In search of virus carriers of the 1988 and 2002 phocine distemper virus outbreaks in European harbour seals
European harbour seal (Phoca vitulina) populations decreased substantially during the phocine distemper virus (PDV) outbreaks of 1988 and 2002. Different hypotheses have stated that various seals and terrestrial carnivore species might be the source of infection. To further analyse these hypotheses, grey (Halichoerus grypus) and ringed (Phoca hispida) seals, polar bears (Ursus maritimus) and minks (Mustela lutreola) were sampled from the North Sea and East Greenland coasts between 1988 and 2004 and investigated by RT-PCR using a panmorbillivirus primer pair. Curcumin acts as anti-tumorigenic and hormone-suppressive agent in murine and human pituitary tumour cells in vitro and in vivo
Curcumin (diferuloylmethane) is the active ingredient of the spice plant Curcuma longa and has been shown to act anti-tumorigenic in different types of tumours. Therefore, we have studied its effect in pituitary tumour cell lines and adenomas. Proliferation of lactosomatotroph GH3 and somatotroph MtT/S rat pituitary cells as well as of corticotroph AtT20 mouse pituitary cells was inhibited by curcumin in monolayer cell culture and in colony formation assay in soft agar. Fluorescence-activated cell sorting (FACS) analysis demonstrated curcumin-induced cell cycle arrest at G2/M. Analysis of cell cycle proteins by immunoblotting showed reduction in cyclin D1, cyclin-dependent kinase 4 and no change in p27kip. FACS analysis with Annexin V-FITC/7-aminoactinomycin D staining demonstrated curcumin-induced early apoptosis after 3, 6, 12 and 24 h treatment and nearly no necrosis. Induction of DNA fragmentation, reduction of Bcl-2 and enhancement of cleaved caspase-3 further confirmed induction of apoptosis by curcumin. Growth of GH3 tumours in athymic nude mice was suppressed by curcumin in vivo. In endocrine pituitary tumour cell lines, GH, ACTH and prolactin production were inhibited by curcumin. Studies in 25 human pituitary adenoma cell cultures have confirmed the antitumorigenic and hormone-suppressive effects of curcumin. Altogether, the results described in this report suggest this natural compound as a good candidate for therapeutic use on pituitary tumours. © 2009 Society for Endocrinology Printed in Great Britain
Heterogeneously Catalyzed Continuous-Flow Hydrogenation Using Segmented Flow in Capillary Columns
Segmented flow in standard GC capillary columns, with a heterogeneous Pd catalyst on the walls, gave rapid information about catalytic processes in them. The residence time and conversion was monitored visually, greatly simplifying bench-scale optimization. Examples show the benefits of the elimination of pore diffusion and axial dispersion. Dilution of the magnetic lattice in the Kitaev candidate -RuCl by Rh doping
Magnetic dilution of a well-established Kitaev candidate system is realized
in the substitutional RuRhCl series (). Optimized
syntheses protocols yield uniformly-doped single crystals and polycrystalline
powders that are isostructural to the parental -RuCl as per X-ray
diffraction. The Rh content is accurately determined by the quantitative
energy-dispersive X-ray spectroscopy technique with standards. We determine the
magnetic phase diagram of RuRhCl for in-plane magnetic fields
from magnetization and specific-heat measurements as a function of and
stacking periodicity, and identify the suppression of the magnetic order at towards a disordered phase, which does not show any clear
signature of freezing into a spin glass. Comparing with previous studies on the
substitution series RuIrCl, we propose that chemical pressure
would contribute to the suppression of magnetic order especially in
RuIrCl and that the zigzag magnetic ground state appears to be
relatively robust with respect to the dilution of the
Kitaev----Heisenberg magnetic lattice. We also discovered a slight
dependence of the magnetic properties on thermal cycling, which would be due to
an incomplete structural transition
Use of a porous membrane for gas bubble removal in microfluidic channels: physical mechanisms and design criteria
We demonstrate and explain a simple and efficient way to remove gas bubbles
from liquid-filled microchannels, by integrating a hydrophobic porous membrane
on top of the microchannel. A prototype chip is manufactured in hard,
transparent polymer with the ability to completely filter gas plugs out of a
segmented flow at rates up to 7.4 microliter/s per mm2 of membrane area. The
device involves a bubble generation section and a gas removal section. In the
bubble generation section, a T-junction is used to generate a train of gas
plugs into a water stream. These gas plugs are then transported towards the gas
removal section, where they slide along a hydrophobic membrane until complete
removal. The system has been successfully modeled and four necessary operating
criteria have been determined to achieve a complete separation of the gas from
the liquid. The first criterion is that the bubble length needs to be larger
than the channel diameter. The second criterion is that the gas plug should
stay on the membrane for a time sufficient to transport all the gas through the
membrane. The third criterion is that the gas plug travel speed should be lower
than a critical value: otherwise a stable liquid film between the bubble and
the membrane prevents mass transfer. The fourth criterion is that the pressure
difference across the membrane should not be larger than the Laplace pressure
to prevent water from leaking through the membrane
Leishmania isoenzyme polymorphisms in Ecuador: Relationships with geographic distribution and clinical presentation
Background: Determinants of the clinical presentation of the leishmaniases are poorly understood but Leishmania species and strain differences are important. To examine the relationship between clinical presentation, species and isoenzyme polymorphisms, 56 Leishmania isolates from distinct presentations of American tegumentary leishmaniasis (ATL) from Ecuador were analyzed.
Methods: Isolates were characterized by multilocus enzyme electrophoresis for polymorphisms of 11 isoenzymes. Patients were infected in four different ecologic regions: highland and lowland jungle of the Pacific coast, Amazonian lowlands and Andean highlands.
Results: Six Leishmania species constituting 21 zymodemes were identified: L. (Viannia) panamensis (21 isolates,
7 zymodemes), L. (V.) guyanensis (7 isolates, 4 zymodemes), L. (V.) braziliensis (5 isolates, 3 zymodemes), L.
(Leishmania) mexicana (11 isolates, 4 zymodemes), L. (L.) amazonensis (10 isolates, 2 zymodemes) and L. (L.) major
(2 isolates, 1 zymodeme). L. panamensis was the species most frequently identified in the Pacific region and was
associated with several clinical variants of cutaneous disease (CL); eight cases of leishmaniasis recidiva cutis (LRC) found in the Pacific highlands were associated with 3 zymodemes of this species. Mucocutaneous leishmaniasis
found only in the Amazonian focus was associated with 3 zymodemes of L. braziliensis. The papular variant of CL,
Uta, found in the Andean highlands was related predominantly with a single zymodeme of L. mexicana.
Conclusion: Our data show a high degree of phenotypic variation within species, and some evidence for associations between specific variants of ATL (i.e. Uta and LRC) and specific Leishmania zymodemes. This study
further defines the geographic distribution of Leishmania species and clinical variants of ATL in Ecuador
Dynamics of droplet formation at T-shaped nozzles with elastic feed lines
We describe the formation of water in oil droplets, which are commonly used in lab-on-a-chip systems for sample generation and dosing, at microfluidic T-shaped nozzles from elastic feed lines. A narrow nozzle forms a barrier for a liquid-liquid interface, such that pressure can build up behind the nozzle up to a critical pressure. Above this critical pressure, the liquid bursts into the main channel. Build-up of pressure is possible when the fluid before the nozzle is compressible or when the channel that leads to the nozzle is elastic. We explore the value of the critical pressure and the time required to achieve it. We describe the fluid flow of the sudden burst, globally in terms of flow rate into the channel and spatially resolved in terms of flow fields measured using micro-PIV. A total of three different stages-the lag phase, a spill out phase, and a linear growth phase-can be clearly discriminated during droplet formation. The lag time linearly scales with the curvature of the interface inside the nozzle and is inversly proportional to the flow rate of the dispersed phase. Detailed statistical analysis plan for the SafeBoosC III trial : a multinational randomised clinical trial assessing treatment guided by cerebral oxygenation monitoring versus treatment as usual in extremely preterm infants
Background: Infants born extremely preterm are at high risk of dying or suffering from severe brain injuries. Treatment guided by monitoring of cerebral oxygenation may reduce the risk of death and neurologic complications. The SafeBoosC III trial evaluates the effects of treatment guided by cerebral oxygenation monitoring versus treatment as usual. This article describes the detailed statistical analysis plan for the main publication, with the aim to prevent outcome reporting bias and data-driven analyses. Methods/design: The SafeBoosC III trial is an investigator-initiated, randomised, multinational, pragmatic phase III trial with a parallel group structure, designed to investigate the benefits and harms of treatment based on cerebral near-infrared spectroscopy monitoring compared with treatment as usual. Randomisation will be 1:1 stratified for neonatal intensive care unit and gestational age (lower gestational age (< 26 weeks) compared to higher gestational age ( 65 26 weeks)). The primary outcome is a composite of death or severe brain injury at 36 weeks postmenstrual age. Primary analysis will be made on the intention-to-treat population for all outcomes, using mixed-model logistic regression adjusting for stratification variables. In the primary analysis, the twin intra-class correlation coefficient will not be considered. However, we will perform sensitivity analyses to address this. Our simulation study suggests that the inclusion of multiple births is unlikely to significantly affect our assessment of intervention effects, and therefore we have chosen the analysis where the twin intra-class correlation coefficient will not be considered as the primary analysis. Discussion: In line with the Declaration of Helsinki and the International Conference on Harmonization Good Clinical Practice guidelines, we have developed and published this statistical analysis plan for the SafeBoosC III trial, prior to any data analysis. Cerebral near-infrared spectroscopy monitoring versus treatment as usual for extremely preterm infants : a protocol for the SafeBoosC randomised clinical phase III trial
Background: Cerebral oxygenation monitoring may reduce the risk of death and neurologic complications in extremely preterm infants, but no such effects have yet been demonstrated in preterm infants in sufficiently powered randomised clinical trials. The objective of the SafeBoosC III trial is to investigate the benefits and harms of treatment based on near-infrared spectroscopy (NIRS) monitoring compared with treatment as usual for extremely preterm infants. Methods/design: SafeBoosC III is an investigator-initiated, multinational, randomised, pragmatic phase III clinical trial. Inclusion criteria will be infants born below 28 weeks postmenstrual age and parental informed consent (unless the site is using 'opt-out' or deferred consent). Exclusion criteria will be no parental informed consent (or if 'opt-out' is used, lack of a record that clinical staff have explained the trial and the 'opt-out' consent process to parents and/or a record of the parents' decision to opt-out in the infant's clinical file); decision not to provide full life support; and no possibility to initiate cerebral NIRS oximetry within 6 h after birth. Participants will be randomised 1:1 into either the experimental or control group. Participants in the experimental group will be monitored during the first 72 h of life with a cerebral NIRS oximeter. Cerebral hypoxia will be treated according to an evidence-based treatment guideline. Participants in the control group will not undergo cerebral oxygenation monitoring and will receive treatment as usual. Each participant will be followed up at 36 weeks postmenstrual age. The primary outcome will be a composite of either death or severe brain injury detected on any of the serial cranial ultrasound scans that are routinely performed in these infants up to 36 weeks postmenstrual age. Severe brain injury will be assessed by a person blinded to group allocation. To detect a 22% relative risk difference between the experimental and control group, we intend to randomise a cohort of 1600 infants. Discussion: Treatment guided by cerebral NIRS oximetry has the potential to decrease the risk of death or survival with severe brain injury in preterm infants. There is an urgent need to assess the clinical effects of NIRS monitoring among preterm neonates. Trial registration: ClinicalTrial.gov, NCT03770741. Registered 10 December 2018
