fibers allowed the recording of action potentials as early as 8 days post-implantation with high signal-to-noise ratio, as long as 3 months in some animals, and with minimal inflammation at the nerve tissue-metal electrode interface. Our findings suggest that regenerative multi-electrode arrays of open design allow early and stable interfacing of neural activity from amputated peripheral nerves and might contribute towards conveying full neural control and sensory feedback to users of robotic prosthetic devices

    Experiencia de evaluación integradora grupal en Meteorología Agrícola

    El aumento del número de alumnos en la carrera de Ingeniería Agronómica de la Facultad de Ciencias Agrarias, UNCuyo, al no estar acompañado por un incremento en la cantidad de docentes, puso en crisis el método de mensuración de conocimientos de los alumnos que era practicado con excelentes resultados a través de la clásica evaluación oral individual. Por ello hubo que reemplazar este método por otro, que manteniendo la calidad de la mensuración, requiriera menos tiempo y docentes por alumno evaluado. A diferencia de otras asignaturas que sustituyeron el sistema tradicional por una evaluación escrita, la asignatura Meteorología Agrícola optó por un sistema mixto constituido por cinco evaluaciones individuales escritas sobre objetivos parciales durante el curso, más una evaluación final integradora. Debido al carácter integrador de esta evaluación final, se decidió realizarla oralmente y para hacerla posible con el tiempo y número de docentes disponibles, se evalúan simultáneamente los conocimientos de cinco alumnos, siendo la calificación individual para cada alumno. Para analizar los resultados de ambos métodos, se realizó: a. la comparación de las frecuencias de notas de evaluaciones orales individuales (antiguo método) y de evaluaciones orales grupales (nuevo método), con un coeficiente de correlación de 0.64 b. la comparación de la nota obtenida por cada alumno con su nota promedio en la carrera, con un coeficiente de correlación de 0.50 c. encuestas a docentes y alumnos. Los resultados obtenidos permiten afirmar que la evaluación oral grupal constituye una forma idónea para evaluar el grado de integración de conocimientos en Meteorología AgrícolaThe increasing number of students in Agronomic Engineer at the Facultad de Ciencias Agrarias UNCuyo was not accompanied by an increase in the teachers´ number. This led to the collapse of traditional student's knowledge assessment method, a classic individual oral examination. It was necessary to adopt a new method requiring less time and teachers per evaluated student. A system with five individual written achievement tests, plus a final integrative evaluation was adopted in Agricultural Meteorology. Its integrative character led to an oral form evaluation and, to make it viable with the available time and teachers, five students are evaluated simultaneously getting individual mark. To analyze the performance of the two methods it was done: a. the comparison of the frequencies of the scores obtained by the two methods, (correlation coefficient: 0.64) b. the comparison of the mark of each single student with his/her score in the career, (correlation coefficient: 0.50) c. surveys to teachers and students.     Modality-Specific Axonal Regeneration: Toward Selective Regenerative Neural Interfaces

    Regenerative peripheral nerve interfaces have been proposed as viable alternatives for the natural control of robotic prosthetic devices. However, sensory and motor axons at the neural interface are of mixed sub-modality types, which difficult the specific recording from motor axons and the eliciting of precise sensory modalities through selective stimulation. Here we evaluated the possibility of using type specific neurotrophins to preferentially entice the regeneration of defined axonal populations from transected peripheral nerves into separate compartments. Segregation of mixed sensory fibers from dorsal root ganglion neurons was evaluated in vitro by compartmentalized diffusion delivery of nerve growth factor (NGF) and neurotrophin-3 (NT-3), to preferentially entice the growth of TrkA+ nociceptive and TrkC+ proprioceptive subsets of sensory neurons, respectively. The average axon length in the NGF channel increased 2.5-fold compared to that in saline or NT-3, whereas the number of branches increased threefold in the NT-3 channels. These results were confirmed using a 3D “Y”-shaped in vitro assay showing that the arm containing NGF was able to entice a fivefold increase in axonal length of unbranched fibers. To address if such segregation can be enticed in vivo, a “Y”-shaped tubing was used to allow regeneration of the transected adult rat sciatic nerve into separate compartments filled with either NFG or NT-3. A significant increase in the number of CGRP+ pain fibers were attracted toward the sural nerve, while N-52+ large-diameter axons were observed in the tibial and NT-3 compartments. This study demonstrates the guided enrichment of sensory axons in specific regenerative chambers, and supports the notion that neurotrophic factors can be used to segregate sensory and perhaps motor axons in separate peripheral interfaces

    Natural ester fluids applications in transformers as a sustainable dielectric and coolant

    New requirements for a better sustainable energy policy around the world is easy to observe, many projects in sustainable energy are developed wherein the academia works together the authorities and commercial firms such as electrical grid utilities. In distribution transformers one of the suggested solutions is to replace mineral oils used as dielectric and coolant for natural ester fluids, they provide improved technical properties making them a safe substitute of mineral oil. Natural esters are based on clean technology in order to guarantee requirements from utilities. The main goal of the present paper is to present a technical-economic analysis obtained from five years of applications of oil-immersed transformers using natural ester fluids in Colombia. The methodology considers forty-four oil-immersed distribution Transformers, a half use mineral oil and the other side use natural ester, operating under the same load regime. Results evidenced the feasibility of implementing this technology because the costs are very similar and the reduction of risks and environmental impact is considerable. © 2019 Author(s)

    Effect of rabbit doe-litter separation on 24-hour changes of luteinizing hormone, follicle stimulating hormone and prolactin release in female and male suckling pups

    BACKGROUND: The daily pattern of nursing of the rabbit pup by the doe is the most important event in the day for the newborn and is neatly anticipated by them. Such anticipation presumably needs a close correlation with changes in hormones that will allow the pups to develop an appropriate behavior. Although a number of circadian functions have been examined in newborn rabbits, there is no information on 24-h pattern of gonadotropin release or on possible sex-related differences in gonadotropin or prolactin (PRL) release of pups. This study examined the 24-h changes of plasma luteinizing hormone (LH), follicle stimulating hormone (FSH) and prolactin (PRL) in 11 days old suckling female and male rabbits left with the mother or after short-term (i.e., 48 h) doe-litter separation. METHODS: Animals were kept under controlled light-dark cycles (16 h – 8 h; lights on at 08:00 h). On day 9 post partum, groups of 6–7 female or male rabbit pups were separated from their mothers starting at 6 different time intervals in the 24 h cycle. Pups were killed 48 h after separation. At each time interval groups of male or female pups that stayed with the mother were killed as controls. Plasma, LH, FSH and PRL levels were measured by specific radioimmunoassays. RESULTS: In pups kept with their mother plasma FSH and LH maxima occurred at the first and second part of the light phase (at 13:00 and 17:00 – 21:00 h, respectively) (females) or as two peaks for each of the hormones (at 13:00 and 01:00 h) (males). PRL release was similar in female and male rabbit pups kept with their mother, showing a 24-h pattern with two peaks, at 13:00 and 01:00 h, respectively. Mean 24-h values of gonadotropins and PRL did not differ between sexes. Isolation of pups for 48 h augmented circulating gonadotropin and PRL levels and distorted hormone 24-h pattern to a similar extent in both sexes. CONCLUSION: Significant sex differences in 24-h changes in LH and FSH, but not in PRL, release occurred in rabbit pups kept with the doe. Separation of newborn pups from their mother augmented circulating gonadotropin and PRL levels and disrupted 24-h rhythmicity of gonadotropin and PRL release similarly in both sexes. The effect of pups' isolation can be attributed either to a modification of the circadian pacemaker or to a masking effect on some of its output overt rhythms

    Tensor Networks for Solving Realistic Time-independent Boltzmann Neutron Transport Equation

    Tensor network techniques, known for their low-rank approximation ability that breaks the curse of dimensionality, are emerging as a foundation of new mathematical methods for ultra-fast numerical solutions of high-dimensional Partial Differential Equations (PDEs). Here, we present a mixed Tensor Train (TT)/Quantized Tensor Train (QTT) approach for the numerical solution of time-independent Boltzmann Neutron Transport equations (BNTEs) in Cartesian geometry. Discretizing a realistic three-dimensional (3D) BNTE by (i) diamond differencing, (ii) multigroup-in-energy, and (iii) discrete ordinate collocation leads to huge generalized eigenvalue problems that generally require a matrix-free approach and large computer clusters. Starting from this discretization, we construct a TT representation of the PDE fields and discrete operators, followed by a QTT representation of the TT cores and solving the tensorized generalized eigenvalue problem in a fixed-point scheme with tensor network optimization techniques. We validate our approach by applying it to two realistic examples of 3D neutron transport problems, currently solved by the PARallel TIme-dependent SN (PARTISN) solver. We demonstrate that our TT/QTT method, executed on a standard desktop computer, leads to a yottabyte compression of the memory storage, and more than 7500 times speedup with a discrepancy of less than 1e-5 when compared to the PARTISN solution.Comment: 38 pages, 9 figure

    Neuroimagen estructural y funcional en las enfermedades priónicas humanas

    INTRODUCTION: Prion diseases are neurodegenerative disorders resulting from the accumulation of a misfolded isoform of the cellular prion protein (PrPc). They can occur as acquired, sporadic or hereditary forms. Although prion diseases show a wide range of phenotypic variations, pathological features and clinical evolution, they are all characterised by a common unfavourable course and a fatal outcome. REVIEW SUMMARY: Some variants, such as kuru, have practically disappeared, while others, for example the variant Creutzfeldt-Jakob (vCJD) or those attributable to iatrogenic causes, are still in force and pose a challenge to current medicine. There are no definitive pre-mortem diagnostic tests, except for vCJD, where a tonsil biopsy detects 100% of the cases. For this reason, diagnostic criteria dependent on statistical probability have had to be created. These require complementary examinations, such as an electroencephalogram (EEG) or the detection of 14-3-3 protein in cerebrospinal fluid (CSF). Only the "pulvinar sign" in magnetic resonance imaging (MRI) has been included as a vCJD diagnostic criterion. The present review discusses neuroimaging findings for each type of prion disease in patients with a definitive histopathological diagnosis. CONCLUSIONS: The aim is to define the usefulness of these complementary examinations as a tool for the diagnosis of this family of neurodegenerative diseases

    Clinical Implications for Carotid Body Neuromodulation

This study received funding from the Fundação para a Ciência e Tecnologia (FCT) (UIDB/05704/2020).Chronic carotid sinus nerve (CSN) electrical modulation through kilohertz frequency alternating current improves metabolic control in rat models of type 2 diabetes, underpinning the potential of bioelectronic modulation of the CSN as a therapeutic modality for metabolic diseases in humans. The CSN carries sensory information from the carotid bodies, peripheral chemoreceptor organs that respond to changes in blood biochemical modifications such as hypoxia, hypercapnia, acidosis, and hyperinsulinemia. In addition, the CSN also delivers information from carotid sinus baroreceptors—mechanoreceptor sensory neurons directly involved in the control of blood pressure—to the central nervous system. The interaction between these powerful reflex systems—chemoreflex and baroreflex—whose sensory receptors are in anatomical proximity, may be regarded as a drawback to the development of selective bioelectronic tools to modulate the CSN. Herein we aimed to disclose CSN influence on cardiovascular regulation, particularly under hypoxic conditions, and we tested the hypothesis that neuromodulation of the CSN, either by electrical stimuli or surgical means, does not significantly impact blood pressure. Experiments were performed in Wistar rats aged 10–12 weeks. No significant effects of acute hypoxia were observed in systolic or diastolic blood pressure or heart rate although there was a significant activation of the cardiac sympathetic nervous system. We conclude that chemoreceptor activation by hypoxia leads to an expected increase in sympathetic activity accompanied by compensatory regional mechanisms that assure blood flow to regional beds and maintenance of hemodynamic homeostasis. Upon surgical denervation or electrical block of the CSN, the increase in cardiac sympathetic nervous system activity in response to hypoxia was lost, and there were no significant changes in blood pressure in comparison to control animals. We conclude that the responses to hypoxia and vasomotor control short-term regulation of blood pressure are dissociated in terms of hypoxic response but integrated to generate an effector response to a given change in arterial pressure.publishersversionpublishe
