Effect of site-specific mutagenesis of tyrosine-55, methionine-110 and histidine-217 in porcine kidney D-amino acid oxidase on its catalytic function
AbstractTo assess the contributions of Tyr-55, Met-110 and His-217 in porcine kidney D-amino acid oxidase (EC 1.4.3.3, DAO) to its catalytic function, we constructed three mutant cDNAs coding for the enzymes possessing Phe-55, Leu-110 and Leu-217 by site-specific mutagenesis. The mutant and wild type cDNAs could be expressed in vitro with similar efficiency. The three mutant enzymes thus synthesized showed catalytic activities comparable to that of the wild type oxidase. It is concluded that Tyr-55, Met-110 and His-217 are not directly involved in the catalytic function
Effect of peer support training for elementary school 3rd graders : Foundation for cooperative learning
本研究では,学校ぐるみで協同学習の取り組みを進めている小学校の3年生児童を対象として,協同学習の下地づくりを行うことをめざしたピア・サポートトレーニングを実施し,聞き方スキル,援助要請行動,サポート入手可能性,サポート提供可能性,攻撃性の観点からその効果を検討することを目的としていた。トレーニングの前後とフォローアップの3時点での効果測定について分析を行った結果,サポート入手可能性においてはトレーニングの効果を示唆する変化がみられた。一方,攻撃性においては,トレーニングによって攻撃性が上昇していることが示され,トレーニングに参加した児童について,イライラ感情や攻撃性への気づきを高めることはできたものの,対処方法の習得,活用,定着までには至らなかったと解釈された。トレーニングの回数,実施期間,およびトレーニング内容の般化に関して改善することで,より明確な効果が得られると考えられる
The Effects of Creative Learning Spiral Model on Making Videos to Creativity and Information Morals
クリエイティブ・ラーニング・スパイラルに基づいたワークショップが子どもの創造性・情報モラルに与える効果について検討した。ワークショップに参加する群 (介入群) とゲームで遊ぶ群(統制群) の2群間で比較した結果,アイデアの産出数と情報モラルに関する記述数の差は有意ではなかった。また,クリエイティブ・ラーニング・スパイラルの各段階の行動に対する自己効力感を調べた結果,介入群の自己効力感は有意に上昇していた。さらに,クリエイティブ・ラーニング・スパイラルに基づいた創造性を育む方法について考察し,子ども間の自由な関わり合いが必要であることを示唆した
Interactive cueing with walk-Mate for Hemiparetic Stroke Rehabilitation
Background: Many techniques that compensate for locomotion problems in daily life using externally controlled stimulation have recently been reported. These techniques are beneficial for effortlessly supporting patients' locomotive functions, but the users of such devices must necessarily remain dependent on them. It is possible that some individuals with gait impairment may be prevented recovering locomotive function. From a rehabilitation viewpoint, it may therefore be supposed that ideally, devices that can be used in daily life to improve the locomotive functions of the body itself should be proposed. Methods: We evaluate the effectiveness of Walk-Mate, which has been used mainly as a gait compensation device, as a gait rehabilitation training device by analyzing improvement in locomotion before, during and after rehabilitation in hemiparetic patients and comparing it with a previous gait training method. Walk-Mate generates a model walking rhythm in response to a user's locomotion in real time, and by indicating this rhythm using auditory stimuli, provides a technology that supports walking by reducing asymmetries and fluctuations in foot contact rhythm. If patients can use the system to learn a regulated walking rhythm, then it may also be expected to fulfil the functions of a gait rehabilitation training device for daily life. Results: With regard to asymmetry, significantly improvements were seen for compensatory movement during training using Walk-Mate, but improvements were not retained as rehabilitative results. Regarding fluctuations in the foot contact period, significant improvement was observed for compensatory movement during training and these significant improvements were retained as rehabilitative results. In addition, it became clear that such improvement could not be adequately obtained by the previously proposed training technique utilizing constant rhythmic auditory stimulation. Conclusions: Walk-Mate effectively compensated for locomotion problems of hemiparetic patients by improving gait rhythm both during and after training, suggesting that locomotive function can be effectively recovered in some patients. The interactive mechanism of Walk-Mate may be capable of simultaneously achieving the aims of gait compensation and gait rehabilitation training methods previously developed under individual frameworks. Walk-Mate is a promising technology for assisting the reintegration of disabled persons into society
Household smoking and dental caries in schoolchildren: the Ryukyus Child Health Study
<p>Abstract</p> <p>Background</p> <p>Secondhand smoke exposure (SHSe) is perhaps one of the most important toxic exposures in childhood. However, epidemiological studies on the relation between SHSe and dental caries are limited and have yielded inconsistent results. The present cross-sectional study examined the potential association between SHSe at home and the prevalence of dental caries in children.</p> <p>Methods</p> <p>Subjects were 20,703 schoolchildren aged 6 to 15 years in Okinawa, Japan. Information on SHSe at home and potential confounding factors was obtained through questionnaires. Data on dental caries were obtained from school records. Children were classified as having decayed and/or filled teeth (DFT) if a dentist diagnosed these conditions. Additionally, we analyzed decayed teeth (DT) and filled teeth (FT) separately. Adjustment was made for sex, age, region of residence, toothbrushing frequency, use of fluoride, sugar intake, and paternal and maternal educational level.</p> <p>Results</p> <p>The prevalence of DFT was 82.0%. Compared with never smoking in the household, former and current household smoking were independently associated with an increased prevalence of DFT (adjusted prevalence ratios [95% confidence intervals] for former household smoking and current light and heavy household smoking were 1.03 [1.00-1.05], 1.04 [1.02-1.05], and 1.04 [1.03-1.06], respectively); when analyzed separately there was an increased prevalence of DT (adjusted prevalence ratios [95% confidence intervals] for former household smoking and current light and heavy household smoking were 1.06 [1.02-1.11], 1.10 [1.06-1.13], and 1.10 [1.07-1.14], respectively) but not FT. A statistically significant dose-response relationship between cumulative smoking in the household and the prevalence of DFT and DT (<it>P </it>for trend < 0.0001), but not FT, was observed. In an analysis of 2 subgroups, subjects who had at least 1 deciduous tooth and subjects who had at least 1 permanent tooth, household smoking exposure was associated with an increased prevalence of DFT and DT not only in those with deciduous but also those with permanent dentition.</p> <p>Conclusion</p> <p>Our findings suggested that household smoking might be associated with an increased prevalence of dental caries in children.</p
Monetary Diet Cost is Associated with not only Favorable but also Unfavorable Aspects of Diet in Pregnant Japanese Women: The Osaka Maternal and Child Health Study
While several observational studies in European countries have shown that higher monetary diet cost is associated with healthier diets, information on the relationship of cost to diet quality in other countries is sparse, including Japan. This cross-sectional study examined the association between monetary diet cost and dietary intake in a group of pregnant Japanese women. Subjects were 596 pregnant Japanese housewives. Dietary intake was estimated using a validated, self-administered, comprehensive diet history questionnaire. Monetary diet cost was calculated using retail food prices. Values of monetary diet cost and nutrient and food intake were energy-adjusted using the density method. Monetary diet cost was associated positively with the intake of protein, total fat, saturated fatty acids, dietary fiber, cholesterol, sodium, potassium, calcium, magnesium, iron, vitamins A, D, E, C, and folate, and inversely with that of carbohydrate. For foods, cost was associated positively with the intake of potatoes, pulses and nuts, fish and shellfish, meat, dairy products, vegetables, and fruits, and inversely with that of rice and bread. No association was seen for noodles, confectioneries and sugars, fats and oils, or eggs. Cost was also associated inversely with dietary energy density. In conclusion, monetary diet cost was associated with not only favorable aspects of diet, including a higher intake of dietary fiber, key vitamins and minerals, fruits, and vegetables and lower dietary energy density, but also unfavorable aspects, including a higher intake of fat and sodium and lower intake of carbohydrate and rice, in a group of pregnant Japanese women
Panretinal Photocoagulation Using Short-Pulse Laser Induces Less Inflammation and Macular Thickening in Patients with Diabetic Retinopathy
We compared the effect of panretinal photocoagulation (PRP) using short-pulse laser (SPL) and conventional laser, regardless of the number of spots, in terms of their effect on the progression of diabetic macular edema (DME) and anterior flare intensity (AFI) in patients with high-risk nonproliferative diabetic retinopathy (non-PDR). Forty-two eyes of 42 patients were subjected to PRP using the conventional argon laser (Conv group) or SPL (SPL group). CRT and AFI levels in the SPL group were significantly lower than those in the Conv group (CRT at 4, 6, and 10 weeks; AFI at 6, 10, and 18 weeks). Eyes of rabbits were photocoagulated using conventional laser with 500 spots (Conv 500s), SPL with 500 spots (SPL 500s), or 1000 spots (SPL 1000s). Vascular endothelial growth factor (VEGF), interleukin-6 (IL-6), intercellular adhesion molecule-1 (ICAM-1), and monocyte chemotactic protein-1 (MCP-1) levels in vitreous humor were measured using an immunoassay. Compared to conventional laser, VEGF, IL-6, and MCP-1 levels were significantly lower in the SPL 1000s and SPL 500s groups. In patients with high-risk non-PDR, SPL has a greater preventive effect on the progression of DME and AFI and produces less inflammatory cytokines than conventional lasers
Unravelling the relation between altruistic cooperativeness trait, smiles, and cooperation: a mediation analysis
IntroductionHuman cooperativeness is an important personality trait. However, the mechanism through which people cooperate remains unclear. Previous research suggests that one of the proposed functions of smiling is to advertise altruistic dispositions, leading to successful cooperation. In particular, studies have reported that Duchenne smiles are honest signals of cooperative intent because they are not easy to produce voluntarily. This study aimed to examine the predictive relationships among altruistic cooperativeness traits, Duchenne smiles, and cooperative behavior.MethodsA total of 90 people were randomly assigned to dyads and filmed while they participated in a ten-minute, unstructured conversation followed by a prisoner’s dilemma game to measure their cooperative behaviors. Their smiles during conversations were classified as Duchenne or non-Duchenne. Participants’ altruistic dispositions were measured before the conversation began using an anonymous prisoner’s dilemma game.ResultsThe results of our linear regression analyses support previous findings that individual’s Duchenne smiles and their own cooperative behavior are positively correlated. However, when we controlled for altruistic cooperativeness, Duchenne smiles no longer correlated with cooperative behavior. The results of the mediation analyses showed that Duchenne smiles and smile synchrony did not mediate the predictive relationship between altruistic cooperativeness and cooperative behavior.DiscussionOur results suggest that human cooperative behavior may be predetermined by altruistic cooperativeness. This calls for the reconsideration of the Duchenne smile as an underlying behavioral mechanism that is effective for signaling altruistic cooperative intent
