    Multi-residues analysis of pre-emergence herbicides in fluvial sediments : application to the mid-Garonne River

    Contamination of man and ecosystems by pesticides has become a major environmental concern. Whereas many studies exist on contamination from agriculture, the effects of urban sources are usually omitted. Fluvial sediment is a complex matrix of pollutants but little is known of its recent herbicide content. This study proposes a method for a fast and reliable analysis of herbicides by employing the accelerated solvent extractor (ASE). The aim of the study is to show the impact of a major town (Toulouse) on the herbicide content in the river. In this study, three herbicide families (i.e. s-triazine, substituted ureas and anilides) were analysed in fluvial sediment fractions at 11 sampling sites along the mid-Garonne River and its tributaries. River water contamination by herbicides is minor, except for at three sites located in urban areas. Among the herbicidal families studied, urban and suburban areas are distinguished from rural areas and were found to be the most contaminated sites during the study period, a winter low-water event. The herbicide content of the coarse sediment fractions is about one third of that found in the fine fractions and usually ignored. The distribution of pesticide concentrations across the whole range of particle sizes was investigated to clarify the role of plant remains on the significant accumulation in the coarse fractions

    Urban Impact on Selected Pre-Emergence Herbicides in Sediment cores

    In order to determine if pre-emergence herbicide pollutant source, mixing with many others from residential and industrial activities, has an effect on pollutant degradation, three sediment cores were sampled in appropriate sites of the Garonne river near the city of Toulouse: two in rural surroundings, one upstream and one downstream from the urban area away from its influence, and one downtown close to Toulouse. Atrazine and DEA were analysed and, using DAR pesticide/metabolite ratio, an inferior metabolisation ratio was highlighted in the urban sediment compared to the rural ones, regardless of sedimentation rate, organic carbon content, topography or differences in the intensity of surrounding activities between rural cores

    Herbicide accumulation and evolution in reservoir sediments

    The aim of the present study was to understand the effect of reservoir configurations on sediment pesticide fate. Two dams were selected on the River Garonne, in southwest France: Carbonne and Golfech, both with reservoirs subject to accumulation of herbicide-contaminated sediment. They are situated upstream and downstream respectively of an agricultural and urban area: the Mid-Garonne. The results presented include pesticide concentrations and C/N ratios in the smaller sediment particles (b2 mm) and values of oxygenation and herbicide concentrations in the water. The dynamic behaviour of sediment in the reservoirs is discussed. The present study shows that the theoretical lifespan (weak remanence in vitro) and the results actually observed in the sediment are conflicting. Pesticide contamination in Carbonne indicates conservation, even accumulation, of herbicide molecules while in Golfech transformation processes clearly dominate. The hydromorphological position of Golfech reservoir, i.e. located at the junction of two rivers with contrasting hydrological regimes and very different oxygenation conditions, leads to accelerated pesticide desorption or degradation.     Erythropoietic protoporphyria

    Erythropoietic protoporphyria (EPP) is an inherited disorder of the haem metabolic pathway characterised by accumulation of protoporphyrin in blood, erythrocytes and tissues, and cutaneous manifestations of photosensitivity. EPP has been reported worldwide, with prevalence between 1:75,000 and 1:200,000. It usually manifests in early infancy upon the first sun exposures. EPP is characterised by cutaneous manifestations of acute painful photosensitivity with erythema and oedema, sometimes with petechiae, together with stinging and burning sensations upon exposure to sunlight, without blisters. These episodes have a variable severity depending on the exposure duration and may result in chronic permanent lesions on exposed skin. As protoporphyrin is a lipophilic molecule that is excreted by the liver, EPP patients are at risk of cholelithiasis with obstructive episodes, and chronic liver disease that might evolve to rapid acute liver failure. In most patients, EPP results from a partial deficiency of the last enzyme of the haem biosynthetic pathway, ferrochelatase, EC 4.99.1.1/FECH (encoded by the FECH gene). EPP appears to be inherited as an autosomal dominant disease, the clinical expression of which is modulated by the presence of the hypomorphic FECH IVS3-48C allele trans, but recessive inheritance with two mutated FECH alleles has also been described. In about 2% of patients, overt disease was recently shown to be caused by gain-of-function mutations in the erythroid-specific aminolevulinic acid synthase 2 (ALAS2/ALAS, EC 2.3.1.27) gene and named X-linked dominant protoporphyria. Diagnosis is established by finding increased levels of protoporphyrin in plasma and red blood cells, and detection of a plasma fluorescence peak at 634 nm. Investigations for hepatic involvement, ferrochelatase activity level, genetic analysis (FECH mutations, presence of the hypomorphic FECH IVS3-48C allele trans and ALAS2 mutations) and family studies are advisable. Differential diagnosis includes phototoxic drug reactions, hydroa vacciniforme, solar urticaria, contact dermatitis, angio-oedema and, in some cases, other types of porphyria. Management includes avoidance of exposure to light, reduction of protoporphyrin levels and prevention of progression of possible liver disease to liver failure. As the major risk in EPP patients is liver disease, a regular follow-up of hepatic involvement is essential. Sequential hepatic and bone marrow transplantation should be considered as a suitable treatment for most severe cases of EPP with hepatic involvement. EPP is a lifelong disorder whose prognosis depends on the evolution of the hepatic disease. However, photosensitivity may have a significant impact on quality of life of EPP patients

    Sex differences in cerebral venous sinus thrombosis after adenoviral vaccination against COVID-19

    Introduction: Cerebral venous sinus thrombosis associated with vaccine-induced immune thrombotic thrombocytopenia (CVST-VITT) is a severe disease with high mortality. There are few data on sex differences in CVST-VITT. The aim of our study was to investigate the differences in presentation, treatment, clinical course, complications, and outcome of CVST-VITT between women and men. Patients and methods: We used data from an ongoing international registry on CVST-VITT. VITT was diagnosed according to the Pavord criteria. We compared the characteristics of CVST-VITT in women and men. Results: Of 133 patients with possible, probable, or definite CVST-VITT, 102 (77%) were women. Women were slightly younger [median age 42 (IQR 28–54) vs 45 (28–56)], presented more often with coma (26% vs 10%) and had a lower platelet count at presentation [median (IQR) 50x109/L (28–79) vs 68 (30–125)] than men. The nadir platelet count was lower in women [median (IQR) 34 (19–62) vs 53 (20–92)]. More women received endovascular treatment than men (15% vs 6%). Rates of treatment with intravenous immunoglobulins were similar (63% vs 66%), as were new venous thromboembolic events (14% vs 14%) and major bleeding complications (30% vs 20%). Rates of good functional outcome (modified Rankin Scale 0-2, 42% vs 45%) and in-hospital death (39% vs 41%) did not differ. Discussion and conclusions: Three quarters of CVST-VITT patients in this study were women. Women were more severely affected at presentation, but clinical course and outcome did not differ between women and men. VITT-specific treatments were overall similar, but more women received endovascular treatment.</p

    Le travail concerne le bassin hydrographique de la Garonne en utilisant les peuplements des poissons comme outils d'analyses et de gestion de la qualité environnementale. La prémière partie aborde l'approche globale à l'échelle du bassin versant. Les patrons de diversité du peuplement de poissons, selon le profil longitudinal et les guildes, en relation avec les facteurs physiques et environnementaux ont été réalisés. L'analyse statistique s'effectue par des méthodes multivariées y compris les réseaux de neurones artificiels. La deuxième partie concerne l'approche à l'échelle locale où deux cas d'anthropisation ont été étudiés.     Observations of the range of fish species in the middle Garonne. By using electrical and net fishing and by making observations at various fish ladders a list of species in the Garonne between Toulouse and Agen has been drawn up. These sectors correspond, moving downstream, to the barbel, bream and euryhaline species zones. Thirty-five species have been found, with cyprinoids being dominant. Carnivores make up less than 10 % of the population sampled. Migrating fish have been observed at the Golfech fish ladder. Recent work carried out to overcome the obstacles that Toulouse and Golfech represented will bring about a balancing out of fish populations according to their natural river habitat zones, particularly for the migrators.Observaciones sobre la fauna de peces del Garona medio. Pescas eléctricas y con redes, así como observaciones en las pasadas de peces permitieron elaborar una lista de las especies que pueblan el Garona entre Toulouse y Agen. Estos sectores corresponden, de arriba abajo, a la zona del barbo, de la brema y de especies euriálinas. Se contaron treinta y cinco especies con predominancia de ciprínidos. Los carniceros no alcanzan los 10 % de la muestra. Los migratorios fueron observados fácilmente en el ascensor de peces de Golfech.     Les changements climatiques et leurs impacts sur la biodiversité font aujourd'hui l'objet d'une attention croissante de la part de la communauté scientifique et des gestionnaires des écosystèmes naturels. En effet, le climat influence la biologie et l'écologie des espèces animales et végétales, depuis leur physiologie jusqu'à leur répartition. Les modifications climatiques pourraient donc avoir des répercussions importantes sur les espèces et les assemblages. Au sein des écosystèmes aquatiques continentaux, les poissons de rivière sont des organismes incapables de réguler leur température corporelle et soumis à une variabilité hydrologique importante ainsi qu'à de fortes pressions anthropiques. Leur réponse aux modifications du climat actuelles et à venir a pourtant été encore peu abordée. L'objectif de ce travail de thèse est donc d'évaluer les impacts potentiels du changement climatique sur les poissons des rivières françaises, et plus particulièrement sur la distribution des espèces et la structure des assemblages. Des données fournies par l'Office National de l'Eau et des Milieux Aquatiques ainsi qu'une approche de modélisation basée sur les niches écologiques des espèces (i.e., modèles de distribution) ont été utilisées. Différentes sources d'incertitude ont également été testées dans une approche d'ensembles afin de prendre en compte la variabilité entre les impacts projetés et fournir ainsi une évaluation robuste de ces impacts. La première partie de ce travail a consisté en l'identification des principaux déterminants environnementaux qui structurent la répartition spatiale des espèces de poisson au sein des réseaux hydrographiques. Globalement, il apparaît qu'une combinaison de facteurs climatiques et de variables décrivant l'habitat local et la position des habitats au sein des réseaux hydrographiques est importante pour expliquer la distribution actuelle des espèces. De plus, les espèces ont toutes des réponses différentes aux facteurs de l'environnement. Dans un second temps, nous avons mis en évidence que le choix de la méthode statistique de modélisation de la niche écologique est crucial, les patrons actuels et futurs de distribution prédits étant fortement contrastés selon la méthode de modélisation considérée. Cette dernière s'avère même être la principale source d'incertitude dans les projections futures, bien plus encore que les modèles climatiques de circulation générale et les scénarios d'émission de gaz à effet de serre. La variabilité entre les prédictions issues de plusieurs techniques de modélisation peut être prise en compte par une approche de consensus. Un modèle consensuel basé sur la valeur moyenne de l'ensemble de prédictions est capable de prédire correctement la distribution actuelle des espèces et la composition des assemblages. Nous avons donc choisi de retenir cette approche pour évaluer au mieux les impacts potentiels du changement climatique sur les poissons des rivières françaises à la fin du 21ème siècle. Nous avons montré que la majorité des espèces de poisson pourrait être affectée par les futures modifications du climat. Seules quelques espèces d'eau froide (e.g. truite fario, chabot) pourraient restreindre leur distribution aux parties les plus apicales des réseaux hydrographiques. Au contraire, les espèces tolérant des températures plus élevées pourraient coloniser de nouveaux habitats et étendre ainsi leur répartition. Ces modifications de la distribution des espèces pourraient conduire à un réarrangement des assemblages au niveau taxonomique et fonctionnel. Une augmentation de la diversité locale et de la similarité régionale (i.e., homogénéisation) sont ainsi prédites simultanément. L'ensemble de ces résultats apporte donc des éléments sur la compréhension de la distribution des poissons d'eau douce et sur les conséquences du changement climatique qui peuvent être envisagées. Ce travail fournit ainsi une base aux acteurs de la gestion de la biodiversité afin d'initier des mesures de conservation concrètes. De plus, les considérations méthodologiques développées dans cette thèse sont une contribution importante à l'amélioration des projections issues de modèles statistiques de distribution et à la quantification de leur incertitude.Climate change and its impact on biodiversity are receiving increasing attention from scientists and people managing natural ecosystems. Indeed, climate has a major influence on the biology and ecology of fauna and flora, from physiology to distribution. Climate change may thus have major consequences on species and assemblages. Among freshwater ecosystems, stream fish have no physiological ability to regulate their body temperature and they have to cope with streams' hydrological variability and strong anthropogenic pressures. Yet their response to current and future climate change has been poorly studied. The aim of this PhD thesis is to assess the potential impact of climate change on fish in French streams, mainly on species distribution and assemblages' structure. Data provided by the Office National de l'Eau et des Milieux Aquatiques combined with a modelling approach based on species' ecological niche (i.e., distribution models) have been used. Several sources of uncertainty have also been considered in an ensemble modeling framework in order to account for the variability between projected impacts and to provide reliable estimates of such impact. First, we have identified the main environmental factors that determine the spatial distribution of fish species within river networks. Overall, it appears that a combination of both climatic variables and variables describing the local habitat and its position within the river network is important to explain the current species distribution. Moreover, each fish species responded differently to the environmental factors. Second, we have highlighted that the choice of the statistical method used to model the fish ecological niche is crucial given that the current and future patterns of distribution predicted by different statistical methods vary significantly. The statistical method appears to be the main source of uncertainty, resulting in more variability in projections than the global circulation models and greenhouse gas emission scenarios. The variability between predictions from several statistical methods can be taken into account by a consensus approach. Consensual predictions based on the computation of the average of the whole predictions ensemble have achieved accurate predictions of the current species distribution and assemblages' composition. We have therefore selected this approach to assess the potential impacts of climate change on fish in French streams at the end of the 21st century with the highest degree of confidence. We have found that most fish species could be sensitive to the future climate modifications. Only a few cold-water species (i.e., brown trout, bullhead) could restrict their distribution to the most upstream parts of river networks. On the contrary, cool- and warm-water fish species could colonize many newly suitable habitats and expand strongly their distribution. These changes of species distribution could lead to a rearrangement of fish assemblages both at the taxonomic and functional levels. An increase in local diversity together with an increase in regional similarity (i.e., homogenization) are therefore expected. All these results bring new insights for the understanding of stream fish species distribution and expected consequences of climate change. This work thus provides biodiversity managers and conservationists with a basis to take efficient preservation measures.
