    ABL1 regulates spindle orientation in adherent cells and mammalian skin

    Despite the growing evidence for the regulated spindle orientation in mammals, a systematic approach for identifying the responsible genes in mammalian cells has not been established. Here we perform a kinase-targeting RNAi screen in HeLa cells and identify ABL1 as a novel regulator of spindle orientation. Knockdown of ABL1 causes the cortical accumulation of Leu-Gly-Asn repeat-enriched-protein (LGN), an evolutionarily conserved regulator of spindle orientation. This results in the LGN-dependent spindle rotation and spindle misorientation. In vivo inactivation of ABL1 by a pharmacological inhibitor or by ablation of the abl1 gene causes spindle misorientation and LGN mislocalization in mouse epidermis. Furthermore, ABL1 directly phosphorylates NuMA, a binding partner of LGN, on tyrosine 1774. This phosphorylation maintains the cortical localization of NuMA during metaphase, and ensures the LGN/NuMA-dependent spindle orientation control. This study provides a novel approach to identify genes regulating spindle orientation in mammals and uncovers new signalling pathways for this mechanism

    Role of Dlg5/lp-dlg, a Membrane-Associated Guanylate Kinase Family Protein, in Epithelial-Mesenchymal Transition in LLc-PK1 Renal Epithelial Cells

    Discs large homolog 5 (Dlg5) is a member of the membrane-associated guanylate kinase adaptor family of proteins, some of which are involved in the regulation of epithelial-to-mesenchymal transition (EMT). Dlg5 has been described as a susceptibility gene for Crohn's disease; however, the physiological function of Dlg5 is unknown. We show here that transforming growth factor-β (TGF-β)-induced EMT suppresses Dlg5 expression in LLc-PK1 cells. Depletion of Dlg5 expression by knockdown promoted the expression of the mesenchymal marker proteins, fibronectin and α-smooth muscle actin, and suppressed the expression of E-cadherin. In addition, activation of JNK and p38, which are stimulated by TGF-β, was enhanced by Dlg5 depletion. Furthermore, inhibition of the TGF-β receptor suppressed the effects of Dlg5 depletion. These observations suggest that Dlg5 is involved in the regulation of TGF-βreceptor-dependent signals and EMT

    Lifestyle factors affecting gastroesophageal reflux disease symptoms: a cross-sectional study of healthy 19864 adults using FSSG scores

    <p>Abstract</p> <p>Background</p> <p>Gastroesophageal reflux disease (GERD) is a very common disorder worldwide, comprised of reflux esophagitis (RE) and non-erosive reflux disease (NERD). As more than half of GERD patients are classified into the NERD group, precise evaluation of bothersome epigastric symptoms is essential. Nevertheless, compared with many reports targeting endoscopic reflux esophagitis, large-scale studies focusing on GERD symptoms have been very scarce.</p> <p>Methods</p> <p>To elucidate lifestyle factors affecting GERD symptoms, 19,864 healthy adults in Japan were analyzed. Sub-analyses of 371 proton pump inhibitor (PPI) users and 539 histamine H<sub>2</sub>-receptor antagonist (H<sub>2</sub>RA) users were also performed. Using the FSSG (Frequency Scale for the Symptoms of GERD) score as a response variable, 25 lifestyle-related factors were univariately evaluated by Student's <it>t</it>-test or Pearson's correlation coefficient, and were further analyzed with multiple linear regression modelling.</p> <p>Results</p> <p>Average FSSG scores were 4.8 ± 5.2 for total subjects, 9.0 ± 7.3 for PPI users, and 8.2 ± 6.6 for H<sub>2</sub>RA users. Among the total population, positively correlated factors and standardized coefficients (β) for FSSG scores are inadequate sleep (β = 0.158), digestive drug users (β = 0.0972 for PPI, β = 0.0903 for H<sub>2</sub>RA, and β = 0.104 for others), increased body weight in adulthood (β = 0.081), dinner just before bedtime (β = 0.061), the habit of midnight snack (β = 0.055), lower body mass index (β = 0.054), NSAID users (β = 0.051), female gender (β = 0.048), lack of breakfast (β = 0.045), lack of physical exercise (β = 0.035), younger age (β = 0.033), antihyperglycemic agents non-users (β = 0.026), the habit of quick eating (β = 0.025), alcohol drinking (β = 0.025), history of gastrectomy (β = 0.024), history of cardiovascular disease (β = 0.020), and smoking (β = 0.018). Positively correlated factors for PPI users are female gender (β = 0.198), inadequate sleep (β = 0.150), lack of breakfast (β = 0.146), antihypertensive agent non-users (β = 0.134), and dinner just before bedtime (β = 0.129), whereas those for H<sub>2</sub>RA users are inadequate sleep (β = 0.248), habit of midnight snack (β = 0.160), anticoagulants non-users (β = 0.106), and antihypertensive agents non-users (β = 0.095).</p> <p>Conclusions</p> <p>Among many lifestyle-related factors correlated with GERD symptoms, poor quality of sleep and irregular dietary habits are strong risk factors for high FSSG scores. At present, usual dose of PPI or H<sub>2</sub>RA in Japan cannot fully relieve GERD symptoms.</p

    The Caenorhabditis elegans Elongator Complex Regulates Neuronal α-tubulin Acetylation

    Although acetylated α-tubulin is known to be a marker of stable microtubules in neurons, precise factors that regulate α-tubulin acetylation are, to date, largely unknown. Therefore, a genetic screen was employed in the nematode Caenorhabditis elegans that identified the Elongator complex as a possible regulator of α-tubulin acetylation. Detailed characterization of mutant animals revealed that the acetyltransferase activity of the Elongator is indeed required for correct acetylation of microtubules and for neuronal development. Moreover, the velocity of vesicles on microtubules was affected by mutations in Elongator. Elongator mutants also displayed defects in neurotransmitter levels. Furthermore, acetylation of α-tubulin was shown to act as a novel signal for the fine-tuning of microtubules dynamics by modulating α-tubulin turnover, which in turn affected neuronal shape. Given that mutations in the acetyltransferase subunit of the Elongator (Elp3) and in a scaffold subunit (Elp1) have previously been linked to human neurodegenerative diseases, namely Amyotrophic Lateral Sclerosis and Familial Dysautonomia respectively highlights the importance of this work and offers new insights to understand their etiology

    Coupling changes in cell shape to chromosome segregation

    Animal cells undergo dramatic changes in shape, mechanics and polarity as they progress through the different stages of cell division. These changes begin at mitotic entry, with cell–substrate adhesion remodelling, assembly of a cortical actomyosin network and osmotic swelling, which together enable cells to adopt a near spherical form even when growing in a crowded tissue environment. These shape changes, which probably aid spindle assembly and positioning, are then reversed at mitotic exit to restore the interphase cell morphology. Here, we discuss the dynamics, regulation and function of these processes, and how cell shape changes and sister chromatid segregation are coupled to ensure that the daughter cells generated through division receive their fair inheritance

    DETERMINATION OF THE V4-CENTER STRUCTURE IN KBr CRYSTAL BY MEANS OF DOUBLE ALIGNMENT CHANNELING TECHNIQUE

    Nous avons déterminé la position des atomes interstitiels impliqué dans les centres V4 de cristaux de KBr par la technique de channeling par alignement double. Une mesure d'abosrption optique d'un cristal de KBr irradié par des protons ou des atomes d'hélium à la température de l'azote liquide nous a assurés que la courbe V4 est un défaut interstitiel principalement produit par le rayonnement. Un faisceau bien collimaté d'ions He+, de 1,25 MeV, a été envoyé parallèlement à la direction [110] des cristaux de KBr. La dépendance angulaire de la proction d'ions hélium renvoyés dans le plan [100] autour de la direction [[MATH]00] et de la direction [[MATH]10] ont été mesurées à la température de l'azote liquide et possèdent respectivement un pic large au centre et un pic double. La méthode de Monte Carlo a été employée pour calculer la dépendance angulaire de la production d'atomes renvoyés pour différentes positions interstitielles dans le channeling en double alignement. La comparaison entre les résultats expérimentaux et calculés indique qu'un atome de Br interstitiel du centre V4 occupe une position proche du centre du corps. Des expériences d'optique et de channeling, nous avons conclu que le centre V4 est une molécule neutre d'halogène dont chaque atome est situé dans la position décrite ci-dessus.The position of interstitial atoms involved in the V4-center in KBr crystal has been determined with the double alignment channeling technique. An optical absorption measurement of KBr crystal irradiated with protons or heliums at liquid nitrogen temperature assured that the V4-center is the predominant radiation-induced interstitial defect. A well collimated beam of 1.25 MeV He+ ions was incident parallel to the [110] direction of KBr crystals. Angular dependence of the yield of the backscattered helium ions in the (100) plane around the [[MATH]00] direction and the [[MATH]10] direction was measured at liquid nitrogen temperature and found to have a broad peak at the enter and a double peak, respectively. The Monte Carlo method was employed to calculate the angular dependence of the backscattering yield for several interstitial positions in the double alignment channeling. Comparison between the experimental and calculated results indicates that an interstitial Br atom of the V4-center occupies a position near the body enter. From the optical and channeling experiments, it was concluded that the V4-center is a neutral halogen molecule, each atom of which is located at the position described above
