Effect of Polyethylene Glycol 3350 on the Handling Properties of Low Salt Wheat Dough Formulations
The effect of polyethylene glycol (PEG) 3350 addition (3%, flour wt. basis) on the properties of dough made from two Canadian Western Red Spring wheat cultivars (Triticum aestivum L. ‘Harvest’ and ‘Pembina’) differing in dough mixing requirements and dough-handling properties was investigated in a low salt dough formulation (1% NaCl, flour wt. basis). PEG was added for experimental purposes to alter water mobility to better understand underlining mechanisms, however would not be used in real bread formulations. For cultivar Harvest, but not Pembina, dough stickiness was reduced by the addition of PEG. Dough freezable water content decreased with the addition of PEG for both cultivars. Rheological measurements showed that PEG increased dough stiffness as measured by the complex modulus |G*|. Creep measurements indicated that the relative elastic component (Jel) increased whereas maximum deformation (Jmax) decreased with the addition of PEG for cultivar Harvest only. Dough made with a weaker cultivar (Harvest) with the addition of PEG performed similarly to dough made with a stronger cultivar (Pembina) without PEG. Results indicate that in a low sodium environment, availability of water is critically important for controlling a number of properties that relate closely to dough machinability, especially in a weaker wheat cultivar
Genetic mapping of leaf rust (Puccinia triticina Eriks) resistance genes in six Canadian spring wheat cultivars
The Canada Western Red Spring wheat (Triticum aestivum L.) cultivars AAC Concord, AAC Prevail, CDC Hughes, Lillian, Glenlea, and elite line BW961 express a spectrum of resistance to leaf rust caused by Puccinia triticina Eriks. This study aimed to identify and map the leaf rust resistance of the cultivars using three doubled haploid populations, AAC Prevail/BW961 (PB), CDC Hughes/AAC Concord (HC), and Lillian/Glenlea (LG). The populations were evaluated for seedling resistance in the greenhouse and adult plant disease response in the field at Morden, MB for 3 years and genotyped with the 90K wheat Infinium iSelect SNP array. Genetic maps were constructed to perform QTL analysis on the seedling and field leaf rust data. A total of three field leaf rust resistance QTL segregated in the PB population, five in the HC, and six in the LG population. In the PB population, BW961 contributed two QTL on chromosomes 2DS and 7DS, and AAC Prevail contributed a QTL on 4AL consistent across trials. Of the five QTL in HC, AAC Concord contributed two QTL on 4AL and 7AL consistent across trials and a QTL on 3DL.1 that provided seedling resistance only. CDC Hughes contributed two QTL on 1DS and 3DL.2. Lillian contributed four QTL significant in at least two of the three trials on 2BS, 4AL, 5AL, and 7AL, and Glenlea two QTL on 4BL and 7BL. The 1DS QTL from CDC Hughes, the 2DS from BW961, the 4AL from the AAC Prevail, AAC Concord, and Lillian, and the 7AL from AAC Concord and Lillian conferred seedling leaf rust resistance. The QTL on 4AL corresponded with Lr30 and was the same across cultivars AAC Prevail, AAC Concord, and Lillian, whereas the 7AL corresponding with LrCen was coincident between AAC Concord and Lillian. The 7DS and 2DS QTL in BW961 corresponded with Lr34 and Lr2a, respectively, and the 1DS QTL in CDC Hughes with Lr21. The QTL identified on 5AL could represent a novel gene. The results of this study will widen our knowledge of leaf rust resistance genes in Canadian wheat and their utilization in resistance breeding
A haplotype map of allohexaploid wheat reveals distinct patterns of selection on homoeologous genomes
BACKGROUND: Bread wheat is an allopolyploid species with a large, highly repetitive genome. To investigate the impact of selection on variants distributed among homoeologous wheat genomes and to build a foundation for understanding genotype-phenotype relationships, we performed population-scale re-sequencing of a diverse panel of wheat lines. RESULTS: A sample of 62 diverse lines was re-sequenced using the whole exome capture and genotyping-by-sequencing approaches. We describe the allele frequency, functional significance, and chromosomal distribution of 1.57 million single nucleotide polymorphisms and 161,719 small indels. Our results suggest that duplicated homoeologous genes are under purifying selection. We find contrasting patterns of variation and inter-variant associations among wheat genomes; this, in addition to demographic factors, could be explained by differences in the effect of directional selection on duplicated homoeologs. Only a small fraction of the homoeologous regions harboring selected variants overlapped among the wheat genomes in any given wheat line. These selected regions are enriched for loci associated with agronomic traits detected in genome-wide association studies. CONCLUSIONS: Evidence suggests that directional selection in allopolyploids rarely acted on multiple parallel advantageous mutations across homoeologous regions, likely indicating that a fitness benefit could be obtained by a mutation at any one of the homoeologs. Additional advantageous variants in other homoelogs probably either contributed little benefit, or were unavailable in populations subjected to directional selection. We hypothesize that allopolyploidy may have increased the likelihood of beneficial allele recovery by broadening the set of possible selection targets
Effects of Genotype and Growing Year on the Nutritional Composition and Pasting Properties of Glabrous Canary Seed (<i>Phalaris canariensis</i> L.) Groat Flours
Canary seed flour is a new food ingredient that the United States Food and Drug Administration (FDA) and Health Canada recently granted Generally Regarded as Safe (GRAS) status. Stability in nutritional composition and functional properties is an essential characteristic of food ingredients for consistency in nutritional quality and performance in processing. This work assessed the effect of genotypic and environmental variation on the nutritional (protein, starch, amylose, oil, dietary fiber, minerals and fat-soluble vitamins) and pasting (as measured in viscosity (peak, trough, breakdown, final, and setback), peak time, and pasting temperatures) properties of Canary seed. The samples included four Canary seed varieties grown in randomized complete block design experiments at one location for two growing seasons. In general, the nutritional composition of Canary seed flour was not affected by genotype, growing year, and their interaction except for starch content, which was significantly affected by the growing year (p p p < 0.001) affected by both genotypic and growing year variation but not their interaction. Our results suggest that the food industry should measure starch and iron content prior to processing to ensure consistency in nutritional labeling. Also, for those applications where starch pasting properties are essential, the manufacturer should consider measuring the RVA pasting viscosities for every batch of raw material. The results have provided the baseline knowledge of which nutritional or functional properties of Canary seed flour can be improved through breeding and agronomy programs to ensure the reliability of Canary seed as an ingredient
In vitro assessment of the starch digestibility of western Canadian wheat market classes and cultivars
The objective of the study was to measure the effect of wheat market class and cultivar on starch digestibility using an in vitro model that mimics chicken digestive tract and relate it to grain characteristics. The study evaluated 18 wheat cultivars from eight western Canadian wheat classes and, each cultivar was replicated four times. Samples were subjected to gastric and small intestine (SI) digestion phases and each sample was assayed in triplicate; glucose release was measured in SI phase. Starch granule distribution, amylose, total starch, crude protein (CP), ash, and non-starch polysaccharides (NSP) were analyzed in all wheat samples. Small intestinal phase times of 15, 60 and 120 min were chosen to approximate digestion in the terminal duodenum, jejunum and ileum. Starch digestibility of wheat classes ranged as follows: 15 min â 33.1 to 49.1%, 60 min â 80.2 to 93.3% and 120 min â 92.4 to 97.6%. Starch digestibility positively correlated with CP, ash, NSP and % large granules whereas, it negatively correlated with total starch, and proportion of small and medium granules. In conclusion, market class and cultivar of western Canadian wheat affects both rate and extent of starch digestibility and it is related to various grain characteristics.The accepted manuscript in pdf format is listed with the files at the bottom of this page. The Soil Microbial Community and Grain Micronutrient Concentration of Historical and Modern Hard Red Spring Wheat Cultivars Grown Organically and Conventionally in the Black Soil Zone of the Canadian Prairies
Micronutrient deficiencies in the diet of many people are common and wheat is a staple food crop, providing a carbohydrate and micronutrient source to a large percentage of the world’s population. We conducted a field study to compare five Canadian red spring wheat cultivars (released over the last century) grown under organic and conventional management systems for yield, grain micronutrient concentration, and soil phospholipid fatty acid (PLFA) profile. The organic system had higher grain Zn, Fe, Mg and K levels, but lower Se and Cu levels. There was no trend in the results to suggest that modern western Canadian hard red spring cultivars have lower grain micronutrient content than historical cultivars. Wheat cultivar choice is important for maximizing grain nutrient levels, which was influenced by management system. It is evident that the emphasis on elevated grain quality in the western Canadian hard red spring class has resulted in the retention of micronutrient quality characters. Three fungal PLFAs were indicators for the organic system, and all three of these indicators were positively correlated with grain Cu concentration. In the organic system, percent arbuscular mycorrhizal fungi were negatively correlated with grain Zn and Fe concentrations, and positively correlated with grain Mn, Cu, K concentrations and grain yield. The organic system had higher levels of fungi in the soil, including arbuscular mycorrhizal fungi. Organic management practices appear to result in elevated levels of grain micronutrient concentration. The hard red spring breeding effort in and for the black soil zone of the northern Great Plains also appears to have led to no diminishment of grain micronutrient concentration. It is evident that both the agronomic system and breeding strategies in this region can be exploited for future increases in grain micronutrient concentration
