    Mutations in porin LamB contribute to ceftazidime-avibactam resistance in KPC-producing Klebsiella pneumoniae.

    Ceftazidime-avibactam (CAZ-AVI) shows promising activity against carbapenem-resistant Klebsiella pneumoniae (CRKP), however, CAZ-AVI resistance have emerged recently. Mutations in KPCs, porins OmpK35 and/or OmpK36, and PBPs are known to contribute to the resistance to CAZ-AVI in CRKP. To identify novel CAZ-AVI resistance mechanism, we generated 10 CAZ-AVI-resistant strains from 14 CAZ-AVI susceptible KPC-producing K. pneumoniae (KPC-Kp) strains through in vitro multipassage resistance selection using low concentrations of CAZ-AVI. Comparative genomic analysis for the original and derived mutants identified CAZ-AVI resistance-associated mutations in KPCs, PBP3 (encoded by ftsI), and LamB, an outer membrane maltoporin. CAZ-AVI susceptible KPC-Kp strains became resistant when complemented with mutated blaKPC genes. Complementation experiments also showed that a plasmid borne copy of wild-type lamB or ftsI gene reduced the MIC value of CAZ-AVI in the induced resistant strains. In addition, blaKPC expression level increased in four of the six CAZ-AVI-resistant strains without KPC mutations, indicating a probable association between increased blaKPC expression and increased resistance in these strains.     Purpose: To explore the efficacy and safety of L-asparaginase in newly-diagnosed extranodal nature killer (NK)/T -cell lymphoma (ENKTL), we conducted a prospective phase II study of L-asparaginase, cyclophosphamide, vincristine, doxorubicin and dexamethasone (CHOP-L) regimen in combination with radiotherapy. Patients and methods: Patients with newly diagnosed ENKTL and an ECOG performance status of 0 to 2 were eligible for enrollment. Treatment included 6-8 cycles of CHOP-L (cyclophosphamide, 750 mg/m(2) day 1; vincristine, 1.4 mg/m(2) day 1 (maximal dose 2 mg), doxorubicin 50 mg/m(2) day 1; dexamethasone 10 mg days 1-8; L-asparaginase 6000 u/m(2) days 2-8). Radiotherapy was scheduled after 4-6 cycles of CHOP-L regimen, depending on stage and primary anatomic site. The primary endpoint was complete response (CR) rate. Results: A total of 38 eligible patients were enrolled. The median age was 40.5 years (range, 15 to 71 years). Their clinical characteristics were male to female ratio, 24: 14; Ann Arbor stage I, 20; II, 11; III, 3; IV, 4. CR and overall response rates were 81.6% (95% CI, 69.3% to 93.9%) and 84.2%, respectively. With a median follow-up of 25 months, the 2-year overall survival, progression-free survival and disease-free survival rates were 80.1% (95% CI, 73.3% to 86.9%), 81% (95% CI, 74.5% to 87.5%) and 93.6% (95% CI, 89.3% to 97.9%), respectively. The major adverse events were myelosuppression, liver dysfunction, and digestive tract toxicities. Grade 3 to 4 leukopenia and neutropenia were 76.3% and 84.2%, respectively. No treatment-related death was observed. Conclusion: CHOP-L chemotherapy in combination with radiotherapy is a safe and highly effective treatment for newly diagnosed ENKTL.OncologyHematologySCI(E)9ARTICLEnull

    Global media as an early warning tool for food fraud; an assessment of MedISys-FF

    Food fraud is a serious problem that may compromise the safety of the food products being sold on the market. Previous studies have shown that food fraud is associated with a large variety of food products and the fraud type may vary from deliberate changing of the food product (i.e. substitution, tampering, dilution etc.) to the manipulation of documents. It is therefore important that all actors within the food supply chain (food producers, authorities), have methodologies and tools available to detect fraudulent products at an early stage so that preventative measures can be taken. Several of such systems exist (i.e. iRASFF, EMA, HorizonScan, AAC-FF, MedISys-FF), but currently only MedISys-FF is publicly online available. In this study, we analyzed food fraud cases collected by MedISys-FF over a 6-year period (2015ā€“2020) and show global trends and developments in food fraud activities. In the period investigated, the system has collected 4375 articles on food fraud incidents from 164 countries in 41 different languages. Fraud with meat and meat products were most frequently reported (27.7%), followed by milk and milk products (10.5%), cereal and bakery products (8.3%), and fish and fish products (7.7%). Most of the fraud was related to expiration date (58.3%) followed by tampering (22.2%) and mislabeling of country of origin (11.4%). Network analysis showed that the focus of the articles was on food products being frauded. The validity of MedISys-FF as an early warning system was demonstrated with COVID- 19. The system has collected articles discussing potential food fraud risks due to the COVID-19 crisis. We therefore conclude that MedISys-FF is a very useful tool to detect early trends in food fraud and may be used by all actors in the food system to ensure safe, healthy, and authentic food

    The organic market is growing, and confidence is key. However, it is hard to distinguish an organic product from another regular product. In this article we examined a consumer NIRS-Vis device for its capabilities to distinguish organic retail milks from conventional milks. The study revealed differences between the two groups, allowing classification of samples, but these differences were not as distinct as those provided by other devices examined previously. Nevertheless, with some improvement of performance of this kind of devices and integration in smart phones, authentication of foods is within reach of citizens in the near future

    Objective. As one of the worldā€™s cultural heritages, Dunhuang murals contain profound cultural, historical, social, and artistic values. In this paper, the evolution and progress of character costumes are analyzed and the development process of Chinese costumes is studied. Methods. This paper compares the characteristics of the costumes in different times; the progress and changes in graphics, styles, and textures; and the different characteristics from the costumes in the modern society. It also compares the coupling degree data of clothing culture in the two periods. Through analysis and research it is found that, compared with traditional algorithms, the integration of clothing culture using deep learning algorithms is more coupling. Through the calculation and analysis of clothing evolution in Dunhuang murals, more design inspiration can be provided for Chinese clothing designers, and more strength and research materials can be contributed to future design and clothing development in combination with ancient and modern clothing culture

    Deoxynivalenol is a common mycotoxin in grains such as wheat and corn. Contaminated food can seriously affect the health of humans and animals. Hence, to ensure the safety and quality of food at crucial segment, it is essential to manage the risk of Fusarium infection and deoxynivalenol (DON). Nowadays, countries around the world attach great importance to predict the risk of food mycotoxins contamination. The research about the prediction and early warning of DON contamination has been progressed recent years. The results would help to reveal the relationship between contamination risk and agronomical information to develop the prediction model. This paper reviewed the latest research results of the prediction of the DON contamination, aiming to summarize the existing research methods and results, and provide the basis for the early warning of mycotoxin in China.</p
