Modelling the Dynamics of Feral Alfalfa Populations and Its Management Implications
BACKGROUND: Feral populations of cultivated crops can pose challenges to novel trait confinement within agricultural landscapes. Simulation models can be helpful in investigating the underlying dynamics of feral populations and determining suitable management options. METHODOLOGY/PRINCIPAL FINDINGS: We developed a stage-structured matrix population model for roadside feral alfalfa populations occurring in southern Manitoba, Canada. The model accounted for the existence of density-dependence and recruitment subsidy in feral populations. We used the model to investigate the long-term dynamics of feral alfalfa populations, and to evaluate the effectiveness of simulated management strategies such as herbicide application and mowing in controlling feral alfalfa. Results suggest that alfalfa populations occurring in roadside habitats can be persistent and less likely to go extinct under current roadverge management scenarios. Management attempts focused on controlling adult plants alone can be counterproductive due to the presence of density-dependent effects. Targeted herbicide application, which can achieve complete control of seedlings, rosettes and established plants, will be an effective strategy, but the seedbank population may contribute to new recruits. In regions where roadside mowing is regularly practiced, devising a timely mowing strategy (early- to mid-August for southern Manitoba), one that can totally prevent seed production, will be a feasible option for managing feral alfalfa populations. CONCLUSIONS/SIGNIFICANCE: Feral alfalfa populations can be persistent in roadside habitats. Timely mowing or regular targeted herbicide application will be effective in managing feral alfalfa populations and limit feral-population-mediated gene flow in alfalfa. However, in the context of novel trait confinement, the extent to which feral alfalfa populations need to be managed will be dictated by the tolerance levels established by specific production systems for specific traits. The modelling framework outlined in this paper could be applied to other perennial herbaceous plants with similar life-history characteristics
Modulation of embryo-forming capacity in culture through the expression of Brassica genes involved in the regulation of the shoot apical meristem
Somatic embryogenesis in Arabidopsis is achieved by culturing bending-cotyledon embryos on a 2,4-D-containing induction medium for 14âd followed by a transfer on to a hormone-free development medium. Several genes orthologous to Arabidopsis SHOOTMERISTEMLESS (STM), CLAVATA 1 (CLV1), and ZWILLE (ZLL) were isolated from Brassica oleracea (Bo), B. rapa (Br), and B. napus (Bn), and ectopically expressed in Arabidopsis to assess their effects on somatic embryogenesis. Ectopic expression of BoSTM, BrSTM, and BnSTM increased the number of somatic embryos, whereas a different effect was observed in lines overexpressing BnCLV1 in which somatic embryo formation was severely repressed. The introduction of BnZLL did not have any effects on Arabidopsis somatic embryogenesis. The increased embryo-forming capacity observed in lines overexpressing Brassica STM was associated with a lower requirement for the inductive signal 2,4-D, and a higher expression of WUSCHEL (WUS) which demarcates the formation of embryogenic cells. This was in contrast to the 35S::BnCLV1 lines which showed the highest requirement for exogenous 2,4-D and a reduced WUS expression. Microarray studies were conducted to monitor global changes in transcript levels during Arabidopsis somatic embryogenesis between the wild-type (WT) line and a BoSTM-overexpressing line, which showed the most pronounced enhancement of somatic embryo yield. The introduction of BoSTM affected the expression of many genes involved in hormone perception and signalling, as well as genes encoding DNA methyltransferases and enzymes of glutathione metabolism. Pharmacological experiments performed to confirm some of the microarray results showed that Arabidopsis somatic embryogenesis is encouraged by a global hypomethylation of the DNA during the induction phase and by a switch of the glutathione pool towards an oxidized state during the subsequent development phase. Both events occurred in the 35S::BoSTM line, but not in the WT line. Altered expression of Brassica STM also had profound effects on B. napus microspore-derived embryogenesis. The yield of microspore-derived embryos increased in lines overexpressing BnSTM and significantly decreased in antisense lines down-regulating BnSTM
Low-temperature plasma treatment induces DNA damage leading to necrotic cell death in primary prostate epithelial cells
Background:In recent years, the rapidly advancing field of low-temperature atmospheric pressure plasmas has shown considerable promise for future translational biomedical applications, including cancer therapy, through the generation of reactive oxygen and nitrogen species.Method:The cytopathic effect of low-temperature plasma was first verified in two commonly used prostate cell lines: BPH-1 and PC-3 cells. The study was then extended to analyse the effects in paired normal and tumour (Gleason grade 7) prostate epithelial cells cultured directly from patient tissue. Hydrogen peroxide (H2O2) and staurosporine were used as controls throughout.Results:Low-temperature plasma (LTP) exposure resulted in high levels of DNA damage, a reduction in cell viability, and colony-forming ability. H2O2 formed in the culture medium was a likely facilitator of these effects. Necrosis and autophagy were recorded in primary cells, whereas cell lines exhibited apoptosis and necrosis.Conclusions:This study demonstrates that LTP treatment causes cytotoxic insult in primary prostate cells, leading to rapid necrotic cell death. It also highlights the need to study primary cultures in order to gain more realistic insight into patient response

The BRCA1-Î11q Alternative Splice Isoform Bypasses Germline Mutations and Promotes Therapeutic Resistance to PARP Inhibition and Cisplatin.
Breast and ovarian cancer patients harboring BRCA1/2 germline mutations have clinically benefitted from therapy with PARP inhibitor (PARPi) or platinum compounds, but acquired resistance limits clinical impact. In this study, we investigated the impact of mutations on BRCA1 isoform expression and therapeutic response. Cancer cell lines and tumors harboring mutations in exon 11 of BRCA1 express a BRCA1-Î11q splice variant lacking the majority of exon 11. The introduction of frameshift mutations to exon 11 resulted in nonsense-mediated mRNA decay of full-length, but not the BRCA1-Î11q isoform. CRISPR/Cas9 gene editing as well as overexpression experiments revealed that the BRCA1-Î11q protein was capable of promoting partial PARPi and cisplatin resistance relative to full-length BRCA1, both in vitro and in vivo Furthermore, spliceosome inhibitors reduced BRCA1-Î11q levels and sensitized cells carrying exon 11 mutations to PARPi treatment. Taken together, our results provided evidence that cancer cells employ a strategy to remove deleterious germline BRCA1 mutations through alternative mRNA splicing, giving rise to isoforms that retain residual activity and contribute to therapeutic resistance. Integrative functional genomic analysis of human brain development and neuropsychiatric risks
INTRODUCTION The brain is responsible for cognition, behavior, and much of what makes us uniquely human. The development of the brain is a highly complex process, and this process is reliant on precise regulation of molecular and cellular events grounded in the spatiotemporal regulation of the transcriptome. Disruption of this regulation can lead to neuropsychiatric disorders. RATIONALE The regulatory, epigenomic, and transcriptomic features of the human brain have not been comprehensively compiled across time, regions, or cell types. Understanding the etiology of neuropsychiatric disorders requires knowledge not just of endpoint differences between healthy and diseased brains but also of the developmental and cellular contexts in which these differences arise. Moreover, an emerging body of research indicates that many aspects of the development and physiology of the human brain are not well recapitulated in model organisms, and therefore it is necessary that neuropsychiatric disorders be understood in the broader context of the developing and adult human brain. RESULTS Here we describe the generation and analysis of a variety of genomic data modalities at the tissue and single-cell levels, including transcriptome, DNA methylation, and histone modifications across multiple brain regions ranging in age from embryonic development through adulthood. We observed a widespread transcriptomic transition beginning during late fetal development and consisting of sharply decreased regional differences. This reduction coincided with increases in the transcriptional signatures of mature neurons and the expression of genes associated with dendrite development, synapse development, and neuronal activity, all of which were temporally synchronous across neocortical areas, as well as myelination and oligodendrocytes, which were asynchronous. Moreover, genes including MEF2C, SATB2, and TCF4, with genetic associations to multiple brain-related traits and disorders, converged in a small number of modules exhibiting spatial or spatiotemporal specificity. CONCLUSION We generated and applied our dataset to document transcriptomic and epigenetic changes across human development and then related those changes to major neuropsychiatric disorders. These data allowed us to identify genes, cell types, gene coexpression modules, and spatiotemporal loci where disease risk might converge, demonstrating the utility of the dataset and providing new insights into human development and disease
Secondary seed dormancy and the seedbank ecology of Brassica napus L. in western Canada
The release of genetically modified, herbicide tolerant canola (Brassica napus L.) genotypes in western Canada has increased interest in the persistence of volunteer canola. B. napus seed may be induced into secondary dormancy in the laboratory, however, little is known of the seedbank ecology and the role of secondary dormancy as a persistence mechanism in this species in the region. The objectives of this research were i) to determine seedbank additions at the time of harvest, ii) determine the role of secondary seed dormancy in seedbank persistence under different management systems, iii) determine the relative importance of factors contributing to secondary dormancy potential and iv) evaluate the role of abscisic acid (ABA) during secondary dormancy induction in B. napus. On farms, average seedbank additions during harvest were approximately 20 times the normal seeding rate of canola. High secondary seed dormancy potential prolonged seedbank persistence in fields, irrespective of tillage system. B. napus exhibited seedling recruitment of a typical summer annual weed where seedling recruitment was only observed in the spring. Seasonal seedling recruitment was the result of two fates: seed death in the shallow seedbank, irrespective of dormancy potential, and increased ungerminability in buried seeds which was related to secondary seed dormancy potential. Among the factors that contribute to secondary seed dormancy potential, genotype was of greatest significance. Seed size was of lesser importance, while the contributions of pre-harvest factors including seed maturity, year, and location were negligible in comparison. Differences in ABA synthesis and the response to ABA application were related to secondary seed dormancy potential and correlated well previous reports linking ABA to seed dormancy. Conclusions that emerge from this research are i) that on some farms, seedbank additions may be lowered by more diligent harvest practices, ii) seedbank persistence of B. napus may be reduced by growing low dormancy genotypes and avoiding seed burial for one year after seedbank establishment and iii) ABA + ABA-glucose ester (ABA-GE) and the ability of seeds to respond to ABA application after seed dormancy induction may potentially be used to identify seed dormancy potential in this species
Wheat and Cereal Rye Inter-Row Living Mulches Interfere with Early Season Weeds in Soybean
Rapid growth of cool-season weeds in the spring exacerbates weed interference during early soybean (Glycine max (L.) Merr.) establishment in northern climates. This study tested the utility of spring-seeded inter-row living mulches in soybean for early season weed suppression using volunteer canola (Brassica napus L.) as a representative model weed species. The effects of the presence or absence of spring wheat (Triticum aestivum L.) or winter cereal rye (Secale cereale L.) living mulches (mulch type) that had been seeded simultaneously with soybean grown using 38 or 76 cm row spacing (spatial arrangement) and the presence or absence of herbicides used for mid-season mulch termination (herbicide regime) were evaluated in three environments in Manitoba, Canada, in 2013 and 2014. Soybean yield was similar in the presence and absence of the living mulches. In the environment that received the lowest precipitation (Carman 2013), the mulches terminated with post-emergence glyphosate resulted in a 55% greater soybean yield compared to the mulches that remained live throughout the growing season. Inter-row mulches that had been living or terminated mid-season reduced volunteer canola seed production by about one-third (up to 9000 seeds mâ2). This study demonstrates the utility of wheat or cereal rye inter-row living mulches for enhanced interference with weeds during early soybean establishment
