    MagicVideo: Efficient Video Generation With Latent Diffusion Models

    We present an efficient text-to-video generation framework based on latent diffusion models, termed MagicVideo. Given a text description, MagicVideo can generate photo-realistic video clips with high relevance to the text content. With the proposed efficient latent 3D U-Net design, MagicVideo can generate video clips with 256x256 spatial resolution on a single GPU card, which is 64x faster than the recent video diffusion model (VDM). Unlike previous works that train video generation from scratch in the RGB space, we propose to generate video clips in a low-dimensional latent space. We further utilize all the convolution operator weights of pre-trained text-to-image generative U-Net models for faster training. To achieve this, we introduce two new designs to adapt the U-Net decoder to video data: a framewise lightweight adaptor for the image-to-video distribution adjustment and a directed temporal attention module to capture frame temporal dependencies. The whole generation process is within the low-dimension latent space of a pre-trained variation auto-encoder. We demonstrate that MagicVideo can generate both realistic video content and imaginary content in a photo-realistic style with a trade-off in terms of quality and computational cost.     Trends in sperm quality by computer-assisted sperm analysis of 49,189 men during 2015–2021 in a fertility center from China

    BackgroundSperm quality, including semen volume, sperm count, concentration, and total and progressive motility (collectively, “semen parameters”), has declined in the recent decades. Computer-assisted sperm analysis (CASA) provides sperm kinematic parameters, and the temporal trends of which remain unclear. Our objective is to examine the temporal trend of both semen parameters and kinematic parameters in Shanghai, China, in the recent years.MethodsThis retrospective study analyzed semen parameters and kinematic parameters of 49,819 men attending our reproductive center by using CASA during 2015–2021. The total sample was divided into two groups: samples that surpassed the WHO guideline (2010) low reference limits (“above reference limit” group, ARL; n = 24,575) and samples that did not (“below reference limit” group, BRL; n = 24,614). One-way analysis of variance, Kruskal–Wallis test, independent samples t-test, and covariance analysis were used to assess the differences among groups. Year, age, and abstinence time were included in the multiple linear regression model of the ARL group to adjust the confounders and depict the trends in sperm quality.ResultsAmong all the total sample and the ARL and BRL groups, the age of subjects increased in recent years. Semen volume and sperm count showed declined tendency with years in the total sample, the ARL and BRL groups, and the subgroup of age or abstinence time, whereas sperm velocities showed increased tendency with years on the contrary. The multiple linear regression model of the ARL group, adjusting for age and abstinence time, confirmed these trends. Semen volume (β1= −0.162; CI: −0.172, −0.152), sperm count (β1= −9.97; CI: −10.813, −9.128), sperm concentration (β1 = −0.535; CI: −0.772, −0.299), motility (β1 = −1.751; CI: −1.830, −1.672), and progressive motility (β1 = −1.12; CI: −0.201, −0.145) decreased with year, whereas curvilinear line velocity (VCL) (β1 = 3.058; CI: 2.912, 3.203), straight line velocity (VSL) (β1 = 2.075; CI: 1.990, 2.161), and average path velocity (VAP) (β1 = 2.305; CI: 2.224, 2.386) increased over time (all p < 0.001). In addition, VCL, VSL, and VAP significantly declined with age and abstinence time.ConclusionThe semen parameters declined, whereas the kinematic parameters increased over the recent years. We propose that, although sperm count and motility declined over time, sperm motion velocity increased, suggesting a possible compensatory mechanism of male fertility

    Postoperative ecchymoma of eyelid after botulinum toxin injection for hemifacial spasm: a case report

    Hemifacial spasm (HFS) is a rare movement disorder characterized by involuntary muscle contractions on one side of the face. Compared to the high therapeutic effect, adverse effects of botulinum toxin treatment for HFS occurred rarely. However, managing HFS patients who are also taking antithrombotic drugs poses a challenge. Here, we present a case of postoperative ecchymoma of the eyelid following a botulinum toxin injection in a patient receiving daily vinpocetine and aspirin antiplatelet therapy. This case highlights the importance of considering the potential risks and formulating a treatment plan that maximizes benefit while minimizing complications in HFS patients undergoing botulinum toxin injections and taking antithrombotic medications. To the best of our knowledge, this is the first reported case of postoperative ecchymoma of the eyelid following a botulinum toxin injection. Further research and additional case reports are needed to better understand the management strategies for this patient population

    Investigating the Long-Term Variation Trends of Absorbing Aerosols over Asia by Using Multiple Satellites

    Absorbing aerosols, consisting of smoke (black carbon (BC) and other organics) and dust (from windblown sources), can have a strong warming effect on the climate and impact atmospheric circulation due to localized heating. To investigate the spatiotemporal and vertical changes of absorbing aerosols across Asia, collocation data from OMI, MODIS, and CALIPSO were used to compare two periods: 2006–2013 and 2014–2021. This study revealed a significant temporal and spatial contrast of aerosol loading over the study region, with a drop in total aerosol concentration and anthropogenic smoke concentration recorded across the Eastern China region (all seasons) and a concurrent increase in the Indian sub-continent region (especially in autumn). The range of aerosol diffusion is affected by the height of the smoke and aerosol plumes, as well as the wind force, and is dispersed eastwards because of the Hadley circulation patterns in the Northern Hemisphere. Smoke from Southeast Asia typically rises to a height of 3 km and affects the largest area in contrast to other popular anthropogenic zones, where it is found to be around 1.5–2 km. The dust in Inner Mongolia had the lowest plume height of 2 km (typically in spring) compared to other locations across the study region where it reached 2–5 km in the summer. This study showed, by comparison with AERONET measurements, that combining data from MODIS and OMI generates more accuracy in detecting aerosol AOD from smoke than using the instruments singularly. This study has provided a comprehensive assessment of absorbing aerosol in Asia by utilizing multiplatform remote-sensed data and has summarized long-term changes in the spatiotemporal distribution and vertical structure of absorbing aerosols

    Neuroprotective Effect of Melatonin on Sleep Disorders Associated with Parkinson’s Disease

    Parkinson’s disease (PD) is a complex, multisystem disorder with both neurologic and systemic manifestations, which is usually associated with non-motor symptoms, including sleep disorders. Such associated sleep disorders are commonly observed as REM sleep behavior disorder, insomnia, sleep-related breathing disorders, excessive daytime sleepiness, restless legs syndrome and periodic limb movements. Melatonin has a wide range of regulatory effects, such as synchronizing circadian rhythm, and is expected to be a potential new circadian treatment of sleep disorders in PD patients. In fact, ongoing clinical trials with melatonin in PD highlight melatonin’s therapeutic effects in this disease. Mechanistically, melatonin plays its antioxidant, anti-inflammatory, anti-excitotoxity, anti-synaptic dysfunction and anti-apoptotic activities. In addition, melatonin attenuates the effects of genetic variation in the clock genes of Baml1 and Per1 to restore the circadian rhythm. Together, melatonin exerts various therapeutic effects in PD but their specific mechanisms require further investigations

    SARS-CoV-2 versus Influenza A Virus: Characteristics and Co-Treatments

    For three years, the novel coronavirus disease 2019 (COVID-19) pandemic, caused by infection of the SARS-CoV-2 virus, has completely changed our lifestyles and prepared us to live with this novel pneumonia for years to come. Given that pre-existing flu is caused by the influenza A virus, we have begun unprecedently co-coping with two different respiratory diseases at the same time. Hence, we draw a comparison between SARS-CoV-2 and influenza A virus based on the general characteristics, especially the main variants’ history and the distribution of the two viruses. SARS-CoV-2 appeared to mutate more frequently and independently of locations than the influenza A virus. Furthermore, we reviewed present clinical trials on combined management against COVID-19 and influenza in order to explore better solutions against both at the same time

    Diffusion Tensor Imaging Radiomics for Diagnosis of Parkinson’s Disease

    Background: Diagnosis of Parkinson’s Disease (PD) based on clinical symptoms and scale scores is mostly objective, and the accuracy of neuroimaging for PD diagnosis remains controversial. This study aims to introduce a radiomic tool to improve the sensitivity and specificity of diagnosis based on Diffusion Tensor Imaging (DTI) metrics. Methods: In this machine learning-based retrospective study, we collected basic clinical information and DTI images from 54 healthy controls (HCs) and 56 PD patients. Among them, 60 subjects (30 PD patients and 30 HCs) were assigned to the training group, whereas the test cohort was 26 PD patients and 24 HCs. After the feature extraction and selection using newly developed image processing software Ray-plus, LASSO regression was used to finalize radiomic features. Results: A total of 4600 radiomic features were extracted, of which 12 were finally selected. The values of the AUC (area under the subject operating curve) in the training group, the validation group, and overall were 0.911, 0.931, and 0.919, respectively. Conclusion: This study introduced a novel radiometric and computer algorithm based on DTI images, which can help increase the sensitivity and specificity of PD screening

    Overexpression of human-derived DNMT3A induced intergenerational inheritance of DNA methylation and gene expression variations in rat brain and testis

    In mammals, DNA methylation patterns are established by various types of DNA methyltransferases and can be stably passed on during cell division, thus creating a paradigm for epigenetic regulation that can mediate long-lasting changes in gene expression even when the initial triggering signal has disappeared. Although functional deficiency of DNMT3A, one of the methyltransferases, leads to abnormal DNA methylation patterns that result in developmental deficits in mammals, the impacts of its overexpression on tissue gene expression and DNA methylation patterns remain unclear. Here, our previously established hDNMT3A transgenic rat model and mRNA sequencing and bisulphite sequencing PCR were used to analyse the impact of hDNMT3A overexpression on tissue transcriptome and methylome, and whether the impact could be inherited intergenerationally was subsequently investigated. Our results revealed that the overexpression of hDNMT3A could induce notable gene expression variations in rat testis and brain. More importantly, 36.02% and 38.89% of these variations could be intergenerationally inherited to offspring without the transmission of the initial endogenic trigger in the brain and testis, respectively. Furthermore, we found that intergenerationally inherited DNA methylation variations in their promoters and exons could be the underlying mechanism. Compared with inheritable variations that were passively induced by environmental factors, these variations were actively induced by endogenous epigenetic modifiers. This study provided evidence for the epigenetic inheritance of endogenous factors that actively induce gene expression and DNA methylation variations; however, more studies are needed to determine the number of generations that these variations can be stably inherited

    Overexpression of Human-Derived DNMT3A Induced Intergenerational Inheritance of Active DNA Methylation Changes in Rat Sperm

    DNA methylation is the major focus of studies on paternal epigenetic inheritance in mammals, but most previous studies about inheritable DNA methylation changes are passively induced by environmental factors. However, it is unclear whether the active changes mediated by variations in DNA methyltransferase activity are heritable. Here, we established human-derived DNMT3A (hDNMT3A) transgenic rats to study the effect of hDNMT3A overexpression on the DNA methylation pattern of rat sperm and to investigate whether this actively altered DNA methylation status is inheritable. Our results revealed that hDNMT3A was overexpressed in the testis of transgenic rats and induced genome-wide alterations in the DNA methylation pattern of rat sperm. Among 5438 reliable loci identified with 64 primer-pair combinations using a methylation-sensitive amplification polymorphism method, 28.01% showed altered amplified band types. Among these amplicons altered loci, 68.42% showed an altered DNA methylation status in the offspring of transgenic rats compared with wild-type rats. Further analysis based on loci which had identical DNA methylation status in all three biological replicates revealed that overexpression of hDNMT3A in paternal testis induced hypermethylation in sperm of both genotype-negative and genotype-positive offspring. Among the differentially methylated loci, 34.26% occurred in both positive and negative offspring of transgenic rats, indicating intergenerational inheritance of active DNA methylation changes in the absence of hDNM3A transmission. Furthermore, 75.07% of the inheritable loci were hyper-methylated while the remaining were hypomethylated. Distribution analysis revealed that the DNA methylation variations mainly occurred in introns and intergenic regions. Functional analysis revealed that genes related to differentially methylated loci were involved in a wide range of functions. Finally, this study demonstrated that active DNA methylation changes induced by hDNMT3A expression were intergenerationally inherited by offspring without transmission of the transgene, which provided evidence for the transmission of active endogenous-factors-induced epigenetic variations
