    Evaluating an Institutional Response to Generative Artificial Intelligence (GenAI): Applying Kotter’s Change Model and Sharing Lessons Learned for Educational Development

    Copyright (c) 2023 Jackie Potter, Katharine Welsh, Laura MilneSince the launch of ChatGPT in November 2022, there has been a dawning understanding in the higher education sector of ways Generative artificial intelligence (GenAI) tools can challenge the traditional roles of academic teaching staff (e.g., Chan and Tsi, 2023) and support learning by students. For example, Mike Sharples in Sabzalieva and Valentini (2023) identifies ten roles that ChatGPT can play which would all support student learners. Media and sector concern has focused on whether GenAI use by students would disrupt the integrity of degrees and awards and there is a good deal of debate on how to adapt assessment, learning outcomes and curricula to reflect and reward unique human competences associated with a discipline or subject and embrace students’ use of GenAI. Educational development colleagues have been at the vanguard of leading higher education provider reactions and responses to the widespread availability and capabilities of GenAI. This case study reflects on a year of action to lead teaching staff and students as well as institutional policy and practice through a series of steps to enable rapid, proportionate and robust change. We apply Kotter’s (1996) eight stage change model to reflect on the activities, achievements and challenges to date. We do not purport to have finished but rather can see, one year in, that increasingly activity is more embedded into structures, routines, the practice of others, and our work as educational developers. We reflect forward too on the ways we will act next to ‘make change stick’ and on our own personal, professional journeys as educational change leaders, all of whom were new appointments in the educational development centre.     A young testicular microenvironment protects Leydig cells against age‐related dysfunction in a mouse model of premature aging

    Testicular Leydig cells (LCs) are the primary source of circulating androgen in men. As men age, circulating androgen levels decline. However, whether reduced LC steroidogenesis results from specific effects of aging within LCs or reflects degenerative alterations to the wider supporting microenvironment is unclear; inability to separate intrinsic LC aging from that of the testicular microenvironment in vivo has made this question difficult to address. To resolve this, we generated novel mouse models of premature aging, driven by CDGSH iron sulfur domain 2 ( Cisd2) deletion, to separate the effects of cell intrinsic aging from extrinsic effects of aging on LC function. At 6 mo of age, constitutive Cisd2-deficient mice display signs of premature aging, including testicular atrophy, reduced LC and Sertoli cell (SC) number, decreased circulating testosterone, increased luteinizing hormone/testosterone ratio, and decreased expression of steroidogenic mRNAs, appropriately modeling primary testicular dysfunction observed in aging men. However, mice with Cisd2 deletion (and thus premature aging) restricted to either LCs or SCs were protected against testicular degeneration, demonstrating that age-related LCs dysfunction cannot be explained by intrinsic aging within either the LC or SC lineages alone.     ‘A storm of post-it notes’:experiences of perceptual capacity in autism and ADHD

    Lab-based tasks suggest autistic people have increased perceptual capacity (i.e., process more information at any one time) compared to non-autistic people. Here, we explored whether this increase is reflected in autistic people’s day-to-day perceptual experiences and, when compared to those with ADHD/neurotypical people, whether commonalities/divergences in these experiences can illuminate differences between neurotypes. UK-based adults (108 autistic, 40 with ADHD, 79 autistic with ADHD, 85 neurotypical) completed an online survey about experiences of attention and distraction. Responses were analysed using thematic analysis. We found that participants of all neurotypes experienced periods of intense focus. Neurodivergent participants reported experiencing a barrage of information; autistic participants found this overwhelming, whereas those with ADHD referred to overload. This finding may reflect increased perceptual capacity for autistic people (adding ecological validity to previous findings regarding increased autistic perceptual capacity) vs. difficulties maintaining attentional priorities for those with ADHD. While differences between neurodivergent and neurotypical people were evident, discrepancies between experiences of neurodivergent groups were more subtle, suggesting that increased perceptual capacity may extend beyond autism. Consequently, perceptual capacity offers a useful framework to promote better understanding of one’s own perceptual experiences, and to guide strategies to ameliorate any challenges encountered

    Partial pressure of oxygen in adipose tissue and its relationship with fatness in a natural animal model of extreme fat deposition, the grey seal

    Excessive adiposity is associated with altered oxygen tension and comorbidities in humans. In contrast, marine mammals have high adiposity with no apparent detrimental effects. However, partial pressure of oxygen (Po2) in their subcutaneous adipose tissue (blubber) and its relationship with fatness have not been reported. We measured Po2 and temperature at different blubber depths in 12 healthy juvenile grey seals. Fatness was estimated from blubber thickness and morphometric parameters. Simultaneously, we monitored breathing pattern; heart rate and arterial blood saturation with a pulse oximeter; and relative changes in total hemoglobin, deoxyhemoglobin, and oxyhemoglobin in blubber capillaries using near-infrared spectroscopy (NIRS) as proxies for local oxygenation changes. Blubber Po2 ranged from 14.5 to 71.4 mmHg (39.2 ± 14.1 mmHg), which is similar to values reported in other species. Blubber Po2 was strongly and negatively associated with fatness (LME: p < 0.0001, R2marginal = 0.53, R2conditional = 0.64, n = 10), but not with blubber depth. No other parameters explained variability in Po2, suggesting arterial blood and local oxygen delivery did not vary within and between measurements. The fall in blubber Po2 with increased fatness in seals is consistent with other animal models of rapid fat deposition. However, the Po2 levels at which blubber becomes hypoxic and consequences of low blubber Po2 for its health and function, particularly in very fat individuals, remain unknown. How seals avoid detrimental effects of low oxygen tension in adipose tissue, despite their high and fluctuating adiposity, is a fruitful avenue to explore

    Apolipoprotein E mediates evasion from hepatitis C virus−neutralizing antibodies

    Background & Aims Efforts to develop an effective vaccine against hepatitis C virus (HCV) have been hindered by the propensity of the virus to evade host immune responses. HCV particles in serum and in cell culture associate with lipoproteins, which contribute to viral entry. Lipoprotein association has also been proposed to mediate viral evasion of the humoral immune response, though the mechanisms are poorly defined. Methods We used small interfering RNAs to reduce levels of apolipoprotein E (apoE) in cell culture−derived HCV−producing Huh7.5-derived hepatoma cells and confirmed its depletion by immunoblot analyses of purified viral particles. Before infection of naïve hepatoma cells, we exposed cell culture−derived HCV strains of different genotypes, subtypes, and variants to serum and polyclonal and monoclonal antibodies isolated from patients with chronic HCV infection. We analyzed the interaction of apoE with viral envelope glycoprotein E2 and HCV virions by immunoprecipitation. Results Through loss-of-function studies on patient-derived HCV variants of several genotypes and subtypes, we found that the HCV particle apoE allows the virus to avoid neutralization by patient-derived antibodies. Functional studies with human monoclonal antiviral antibodies showed that conformational epitopes of envelope glycoprotein E2 domains B and C were exposed after depletion of apoE. The level and conformation of virion-associated apoE affected the ability of the virus to escape neutralization by antibodies. Conclusions In cell-infection studies, we found that HCV-associated apoE helps the virus avoid neutralization by antibodies against HCV isolated from chronically infected patients. This method of immune evasion poses a challenge for the development of HCV vaccines

    Context reinstatement in recognition: memory and beyond

    Context effects in recognition tests are twofold. First, presenting familiar contexts at a test leads to an attribution of context familiarity to a recognition probe, which has been dubbed ‘context-dependent recognition’. Second, reinstating the exact study context for a particular target in a recognition test cues recollection of an item-context association, resulting in 'context-dependent discrimination'. Here we investigated how these two context effects are expressed in metacognitive monitoring (confidence judgments) and metacognitive control ('don’t know' responding) of retrieval. We used faces as studied items, landscape photographs as study and test contexts and both free- and forced-report 2AFC recognition tests. In terms of context-dependent recognition, the results document that presenting familiar contexts at test leads to higher confidence and lower rates of 'don’t know responses compared to novel contexts, while having no effect on forced-report recognition accuracy. In terms of context-dependent discrimination, the results show that reinstated contexts further boost confidence and reduce 'don’t know' responding compared to familiar contexts, while affecting forced-report recognition accuracy only when contribution of recollection to recognition performance is high. Together, our results demonstrate that metacognitive measures are sensitive to context effects, sometimes even more so than recognition measures

    Basal bud banks are the primary determinants of survival in woody legume seedlings after clipping

    Disturbances determine the structure of savannas by influencing growth at all stages of a tree's life. Since the seedling stage is such a significant part of a tree's life cycle, greater knowledge of this stage informs woody management and encroachment prevention. In this study, we tested the effect of a suite of seedling functional traits on seedling survival after experimentally-induced disturbance. Twelve mimosoid legume tree species, an important group in African savannas, with different provenances, were grown from seed under experimental conditions. Seedlings were clipped at 1 cm above ground at age 30 days, to simulate disturbance, and their survival recorded. A number of above- and below-ground traits quantifying height, biomass, bud bank and architecture were measured prior to clipping in order to assess what traits could be responsible for post-clipping survival. We also quantified root contraction by calculating the change in height of the lowest bud between the ages of 6 and 60 days. Fire regime and browser biomass of the native range of each species were calculated to test whether these factors predicted seedling survival. Seedling survival was primarily predicted by the whether the plant possessed a bud below 1 cm above ground level, i.e. below the height of clipping. For the first time we report that, in several Vachellia species, root contraction occurred, bringing basal buds closer to the ground, and in some cases below the ground, thus increasing seedling survival after clipping. No environmental variables were significant predictors of survival. Our results suggest that, at this young age, buds close to the soil surface are the most important trait predicting seedling survival, while other traits that are important for saplings and adults do not significantly influence the survival of seedlings.
