    Grandisin induces apoptosis in leukemic K562 cells

    In this study, the potential antileukemic activity of grandisin, a lignan extracted from Piper solmsianum, was evaluated against the leukemic line K562. The cytotoxicity of grandisin (0.018 to 2.365 µM) was evaluated in K562 and normal peripheral blood lymphocytes by Trypan Blue Exclusion and MTT methods after 48h exposure to the drug. In both methods, cellular viability was concentration-dependent and the IC50 values were lower than 0.85µM. Analysis of K562 cells after treatment with grandisin showed that the cell cycle was arrested in the G1 phase with a 12.31% increase, while both S and G2 phases decreased. Morphological studies conducted after the exposure of K562 to grandisin revealed changes consistent with the apoptosis process, which was confirmed by anexin V stain and caspase activation. Thus, lignan grandisin showed antileukemic activities against the K562 cell line and the cell death process occurred via apoptosis

    Principais complicações pós-colecistectomia: uma revisão de literatura

    This article aims to analyze the main post-cholecystectomy complications. Integrative literature review carried out through a search in the Pubmed, Scielo, Lilacs and BVS databases, with publication time limited between the years 2019 and 2023. After evaluation by 2 reviewers, duplicate texts or those related to the topic were excluded . 6 articles were analyzed. In the year 2023, it had the highest number of articles published (n=4). The main aspects highlighted in the articles analyzed were early complications, such as bleeding, acute pancreatitis and perforation. Given the data presented, there is a need for a multidisciplinary approach to avoid post-cholecystectomy complications.Este artigo tem por objetivo analisar quais são as principais complicações pós-colecistectomia. Revisão de literatura integrativa feita por meio da busca nas bases de dados Pubmed, Scielo, Lilacs e BVS, com tempo de publicação delimitado entre o ano de 2019 e 2023. Após avaliação por 2 revisores, foram excluídos os textos duplicados ou que tangenciam o tema. Foram analisados 6 artigos. No ano de 2023, obteve o maior número de artigos publicados (n=4). Os principais aspectos ressaltados nos artigos analisados foam complicações precoces, como sangramentos, pancreatite aguda e perfuração. Diante dos dados apresentados, constata-se a necessidade de uma abordagem multiprofissional para evitar as complicações pós-colecistectomia

    Planos alimentares no consumo, na digestibilidade e no desempenho de novilhas leiteiras de distintos grupos raciais

    The objective of this work was to evaluate the effects of nutritional plans and breed composition on nutrient intake and digestibility, nitrogen balance, and performance of dairy heifers in tropical conditions. Thirty-six heifers – 12 Holstein, 12 Gyr, and 12 Gyrolando F1 (½ Holstein × ½ Gyr) – were housed in a tie-stall system and randomly distributed, adopting a completely randomized design in a 3×3 factorial arrangement (nutritional plans and breed composition). The diets were formulated to enable weight gains of 200, 400, and 800 g per day, corresponding to the nutritional plans 11, 14, and 19 g kg-1 body weight (BW), respectively. The intake of dry matter, fibrous fractions (g kg-1 BW-0.75), and metabolizable energy (kcal-1 BW-0.75) in the 11-g kg-1 plan were greater for Gyr heifers than for Gyrolando and Holstein heifers. Ether extract digestibility was 13.85% higher for Gyr heifers compared with Gyrolando F1 in the 19-g kg-1 plan. The increase in the nutritional plans shows a quadratic response to nutrient intake and a linear response to average daily gain, but does not affect the digestibility coefficients. Gyr and Gyrolando F1 heifers present similar nutritional efficiency to that of Holstein heifers.O objetivo deste trabalho foi avaliar os efeitos do plano alimentar e do grupo racial sobre o consumo e a digestibilidade de nutrientes, o balanço de nitrogênio e o desempenho de novilhas leiteiras em condições tropicais. Trinta e seis novilhas – 12 Holandês, 12 Gir e 12 Girolando F1 (½ Holandês × ½ Gir) – foram alojadas em sistema “tie-stall” e distribuídas aleatoriamente, tendo-se adotado delineamento inteiramente casualizado, em arranjo fatorial 3x3 (planos alimentares e grupos raciais). As dietas foram formuladas para possibilitar ganhos de peso de 200, 400 e 800 g por dia, correspondentes aos planos alimentares de 11, 14 e 19 g kg-1 de peso corporal (PC), respectivamente. As novilhas Gir dentro do plano de 11 g kg-1 consumiram mais matéria seca, fração fibrosa (g kg-1 de PC-0,75) e energia metabolizável (kcal de PC-0,75) do que as novilhas Girolando e Holandês. A digestibilidade do extrato etéreo foi 13,85% superior para as novilhas Gir, em comparação às Girolando F1 no plano de 19 g kg-1. O aumento do plano nutricional tem efeito quadrático no consumo de nutrientes e linear no ganho de peso médio diário, mas não influencia o coeficiente de digestibilidade. Novilhas Gir leiteiras e Girolando F1 apresentam eficiência nutricional semelhante à das novilhas Holandês

    The inventory of geological heritage of the state of SĂŁo Paulo, Brazil: Methodological basis, results and perspectives

    An inventory of geological sites based on solid and clear criteria is a first step for any geoconservation strategy. This paper describes the method used in the geoheritage inventory of the State of SĂŁo Paulo, Brazil, and presents its main results. This inventory developed by the geoscientific community aimed to identify geosites with scientific value in the whole state, using a systematic approach. All 142 geosites representative of 11 geological frameworks were characterised and quantitatively evaluated according to their scientific value and risk of degradation, in order to establish priorities for their future management. An online database of the inventory is under construction, which will be available to be easily consulted and updated by the geoscientific community.     Residual biomass potential of commercial and pre-commercial sugarcane cultivars

    Sugarcane (Saccharum spp.) is an efficient and sustainable alternative for energy generation compared to non-renewable sources. Currently, during the mechanized harvest process, the straw left in the field can be used in part for the second generation ethanol and increasing the electric energy production. Thus, this study aimed to provide information on the potential for residual biomass cultivars of sugarcane cropping system. This study provides the following information: yield of straw, depending on the calculated leaf area index and the number of tillers per linear meter; primary energy production of several sugarcane genotypes; contribution of dry tops and leaves; biomass yield; and evaluation of fiber, cellulose, hemicellulose and lignin. Preliminary results obtained by researchers of the State of SĂŁo Paulo, Brazil, and reCviews related studies are presented. The results suggest that the production of sugarcane straw content varies according to the cultivars; the greater mass of sugarcane straw is in the top leaves and that the potential for the crude energy production of sugarcane per area unit can be increased using fiber-rich species or species that produce more straw. The straw indexes was shown to be a good indicator and allow the estimation of straw volumes generated in a sugarcane crop. The cellulose, hemicellulose and lignin composition in sugarcane is distinct among varieties. Therefore, it is possible to develop distinct biomass materials for energy production and for the development of sugarcane mills using biochemical processes and thermal routes

    Pervasive gaps in Amazonian ecological research

    Biodiversity loss is one of the main challenges of our time,1,2 and attempts to address it require a clear un derstanding of how ecological communities respond to environmental change across time and space.3,4 While the increasing availability of global databases on ecological communities has advanced our knowledge of biodiversity sensitivity to environmental changes,5–7 vast areas of the tropics remain understudied.8–11 In the American tropics, Amazonia stands out as the world’s most diverse rainforest and the primary source of Neotropical biodiversity,12 but it remains among the least known forests in America and is often underrepre sented in biodiversity databases.13–15 To worsen this situation, human-induced modifications16,17 may elim inate pieces of the Amazon’s biodiversity puzzle before we can use them to understand how ecological com munities are responding. To increase generalization and applicability of biodiversity knowledge,18,19 it is thus crucial to reduce biases in ecological research, particularly in regions projected to face the most pronounced environmental changes. We integrate ecological community metadata of 7,694 sampling sites for multiple or ganism groups in a machine learning model framework to map the research probability across the Brazilian Amazonia, while identifying the region’s vulnerability to environmental change. 15%–18% of the most ne glected areas in ecological research are expected to experience severe climate or land use changes by 2050.     Hypertrophic cardiomyopathy (HCM) is a form of genetically caused heart muscle disease, characterized by the thickening of the ventricular walls. Diagnosis requires detection through imaging methods (Echocardiogram or Cardiac Magnetic Resonance) showing any segment of the left ventricular wall with a thickness > 15 mm, without any other probable cause. Genetic analysis allows the identification of mutations in genes encoding different structures of the sarcomere responsible for the development of HCM in about 60% of cases, enabling screening of family members and genetic counseling, as an important part of patient and family management. Several concepts about HCM have recently been reviewed, including its prevalence of 1 in 250 individuals, hence not a rare but rather underdiagnosed disease. The vast majority of patients are asymptomatic. In symptomatic cases, obstruction of the left ventricular outflow tract (LVOT) is the primary disorder responsible for symptoms, and its presence should be investigated in all cases. In those where resting echocardiogram or Valsalva maneuver does not detect significant intraventricular gradient (> 30 mmHg), they should undergo stress echocardiography to detect LVOT obstruction. Patients with limiting symptoms and severe LVOT obstruction, refractory to beta-blockers and verapamil, should receive septal reduction therapies or use new drugs inhibiting cardiac myosin. Finally, appropriately identified patients at increased risk of sudden death may receive prophylactic measure with implantable cardioverter-defibrillator (ICD) implantation.La miocardiopatĂ­a hipertrĂłfica (MCH) es una forma de enfermedad cardĂ­aca de origen genĂ©tico, caracterizada por el engrosamiento de las paredes ventriculares. El diagnĂłstico requiere la detecciĂłn mediante mĂ©todos de imagen (Ecocardiograma o Resonancia MagnĂ©tica CardĂ­aca) que muestren algĂşn segmento de la pared ventricular izquierda con un grosor > 15 mm, sin otra causa probable. El análisis genĂ©tico permite identificar mutaciones en genes que codifican diferentes estructuras del sarcĂłmero responsables del desarrollo de la MCH en aproximadamente el 60% de los casos, lo que permite el tamizaje de familiares y el asesoramiento genĂ©tico, como parte importante del manejo de pacientes y familiares. Varios conceptos sobre la MCH han sido revisados recientemente, incluida su prevalencia de 1 entre 250 individuos, por lo tanto, no es una enfermedad rara, sino subdiagnosticada. La gran mayorĂ­a de los pacientes son asintomáticos. Por fim, os pacientes adequadamente identificados com risco aumentado de morta sĂşbita podem receber medida profilática com implante de cardiodesfibrilador implantável (CDI)
