Psychometric properties of the Brazilian version of the Pittsburgh Sleep Quality Index Addendum for PTSD (PSQI-A)
Objective: Sleep disturbances play a fundamental role in the pathophysiology posttraumatic stress disorder (PTSD), and are not only a secondary feature. The aim of this study was to validate and assess the psychometric properties of the Brazilian version of the Pittsburgh Sleep Quality Index Addendum for PTSD (PSQI-A-BR), a self-report instrument designed to assess the frequency of seven disruptive nocturnal behaviors, in a sample of participants with and without PTSD. Methods: PSQI-A was translated into Brazilian Portuguese and applied to a convenience sample of 190 volunteers, with and without PTSD, who had sought treatment for the consequences of a traumatic event. Results: The PSQI-A-BR displayed satisfactory internal consistency (Cronbach's coefficient of 0.83 between all items) and convergent validity with the Clinician Administered PTSD Scale (CAPS), even when excluding sleep-related items (r = 0.52). Test-retest yielded high agreement in the global PSQI-A-BR, with good stability over time (r = 0.88). A global PSQI-A-BR cutoff score of 7 yielded a sensitivity of 79%, specificity of 64%, and a global score of 7 yielded a positive predictive value of 93% for discriminating participants with PTSD from those without PTSD. Disarming Pseudomonas aeruginosa Virulence by the Inhibitory Action of 1,10-Phenanthroline-5,6-Dione- Based Compounds: Elastase B (LasB) as a Chemotherapeutic Target
negative pathogen Pseudomonas aeruginosa, and this enzyme orchestrates several physiopathological events during bacteria-host interplays. LasB is considered to be a potential target for the development of an innovative chemotherapeutic approach, especially against multidrug-resistant strains. Recently, our group showed that 1,10-phenanthroline-5,6-dione (phendione), [Ag(phendione)2]ClO4 (Ag-phendione) and [Cu(phendione)3](ClO4)2.4H2O (Cu-phendione) had anti-P. aeruginosa action against both planktonic- and biofilm-growing cells. In the present work, we have evaluated the effects of these compounds on the (i) interaction with the lasB active site using in silico approaches, (ii) lasB proteolytic activity by using a specific fluorogenic peptide substrate, (iii) lasB gene expression by real time-polymerase chain reaction, (iv) lasB protein secretion by immunoblotting, (v) ability to block the damages induced by lasB on a monolayer of lung epithelial cells, and (vi) survivability of Galleria mellonella larvae after being challenged with purified lasB and lasB-rich bacterial secretions. Molecular docking analyses revealed that phendione and its Ag+ and Cu2+ complexes were able to interact with the amino acids forming the active site of lasB, particularly Cu-phendione which exhibited the most favorable interaction energy parameters. Additionally, the test compounds were effective inhibitors of lasB activity, blocking the in vitro cleavage of the peptide substrate, aminobenzyl-Ala-Gly-Leu-Ala-p-nitrobenzylamide, with Cu- phendione having the best inhibitory action (Ki = 90 nM). Treating living bacteria with a sub-inhibitory concentration (1/2 Ă MIC value) of the test compounds caused a significant reduction in the expression of the lasB gene as well as its mature protein production/secretion. action for lung epithelial cells, reducing the A549 monolayer damage by approximately 32 and 42%, respectively. Interestingly, Cu-phendione mitigated the toxic effect of both purified lasB molecules and lasB-containing bacterial secretions in the in vivo model, increasing the survival time of G. mellonella larvae. Collectively, these data reinforce the concept of lasB being a veritable therapeutic target and phendione-based compounds (mainly Cu-phendione) being prospective anti-virulence drugs against P. aeruginosa
Neonatal outcomes associated with tobacco, alcohol, and crack use during pregnancy in three Neonatal Intensive Care Units
Abstract Despite the prevalence of substance use during pregnancy, studies focusing exclusively on Neonatal Intensive Care Units (NICU) admissions remain limited. This study investigates the impact of maternal use of tobacco, alcohol, and/or crack, on neonatal outcomes among infants admitted to three Brazilian NICUs. Additionally, the investigation explores the impact of substance use on DNA damage in newborns. Over a one-year period, data from 254 newborns were collected through medical records, accompanied by blood samples. Findings revealed that 16.1% of newborns had mothers reporting substance use during pregnancy. Significant associations were found between maternal substance use and adverse neonatal outcomes, including low birth weight, preterm birth, and sexually transmitted infections. Maternal variables linked to substance use encompassed non-white skin color, low education, non-masonry housing, lower income, diseases in other children, and fewer prenatal consultations. Notably, neonatal DNA damage showed no significant association with substance use. Our results underscore the substantial impact of maternal substance use on NICU-admitted infants, emphasizing the necessity for targeted interventions that address both neonatal health and maternal well-being, thereby underscoring the crucial role of comprehensive care in NICU settings
TRY plant trait database â enhanced coverage and open access
Plant traits - the morphological, anatomical, physiological, biochemical and phenological characteristics of plants - determine how plants respond to environmental factors, affect other trophic levels, and influence ecosystem properties and their benefits and detriments to people. Plant trait data thus represent the basis for a vast area of research spanning from evolutionary biology, community and functional ecology, to biodiversity conservation, ecosystem and landscape management, restoration, biogeography and earth system modelling. Since its foundation in 2007, the TRY database of plant traits has grown continuously. It now provides unprecedented data coverage under an open access data policy and is the main plant trait database used by the research community worldwide. Increasingly, the TRY database also supports new frontiers of traitâbased plant research, including the identification of data gaps and the subsequent mobilization or measurement of new data. To support this development, in this article we evaluate the extent of the trait data compiled in TRY and analyse emerging patterns of data coverage and representativeness. Best species coverage is achieved for categorical traits - almost complete coverage for âplant growth formâ. However, most traits relevant for ecology and vegetation modelling are characterized by continuous intraspecific variation and traitâenvironmental relationships. These traits have to be measured on individual plants in their respective environment. Despite unprecedented data coverage, we observe a humbling lack of completeness and representativeness of these continuous traits in many aspects. We, therefore, conclude that reducing data gaps and biases in the TRY database remains a key challenge and requires a coordinated approach to data mobilization and trait measurements. This can only be achieved in collaboration with other initiatives

Effects of Circuit Weight-Interval Training on Physical Fitness, Cardiac Autonomic Control, and Quality of Life in Sedentary Workers
Sedentary behaviors, those that involve sitting and low levels of energy expenditure, have been associated with several adverse cardiometabolic effects. This study evaluated the chronic effects of a combined circuit weight interval training (CWIT) on physical fitness, quality of life, and heart rate variability (HRV), and compared the effects of CWIT-induced autonomic adaptations on different postures in adult sedentary workers. Twenty-seven sedentary workers (age 36.9 ± 9.2 years old, 13 men and 14 women) were divided into two groups: control, who continued their sedentary behavior, and experimental, who were submitted to a CWIT for 12 weeks, completing two ~40 min sessions per week. Monitoring of 8th, 16th, and 24th sessions revealed a moderate training load during sessions. Participants exhibited an improved aerobic capacity (VO2max, 34.03 ± 5.36 vs. 36.45 ± 6.05 mL/kg/min, p < 0.05) and flexibility (22.6 ± 11.4 vs. 25.3 ± 10.1 cm, p < 0.05) after the training period. In addition, they showed greater quality of life scores. However, the CWIT did not change body composition. Interestingly, more HRV parameters were improved in the seated position. The CWIT used in the current study was associated with improvements in several fitness and quality of life parameters, as well as in cardiac autonomic control of HR in adult sedentary workers. Examination of different body positions when evaluating changes in HRV appears to be a relevant aspect to be considered in further studies. Future randomized controlled trials (RCTs) with larger samples of both sexes should confirm these promising results

Produção, teor foliar e qualidade de frutos do pessegueiro 'Chimarrita' em função da adubação nitrogenada, na região da Lapa-PR Production, leaves concentracion and fruit quality of the peach orchard 'Chimarrita' as function of N fertilization, at Lapa region, Parana state - Brazil
A produção integrada de pĂȘssego (Prunus persica L. Batsch) regulamenta a dose de N a ser aplicada, a_fim de manter a produtividade, sem afetar a sanidade da planta e a qualidade ambiental. Contudo, existe uma grande carĂȘncia de informação sobre a dose de N necessĂĄria na manutenção da produtividade de pomares na regiĂŁo da Lapa. Assim, avaliaram-se doses de N na produção, no teor foliar e na qualidade de frutos da cultivar 'Chimarrita', por trĂȘs anos, em um pomar comercial com 5 anos de idade, localizado no municĂpio da Lapa-PR. O delineamento experimental foi em blocos ao acaso, com trĂȘs tratamentos e trĂȘs repetiçÔes. Os tratamentos consistiram de doses de N (40; 80 e 160 kg de N ha-1 ano-1), na forma de urĂ©ia. A redução na adubação nitrogenada em pomar estabelecido afetou negativamente a produtividade para o segundo ano. Com um decrĂ©scimo acumulado de 8,4 t ha-1 em trĂȘs anos, para a menor dose quando comparada com a maior dose. A produtividade variou com o nĂșmero de frutos, visto que a massa mĂ©dia dos frutos e o calibre nĂŁo foram suficientes para afetar a produção. A adubação de N nĂŁo afetou os aspectos qualitativos dos frutos avaliados (sĂłlidos solĂșveis totais, acidez total titulĂĄvel e firmeza da polpa), que ficaram em geral dentro dos padrĂ”es normais da cultivar. Para os teores foliares, somente o N foi afetado pela adubação nitrogenada para os trĂȘs anos avaliados, mas ficando para todos os tratamentos em nĂveis considerados normais. Os teores foliares de Ca, Mg, Fe e Zn ficaram abaixo do preconizado para o pĂȘssego.<br>The peach (Prunus persica L. However, there is a great lack of local information about N rate that it is necessary to achieve high yield for orchard at Lapa origin. So, an experiment was implanted in 5-year old commercial orchard, in the Lapa County, ParanĂĄ State, Brazil where it was evaluated N rates effect over the fruit yield, leaves concentration and quality, during three years. The experimental design was a randomized block with three treatments and three replications. The treatments were three N rates (40, 80 and 160 kg of N ha-1 ano-1), using urea as N source. The reduction in nitrogen fertilization, in established orchard, diminished the yield only for the second year. There was an accumulated yield decrease of 8,4 t ha-1 during three years, comparing the smallest to largest rate. The yield was straight associated with fruit number since the fruit mass and caliber were not affected by N application. The N fertilization did not effect fruit qualitative proprieties evaluated (total soluble solids, acidity total titratable and firmness of the pulp) and them values stayed within range concentration considered normal for cultivar. Leave tissue analysis showed that only N concentration was affected by the N fertilization for the three evaluated years, but being in normal levels for all treatments. The Ca, Mg, Fe and Zn leaves concentration was below normal level established for peach.
