    Development of a Novel Antibacterial Medicine that Targets a Characteristic Lipid of the Cell Membranes of <em>Helicobacter pylori</em>

    Helicobacter pylori is one of the most prevalent causes of gastritis. This pathogen colonizes for many years human stomach and asymptomatically leads the persons to chronic gastritis. The eradication of H. pylori from human stomach is, therefore, important in order to prevent the digestive diseases including peptic ulcers and gastric cancer that develop via chronic atrophic gastritis. Wide-spectrum antibiotics such as amoxicillin and metronidazole are used for the treatment for H. pylori infectious diseases. However, the H. pylori strains resistant to these antibiotics are increasing year by year around the world. On this basis, we need urgently to develop the antibacterial medicines that act on H. pylori with a novel mechanism. Recent studies by our group have demonstrated that H. pylori shows susceptibility to the bactericidal action of indene compounds derived from decomposition of vitamin D. The bactericidal action of indene compounds is selective not against commonplace bacteria but against H. pylori. The indene compounds turned out to target the H. pylori’s phosphatidylethanolamine that retains a myristic acid as the saturated fatty acid side chain. These findings will contribute to the development of new antibacterial medicines specialized to the treatment for H. pylori infectious diseases

    Program Management for Sustainable University CubeSat Programs Based on the Experience of Five Generations of CubeSat Projects, BIRDS Program

    BIRDS program is a university CubeSat program whose primary mission is capacity building of non-space faring countries. It has been run by Kyushu Institute of Technology, Japan, where a group of foreign and Japanese students designs, builds, tests and operates CubeSats. The program started in 2015. Roughly every year since 2017, BIRDS program delivered and launched multiple CubeSats from International Space Station. In total, the program generated 17 CubeSats in five generations. The satellites have been designed in a way so that even satellite beginners can go through the satellite system life cycles from the mission definition to the operation in two years, the duration of Master course. Subsequent generations of students overlap in the laboratory so that they can inherit the know-hows and the experience directly in-person. The satellite design has been modified based on the lessons learned in the former generations, especially during the operation phase. After going through the series of developments and operations, the satellite bus has become very mature. Currently, the initiative to open-source the BIRDS bus is also running. The program, rather than project, management aspects of university CubeSat program is discussed

    CubeSat Electrical Interface Standardization for Faster Delivery and More Mission Success

    In 2019, a new project to standardize the CubeSat electrical interface started based on the heritage of the small satellite related standard activities, such as ISO-19683 (testing) and ISO-TS-20991 (requirements). The project aims at registering at new work item at ISO/TC20/SC14 in summer 2021 by summer 2021 and publish the standard by fall 2024. Discussion has been made utilizing various gathering opportunities of the CubeSat community. A survey on the CubeSat interface has been distributed the CubeSat community to collect the satellite developers’ experience and desires regarding the interface, and the CubeSat vendors’ reality and desires. A research work to identify the difficulties associated integrating CubeSat components from different vendors is on-going. Initial finding suggest that clear definition of interface related information, especially the digital data communication, in the user manual is really needed. A framework of the standard has been drafted, which is mainly made of four parts. (1) Interface among components, (2) Interface between CubeSat bus (platform) and mission payloads, (3) Document specification to describe the information related to component interface, (4) Document specification to describe the information related to CubeSat bus (platform) interface

    Identification of reliable markers of chemo- and radiosensitivity and the key molecules that enhance the susceptibility of head and neck squamous cell carcinoma (HNSCC) to anticancer treatments is highly desirable. Previously, we have reported that regenerating gene (REG) Ⅲ expression was such a marker associated with an improved survival rate for HNSCC patients. In the present study, we investigated the stimulators for induction of REG Ⅲ expression using REG Ⅲ promoter assay in HNSCC cells transfected with REG Ⅲ promoter vector. We tested inflammatory cytokines, growth factors, polyphenols, PPARγ activator of thiazolidinediones, and histone deacetylase inhibitors, and found that 3,4',5-trihydroxy-trans-stilbene (resveratrol) significantly increased the REG Ⅲ promoter activity and the mRNA levels of REG Ⅲ in HNSCC cells. Moreover, we demonstrated the effect of resveratrol on cancer cell progression, such as cell proliferation, chemo‑ and radiosensitivity and cancer invasion of HNSCC cells. Resveratrol significantly inhibited cell growth, enhanced chemo‑ and radiosensitivity, and blocked cancer invasion of HNSCC cells.     An Overview of the BIRDS-4 Satellite Project and the First Satellite of Paraguay

    The Joint Global Multi-National Birds or BIRDS program, is a multinational small satellite project led by Kyushu Institute of Technology (Japan). The BIRDS program gives to non-space faring nations the opportunity to design, integrate, build, test, launch, and operate their country’s first satellite. This paper focuses on BIRDS-4, a constellation of three 1U CubeSats belonging to Paraguay (GuaraniSat-1: Paraguay’s first satellite), Philippines (Maya-2), and Japan (Tsuru). BIRDS-4 members are graduate students enrolled in Space Engineering International Course (SEIC) at Kyushu Institute of Technology. This constellation will execute nine missions such as Earth Imaging, Total Ionization Dose measurements, evaluation of Perovskite solar cell performance in space, and the use of “Satellite Structure as Antenna” for CW transmission. More importantly, the satellite will conduct a Store-and-Forward mission to test the technical viability of the chosen hardware, such that if proven successful, will be used for future satellite missions to gather data in remote areas. The satellites were launched on February 22, 2021 and deployed from the International Space Station on March 14, 2021.This paper describes the background, missions, stakeholders, lessons learned, and initial operational results after the deployment from ISS. Finally, this paper shall discuss the significance of this satellite for Paraguay, which has been a non-space-faring nation up until now

    日本における基質特異性拡張型β-ラクタマーゼ産生肺炎桿菌の分子遺伝学的特徴:CTX-M-15の優位性と高病原性クローンの出現

    Objective: To provide data on the molecular characteristics of extended-spectrum β-lactamase (ESBL)-producing Klebsiella pneumoniae clinical isolates in Japan. Methods: A total of 100 clinical isolates of ESBL-producing K. pneumoniae collected throughout Japan between June and July 2018 were studied. ESBL genes were analyzed using PCR and DNA sequencing. Transferability of ESBL genes was investigated by conjugation experiments. Plasmid replicon types, virulence genes (rmpA, rmpA2, iucA, iroB, and peg-344) associated with hypervirulent K. pneumoniae (hvKp), and capsule types were detected using PCR. Genotyping was performed using multilocus sequence typing. Results: All ESBL-producing isolates carried blaCTX-M genes. The most predominant CTX-M-type identified was CTX-M-15 (n=55). We identified 24 sequence types (STs) among the CTX-M-15 producers, with ST25 (n=8) being the most common. Most of the transconjugants carrying blaCTX-M-15 contained the FIIk replicon. Of the 100 ESBL-producing isolates, 31 were hvKp defined by the presence of the virulence genes. These ESBL-producing hvKp isolates belonged to eight STs (STs 23, 25, 36, 65, 86, 268, 412, and 4492), with five capsule types (K1, K2, K20, K57, and undefined). Conclusions: CTX-M-15 was the predominant ESBL among K. pneumoniae isolates from Japan.     A subcentimeter duodenal neuroendocrine neoplasm with a liver metastasis upgraded to G3: a case report

    BACKGROUND: Although duodenal neuroendocrine neoplasms (DuNENs) usually have indolent phenotypes, some DuNENs exhibit aggressive clinical manifestations. Tumor size > 1 cm, lymph node metastasis, and high grade have been associated with poor prognosis. However, preoperative risk evaluation is often difficult, because Ki-67 index on biopsy is frequently underestimated due to the intratumor heterogeneity. Here, we present a case of a subcentimeter DuNEN with a low Ki-67 index on endoscopic biopsy, who developed lymph node metastasis and high-grade liver metastasis. CASE PRESENTATION: The patient was a 52-year-old female who presented an epigastric pain. Esophagogastroduodenoscopy revealed a duodenal submucosal lesion with a size of 8 mm. The endoscopic biopsy showed DuNEN with a Ki-67 index of 3.3% (G2 categorized by the World Health Organization 2019 classification). We performed an open partial duodenectomy with adjacent lymph node dissection. Pathological examination of the resected specimens revealed a Ki-67 index of 13.5% (G2) in the "hot spot" and lymph node metastasis. A hepatic low-density area detected on preoperative contrast-enhanced computed tomography appeared to be a liver metastasis on postoperative gadoxetic acid-enhanced magnetic resonance imaging. Subsequently, we performed a laparoscopic partial hepatectomy. Pathological examination of the liver specimen showed a metastatic neuroendocrine tumor with a Ki-67 index of 27.5% (NET-G3). The patient has been alive for 14 months since the hepatectomy. CONCLUSIONS: This case shows the possibility of high malignant potential of DuNEN even if the primary lesion is < 1 cm and has a low Ki-67 index on biopsy

    Almost all cases of carbapenemase-producing Enterobacteriaceae infections in Japan are caused by blaIMP-positive Enterobacteriaceae (especially blaIMP-6) and infections caused by other types of carbapenemase-producing Enterobacteriaceae are quite rare. We examined drug resistance genes co-harboring with blaIMP-6 and their inoculum size effects. We screened β-lactamase genes, plasmid-mediated quinolone resistance (PMQR) genes, and aminoglycoside-modifying enzyme genes by PCR and performed sequencing for 14 blaIMP-6-positive Enterobacteriaceae. Further, all PMQR-positive isolates were submitted to conjugation and inoculum effect evaluation. Our data showed that 13 of the 14 isolates harbored CTX-M-2 and one co-harbored CTX-M-2 and CTX-M-1 as extended-spectrum β-lactamases. All isolates carried one or more PMQRs; aac(6’)-Ib-cr was the most prevalent (92.8%), and was followed by oqxA (64.3%), qnrS (50%), oqxAB (21.4%), and qnrB (14.3%). However, Klebsiella pneumoniae contains chromosomal OqxAB. Inoculum size effects were significant in all strains for meropenem, 13 strains for imipenem, 7 for levofloxacin, and 3 for amikacin. We observed that 11 of the experimental strains (100%), 8 strains (72.7%), and 1 strain showed inoculum size effects for meropenem, imipenem, and amikacin, respectively. However, four strains harbored qnr genes and two strains harbored qnr genes and QRDR mutations concurrently; no inoculum size effect was seen for levofloxacin. The blaIMP-6-positive Enterobacteriaceae that we studied was found to harbor at least one plasmid-mediated drug resistance gene. The inoculum size effect for carbapenems was thought to be mainly due to IMP-6-type metallo-β-lactamase; however qnrB and qnrS also had a minimal impact on the inoculum size effect for levofloxacin.博士(医学)・乙第1463号・令和2年6月30日Copyright: © 2019 Ogawa et al.     Spectral evolution of GRB 060904A observed with Swift and Suzaku -- Possibility of Inefficient Electron Acceleration

    We observed an X-ray afterglow of GRB 060904A with the Swift and Suzaku satellites. We found rapid spectral softening during both the prompt tail phase and the decline phase of an X-ray flare in the BAT and XRT data. The observed spectra were fit by power-law photon indices which rapidly changed from Γ=1.510.03+0.04\Gamma = 1.51^{+0.04}_{-0.03} to Γ=5.300.59+0.69\Gamma = 5.30^{+0.69}_{-0.59} within a few hundred seconds in the prompt tail. This is one of the steepest X-ray spectra ever observed, making it quite difficult to explain by simple electron acceleration and synchrotron radiation. Then, we applied an alternative spectral fitting using a broken power-law with exponential cutoff (BPEC) model. It is valid to consider the situation that the cutoff energy is equivalent to the synchrotron frequency of the maximum energy electrons in their energy distribution. Since the spectral cutoff appears in the soft X-ray band, we conclude the electron acceleration has been inefficient in the internal shocks of GRB 060904A. These cutoff spectra suddenly disappeared at the transition time from the prompt tail phase to the shallow decay one. After that, typical afterglow spectra with the photon indices of 2.0 are continuously and preciously monitored by both XRT and Suzaku/XIS up to 1 day since the burst trigger time. We could successfully trace the temporal history of two characteristic break energies (peak energy and cutoff energy) and they show the time dependence of t3t4\propto t^{-3} \sim t^{-4} while the following afterglow spectra are quite stable. This fact indicates that the emitting material of prompt tail is due to completely different dynamics from the shallow decay component. Therefore we conclude the emission sites of two distinct phenomena obviously differ from each other.Comment: 19 pages, 9 figures, accepted for publication in PASJ (Suzaku 2nd Special Issue
