Clonal variability of several grapevine cultivars (V. vinifera L.) grown in the Emilia-Romagna
Clonal selection has been performed over the past 2 decades by the University of Bologna to maintain the traditional grapevine cultivars grown in the Emilia-Romagna. Around 1980 budwood canes from several biotypes of the cvs Lambrusco di Sorbara, Lambrusco Salamino, Lambrusco Grasparossa, Lambrusco Maestri and Fortana were collected from old vineyards and used to establish a preliminary trial. The vines were tested for their virus status and compared for yield, grape quality, leaf characters and phenological phases in order to evaluate the biotype variability and clonal repeatability within each cultivar. L. Salamino, L. Grasparossa and L. Maestri showed very low degrees of genetic determination for yield and quality, while Fortana and L Sorbara exhibited quite high degrees. The results in both cases were independent on the virus status of the vines. While for cvs L. Salamino, L. Maestri and L. Grasparossa selection can be made only on the basis of virus status, good selection potentials were found with cvs L. Sorbara and Fortana. Fortana also exhibited marked differences in leaf morphology and phenological phases. Further investigations are needed to better characterize the diversity among biotypes of this variety, since the delimitation between cultivars and clones remains questionable
YAP and β-catenin co-operate to drive oncogenesis in basal breast cancer
Targeting cancer stem cells (CSCs) can serve as an effective approach toward limiting resistance to therapies and the development of metastases in many forms of cancer. While basal breast cancers encompass cells with CSC features, rational therapies remain poorly established. Here, we show that receptor tyrosine kinase Met signalling promotes the activity of the Hippo component YAP in basal breast cancer. Further analysis revealed enhanced YAP activity within the CSC population. Using both genetic and pharmaceutical approaches, we show that interfering with YAP activity delays basal cancer formation, prevents luminal to basal trans-differentiation and reduces CSC survival. Gene expression analysis of YAP knock-out mammary glands revealed a strong decrease in β-catenin target genes in basal breast cancer, suggesting that YAP is required for nuclear β-catenin activity. Mechanistically, we find that nuclear YAP interacts and overlaps with β-catenin and TEAD4 at common gene regulatory elements. Analysis of proteomic data from primary breast cancer patients identified a significant upregulation of the YAP activity signature in basal compared to other breast cancers, suggesting that YAP activity is limited to basal types. Our findings demonstrate that in basal breast cancers, β-catenin activity is dependent on YAP signalling and controls the CSC program. These findings suggest that targeting the YAP/TEAD4/β-catenin complex offers a potential therapeutic strategy for eradicating CSCs in basal (triple-negative) breast cancers
a case study on optimum tip speed ratio and pitch angle laws for wind turbine rotors operating in yawed conditions
The values of the tip speed ratio and blade pitch angle that yield maximum power coefficient are calculated for a rotor operating in yawed conditions. In a first step, the power coefficient is determined using a model based on the blade element momentum theory (BEMT) which includes a Prandtl-Glauert root-tip losses correction, a non-uniform model for the axial and tangential induction factors, and a model of the rotational augmentation effects. The BEMT model is validated with the experimental data from the NREL-UAE. The maximum values of the power coefficient are determined for different yaw angles and the corresponding values of the tip speed ratio and blade control angle are obtained. The maximum power coefficient using these optimum laws is compared to the maximum power coefficient using the optimum laws of the non-yawed case and it is shown that there is a gain in the power coefficient. For the case study presented in this paper it has been found that for yaw angles of 30° about 10% of the power coefficient can be recovered
Role of Portion Size in the Context of a Healthy, Balanced Diet: A Case Study of European Countries
Over the past decades, a generalised increase in food portion sizes has probably contributed to the growing global obesity epidemic. Increasing awareness of appropriate portion sizes could contribute to reversing this trend through better control of calorie intake. In this study, a comparison of standard portion sizes in European countries for various food categories shows a wide variability of their importance for food, nutrient, and energy consumption according to government and institutional websites. On the other hand, the overall averages appear to be largely in line with the values indicated by the Italian Society of Human Nutrition, which is the most comprehensive and detailed document among those evaluated. The exceptions are milk and yoghurt, for which the reference portions in Europe are generally higher, and vegetables and legumes, for which portions are smaller than those reported in the Italian document. Moreover, the portion sizes of staple foods (e.g., pasta and potatoes) vary according to different food traditions. It is reasonable to consider that the creation of harmonised standard reference portions common to the European countries, based on international guidelines and scientific evidence, would significantly contribute to consumers' nutritional education and ability to make informed choices for a healthy diet
