    Collecting, archiving and processing DNA from wildlife samples using FTA(® )databasing paper

    BACKGROUND: Methods involving the analysis of nucleic acids have become widespread in the fields of traditional biology and ecology, however the storage and transport of samples collected in the field to the laboratory in such a manner to allow purification of intact nucleic acids can prove problematical. RESULTS: FTA(® )databasing paper is widely used in human forensic analysis for the storage of biological samples and for purification of nucleic acids. The possible uses of FTA(® )databasing paper in the purification of DNA from samples of wildlife origin were examined, with particular reference to problems expected due to the nature of samples of wildlife origin. The processing of blood and tissue samples, the possibility of excess DNA in blood samples due to nucleated erythrocytes, and the analysis of degraded samples were all examined, as was the question of long term storage of blood samples on FTA(® )paper. Examples of the end use of the purified DNA are given for all protocols and the rationale behind the processing procedures is also explained to allow the end user to adjust the protocols as required. CONCLUSIONS: FTA(® )paper is eminently suitable for collection of, and purification of nucleic acids from, biological samples from a wide range of wildlife species. This technology makes the collection and storage of such samples much simpler

    To remove or to replace traditional electronic games? A crossover randomised controlled trial on the impact of removing or replacing home access to electronic games on physical activity and sedentary behaviour in children aged 10-12 years.

Available from BMJ Publishing Group via the DOI in this record.OBJECTIVE: To evaluate the impact of (1) the removal of home access to traditional electronic games or (2) their replacement with active input electronic games, on daily physical activity and sedentary behaviour in children aged 10-12 years. DESIGN: Crossover randomised controlled trial, over 6 months. SETTING: Family homes in metropolitan Perth, Australia from 2007 to 2010. PARTICIPANTS: 10-year-old to 12-year-old children were recruited through school and community media. From 210 children who were eligible, 74 met inclusion criteria, 8 withdrew and 10 had insufficient primary outcome measures, leaving 56 children (29 female) for analysis. INTERVENTION: A counterbalanced randomised order of three conditions sustained for 8 weeks each: no home access to electronic games, home access to traditional electronic games and home access to active input electronic games. MAIN OUTCOME MEASURES: Primary outcome was accelerometer assessed moderate/vigorous physical activity (MVPA). Secondary outcomes included sedentary time and diary assessed physical activity and sedentary behaviours. RESULTS: Daily MVPA across the whole week was not significantly different between conditions. However, compared with home access to traditional electronic games, removal of all electronic games resulted in a significant increase in MVPA (mean 3.8 min/day, 95% CI 1.5 to 6.1) and a decrease in sedentary time (4.7 min/day, 0.0 to 9.5) in the after-school period. Similarly, replacing traditional games with active input games resulted in a significant increase in MVPA (3.2 min/day, 0.9 to 5.5) and a decrease in sedentary time (6.2 min/day, 1.4 to 11.4) in the after-school period. Diary reports supported an increase in physical activity and a decrease in screen-based sedentary behaviours with both interventions. CONCLUSIONS: Removal of sedentary electronic games from the child's home and replacing these with active electronic games both resulted in small, objectively measured improvements in after-school activity and sedentary time. Parents can be advised that replacing sedentary electronic games with active electronic games is likely to have the same effect as removing all electronic games.     A time-resolved fluorescence immunoassay for the measurement of testosterone in saliva: Monitoring of testosterone replacement therapy with testosterone buciclate

    Monitoring of testosterone replacement therapy requires a reliable method for testosterone measurement. Determination of salivary testosterone, which reflects the hormone's biologically active plasma fraction, is a superior technique for this purpose. The aim of the present study was to establish a new sensitive time-resolved fluorescence immunoassay for the accurate measurement of testosterone levels in saliva and to validate it by monitoring testosterone replacement therapy in eight hypogonadal men. A clinical phase I- study with the new ester testosterone buciclate was performed to search for new testosterone preparations to produce constant serum levels in the therapy of male hypogonadism. After two control examinations eight male patients with primary hypogonadism were randomly assigned to two treatment groups (n = 2x4) and given single doses of either 200 mg (group I) or 600 mg (group II) testosterone buciclate intramuscularly. Saliva and blood samples were obtained 1, 2, 3, 5 and 7 days post injection and then weekly for three months. The time-resolved fluorescence immunoassay for salivary testosterone shows a detection limit of 16 pmol/l, an intra-assay CV of 8.9% (at a testosterone concentration of 302 pmol/l), an inter-assay CV of 8.7% (at a testosterone concentration of 305 pmol/l) and a good correlation with an established radioimmunsassay of r = 0.89. The sample volume required by this method is only 180 mu l for extraction and duplicate determination. The assay procedure requires no more than three hours. In group I (200 mg) testosterone did not increase to normal levels either in saliva or in serum. However, in group II, androgen levels increased significantly and were maintained in the normal range for up to 12 weeks with maximal salivary testosterone levels of 303 +/- 18 pmol/l (mean+/-SE) and maximal testosterone levels of 13.1 +/- 0.9 nmol/l (mean+/-SE) in serum in study week 6 and 7. The time-resolved fluorescence immunoassay for salivary testosterone provides a useful tool for monitoring androgen status in men and women and is well suited for the follow-up of testosterone replacement therapy on an outpatient basis. The long-acting ester testosterone buciclate is a promising agent for substitution therapy of male hypogonadism and in combination with testosterone monitoring in saliva offers an interesting new perspective for male contraception

    A mathematical modelling study of an athlete's sprint time when towing a weighted sled

The final publication is available at Springer via http://dx.doi.org/10.1007/s12283-013-0114-2.This study used a mathematical model to examine the effects of the sled, the running surface, and the athlete on sprint time when towing a weighted sled. Simulations showed that ratio scaling is an appropriate method of normalising the weight of the sled for athletes of different body size. The relationship between sprint time and the weight of the sled was almost linear, as long as the sled was not excessively heavy. The athlete’s sprint time and rate of increase in sprint time were greater on running surfaces with a greater coefficient of friction, and on any given running surface an athlete with a greater power-to-weight ratio had a lower rate of increase in sprint time. The angle of the tow cord did not have a substantial effect on an athlete’s sprint time. This greater understanding should help coaches set the training intensity experienced by an athlete when performing a sled-towing exercise

    Inherited polyglutamine spinocerebellar ataxias in South Africa

    Objective. To determine the frequency and distribution ofpolyglutamine spinocerebellar ataxias (SCAs) from referrals over a24-year period to the National Health Laboratory Service (NHLS)in South Africa (SA).Methods. Paper-based clinical reports in the University ofCape Town laboratory and the NHLS electronic patient recorddatabase spanning a 24-year period were mined for informationregarding the molecular diagnosis, ethnicity and CAG repeatlength for individuals referred for molecular genetic testing for thepolyglutamine SCAs.Results. SCA1 and 7 are the most frequent types of polyglutamineSCA in the SA patient population, followed by SCA2, 3 and 6.SCA1 is the most common type in the coloured, white and Indianpopulations, whereas the majority of indigenous black Africanpatients are affected with SCA7 and 2. Of individuals tested, 22%were found to be positive for one of the polyglutamine SCAs.Conclusion. Although trends in the frequency and distributionof the polyglutamine SCAs in SA have not changed significantlysince our previous study in 2003, they differ remarkably from thosereported elsewhere, and reflect the unique genetic and demographicbackground of SA. The provision of accurate and complete patientinformation and family history is crucial to the diagnostic process,to enable comprehensive epidemiological studies and assist indeveloping therapeutic and patient management strategies

    Spiritual wellbeing and its relationship to adolescent resilience. A case study of Australian youth attending one local church.

    The purpose of this study was to examine the relationships and connections within the family and the local Christian church community influencing spiritual wellbeing of young people engaged with Christian spirituality. Questions have arisen recently about the role of spiritual wellbeing in strengthening resilience of youth. To explore this association, this research focused on the relationships and connectedness of people who attend one religious organisation as one means of enhancing their spiritual wellbeing. Two separate but complementary theories underpinned this research: the bioecological theory of human development (Bronfenbrenner 2001a) and the international family strengths model (DeFrain & Asay 2007a). In line with the purposes of an instrumental case study, different sources of data (quantitative and qualitative) were collected on the phenomenon of interest—spiritual wellbeing. Utilising a survey method, a theoretical purposive sample of sixty five people participated in this study. Through an abductive analysis process, the research identified a model of five spiritual strengths that enhance peace and life satisfaction and strengthen youth resilience within the lives of the young people in this study. Spiritual wellbeing and resilience were shown to be interrelated and ecologically bound. This case study presents one possible explanation for the often observed yet poorly understood relationship between spiritual wellbeing and positive youth outcomes, such as resilience. The author recommends measurement of resilience and strategies that aim to strengthen youth resilience should include spiritual wellbeing however, further research is required that considers how best to incorporate spiritual wellbeing into both resilience measures and health promoting strategies. It is also recommended that replica studies with people who belong to other spirituality types, and who may implement differing spiritual practices, are needed to test the proposed model of spiritual strengths and the identified relationship to resilience

    In this paper we develop a formal dynamic version of Chain Event Graphs (CEGs), a particularly expressive family of discrete graph- ical models. We demonstrate how this class links to semi-Markov models and provides a convenient generalization of the Dynamic Bayesian Network (DBN). In particular we develop a repeating time-slice Dynamic CEG providing a useful and simpler model in this family. We demonstrate how the Dynamic CEG’s graphical formulation exhibits asymmetric conditional independence statements and also how each model can be estimated in a closed form enabling fast model search over the class. The expressive power of this model class together with its estimation is illustrated throughout by a variety of examples that include the risk of childhood hospitalization and the efficacy of a flu vaccine
