    Rapid and selective manipulation of milk fatty acid composition in mice through the maternal diet during lactation

    Dietary fatty acid (FA) composition in early postnatal life can modulate growth and development and later metabolic health. Investigating programming effects of early dietary FA manipulations in rodents may be stressful and complicated due to the need of artificial feeding techniques. It is largely unknown to what extent breast milk (BM) FA composition can be directly manipulated by the diet. We exposed dams to different dietary FA compositions from postnatal day (PN) 2 until PN28. Dams with litters were randomly assigned to control (CTRL), high-medium-chain FA (MCFA), low-linoleic acid (LowLA), high-n-3 long-chain PUFA (n-3LCP) or high-n-3LCP and MCFA (n-3LCP/MCFA) diets, and diets were continued after weaning until PN28. FA compositions were determined in feeds, milk and in erythrocytes. BM MCFA content was independent from dietary MCFA intake. In contrast, the LowLA diet reduced BM LA content by about 50 % compared with the CTRL diet at PN7. BM of dams fed the n-3LCP or n-3LCP/MCFA diet contained about 6-fold more n-3 LCP than BM of the dams fed the CTRL diet at PN7. These changes in milk FA composition established after 5 d of dietary exposure did not further change over the lactation period. At PN28, the erythrocyte FA composition of the male pups correlated with analysed milk FA profiles. In conclusion, manipulation of the diet of lactating mice can strongly and rapidly affect BM FA composition, in particular of n-6 LA and n-3 LCP. Our present findings will facilitate mechanistic studies on the programming of adult metabolic health by dietary FA in the early postnatal period via direct and selective manipulation of the maternal diet

    Evaluating the effects of storage conditions on dry matter loss and nutritional quality of grain legume fodders in West Africa

    Feed scarcity is a major challenge for livestock production in West Africa, especially during the dry season when grass quality and quantity on grazing lands are inadequate. In the dry season, crop residues are a key source of livestock feed. The residues of grain legumes, also known as grain legume fodders (GLFs), are stored and traded for feeding in the dry season. The objectives of our experiment were to evaluate the effects of storage conditions and duration on dry matter (DM) and nutritional quality of GLFs, and to assess the risk of aflatoxin in stored groundnut fodder. The experiment was designed as a factorial trial with 18 treatment combinations with four replicates (4 farms). The treatments included: 3 types of GLFs (cowpea, groundnut and soybean fodder), 3 types of storage locations (rooftop, room and tree-fork) and 2 types of packaging (packed in polythene sacks and unpacked but tied with rope). Over a 120 day storage period, DM quantity reduced by an average of 24 % across all storage conditions, showing a range from 14 % in the best condition (sacks and rooms) to 35 % in the worst condition (bundles tied with rope and stored on rooftops or tree-forks). Soybean fodder had no leaves, the lowest crude protein content (CP) and organic matter digestibility (OMD), and the highest content of cell wall components compared to cowpea and groundnut fodder. These nutritional quality parameters in soybean fodder hardly changed during storage. Cowpea and groundnut fodder showed a decrease in leaf-to-stem ratio (LSR), CP and OMD, and an increase in the content of cell wall components during storage, but their nutritional value remained better than that of soybean fodder. Storage in sacks resulted in less DM loss, in less reduction of LSR and in a smaller increase of the content of cell wall components than storage of bundles tied with rope. Our study shows that the DM loss, the decrease in LSR, and the increase in the content of cell wall components can be prevented partly by storing GLFs in sacks instead of tying bundles with rope, and to a minor extent by storing in rooms instead of in the open air. Aflatoxin was not detectable in the groundnut fodder samples. Our results highlight that attention to storage conditions can improve the feeding value of GLFs which are key for livestock nutrition during the dry season.</p

    Gene-expression of metastasized versus non-metastasized primary head and neck squamous cell carcinomas: A pathway-based analysis

    Background: Regional lymph node metastasis is an important prognostic factor in head and neck squamous cell carcinoma (HNSCC) and plays a decisive role in the choice of treatment. Here, we present an independent gene expression validation study of metastasized versus non-metastasized HNSCC. Methods: We used a dataset recently published by Roepman et al. as reference dataset and an independent gene expression dataset of 11 metastasized and 11 non-metastasized HNSCC tumors as validation dataset. Reference and validation studies were performed on different microarray platforms with different probe sets and probe content. In addition to a supervised gene-based analysis, a supervised pathway-based analysis was performed, evaluating differences in gene expression for predefined tumorigenesis- and metastasis related gene sets. Results: The gene-based analysis showed 26 significant differentially expressed genes in the reference dataset, 21 of which were present on the microarray platform used in the validation study. 7 of these genes appeared to be significantly expressed in the validation dataset, but failed to pass the correction for multiple testing. The pathway-based analysis revealed 23 significant differentially expressed gene sets, 7 of which were statistically validated. These gene sets are involved in extracellular matrix remodeling (MMPs, MMP regulating pathways and the uPA system), hypoxia and angiogenesis (HIF1α regulated angiogenic factors and HIF1α regulated invasion). Conclusion: Pathways that are differentially expressed between metastasized and non-metastasized HNSCC are involved in the processes of extracellular matrix remodeling, hypoxia and angiogenesis. A supervised pathway-based analysis enhances the understanding of the biological context of the results, the comparability of results across different microarray studies, and reduces multiple testing problems by focusing on a limited number of pathways of interest instead of analyzing the large number of probes available on the microarray

[email protected] Abstract BACKGROUND: Video game seizures have been reported in photosensitive and non-photosensitive patients with epilepsy. The game Super Mario World, has led to many cases of first seizures. We examined whether this game was indeed more provocative than other programs and whether playing the game added to this effect. METHODS: We prospectively investigated 352 patients in four European cities, using a standard protocol including testing of a variety of visual stimuli. We correlated historical data on provocative factors in daily life with electroencephalographic laboratory findings. RESULTS: The video game, Super Mario World proved more epileptogenic than standard TV programs and as provocative as programs with flashing lights and patterns. Most striking was the fact that video game-viewing and-playing on the 50 and 100 Hz TV was significantly more provocative than viewing the standard program (P < 0.001, P < 0.05 respectively). Playing the video game Mario World on a 50 Hz TV, appeared to be significantly more provocative than playing this game on the 100 Hz TV (P < 0.001). Of 163 patients with a history of TV-, VG- or CG-seizures, 85% of them showed epileptiform discharges in response to photic stimulation, 44% to patterns, 59% to 50 Hz TV and 29% to 100 Hz TV. CONCLUSIONS: Children and adolescents with a history of video game seizures are, in the vast majority, photosensitive and should be investigated with standardised photic stimulation. Games and programs with bright background or flashing images are specifically provocative. Playing a video game on a 100 Hz TV is less provocative [published with videosequences].
