    4-Hydroxy-2-Nonenal-Modified Glyceraldehyde-3-Phosphate Dehydrogenase Is Degraded by Cathepsin G in Rat Neutrophils

    Degradation of oxidized or oxidatively modified proteins is an essential part of the antioxidant defenses of cells. 4-Hydroxy-2-nonenal, a major reactive aldehyde formed by lipid peroxidation, causes many types of cellular damage. It has been reported that 4-hydroxy-2-nonenal-modified proteins are degraded by the ubiquitin-proteasome pathway or, in some cases, by the lysosomal pathway. However, our previous studies using U937 cells showed that 4-hydroxy-2-nonenal-modified glyceraldehyde-3-phosphate dehydrogenase is degraded by cathepsin G. In the present study, we isolated the 4-hydroxy-2-nonenal-modified glyceraldehyde-3-phosphate dehydrogenase-degrading enzyme from rat neutrophils to an active protein fraction of 28ā€‰kDa. Using the specific antibody, the 28ā€‰kDa protein was identified as cathepsin G. Moreover, the degradation activity was inhibited by cathepsin G inhibitors. These results suggest that cathepsin G plays a crucial role in the degradation of 4-hydroxy-2-nonenal-modified glyceraldehyde-3-phosphate dehydrogenase

    Physical and Histological Comparison of Hydroxyapatite, Carbonate Apatite, and Ī²-Tricalcium Phosphate Bone Substitutes

    Three commercially available artificial bone substitutes with different compositions, hydroxyapatite (HAp; NeoboneĀ®), carbonate apatite (CO3Ap; CytransĀ®), and Ī²-tricalcium phosphate (Ī²-TCP; CerasorbĀ®), were compared with respect to their physical properties and tissue response to bone, using hybrid dogs. Both NeoboneĀ® (HAp) and CerasorbĀ® (Ī²-TCP) were porous, whereas CytransĀ® (CO3Ap) was dense. Crystallite size and specific surface area (SSA) of NeoboneĀ® (HAp), CytransĀ® (CO3Ap), and CerasorbĀ® (Ī²-TCP) were 75.4 Ā± 0.9 nm, 30.8 Ā± 0.8 nm, and 78.5 Ā± 7.5 nm, and 0.06 m2/g, 18.2 m2/g, and 1.0 m2/g, respectively. These values are consistent with the fact that both NeoboneĀ® (HAp) and CerasorbĀ® (Ī²-TCP) are sintered ceramics, whereas CytransĀ® (CO3Ap) is fabricated in aqueous solution. Dissolution in pH 5.3 solution mimicking Howshipā€™s lacunae was fastest in CO3Ap (CytransĀ®), whereas dissolution in pH 7.3 physiological solution was fastest in Ī²-TCP (CerasorbĀ®). These results indicated that CO3Ap is stable under physiological conditions and is resorbed at Howshipā€™s lacunae. Histological evaluation using hybrid dog mandible bone defect model revealed that new bone was formed from existing bone to the center of the bone defect when reconstructed with CO3Ap (CytransĀ®) at week 4. The amount of bone increased at week 12, and resorption of the CO3Ap (CytransĀ®) was confirmed. Ī²-TCP (CerasorbĀ®) showed limited bone formation at week 4. However, a larger amount of bone was observed at week 12. Among these three bone substitutes, CO3Ap (CytransĀ®) demonstrated the highest level of new bone formation. These results indicate the possibility that bone substitutes with compositions similar to that of bone may have properties similar to those of bone

    Successful Resection of locally infiltrative Glomus Tumor without pulmonary resection

    Introduction: Extracutaneous glomus tumors occurring in the bronchus is very rare. Complete resection is basic procedure for treatment of glomus tumor. We present a glomus tumor of the left main bronchus that was successfully treated with rigid bronchoscopy followed by sleeve resection of the left main bronchus. Presentation of case: A 56-year-old man underwent two term resections to glomus tumor that originated from the left main bronchus. Firstly, we performed palliative resection with rigid bronchoscopy to make the correct diagnosis and evaluate the extent of the tumor. We subsequently performed curative resection. No complications or recurrence has occurred since the operation took place one year ago. Discussion: Before curative resection, it is important to confirm the diagnosis and spread of the tumor. Therefore, palliative tumor resection by rigid bronchoscopy was useful to make the correct diagnosis, evaluate the extent of the tumor and open the bronchial lumen. After bronchoscopic treatment, curative pulmonary resection was performed and preservation of lung function was successful. Conclusion: Two term resections enabled us to make an accurate diagnosis and evaluation, thereby preserving respiratory function without pulmonary resection

    Altered Growth Trajectory in Children Born to Mothers with Gestational Diabetes Mellitus and Preeclampsia

    Purpose: Gestational diabetes mellitus (GDM) and preeclampsia are leading causes of mortality and morbidity in mothers and children. High childhood body mass index (BMI) is among their myriad of negative outcomes. However, little is known about the trajectory of the child BMI exposed to GDM and co-occurring preeclampsia from early to mid-childhood. This study examined the independent and joint impact of GDM and preeclampsia on childhood BMI trajectory. Methods: A population-based sample of 356 mothers were recruited from OB/GYN clinics in New York. Their children were then followed annually from 18 to 72 months. Maternal GDM and preeclampsia status were obtained from medical records. Child BMI was calculated based on their height and weight at annual visits. Results: Hierarchical Linear Modeling was used to evaluate the trajectories of child BMI exposed to GDM and preeclampsia. BMI trajectory by GDM decreased (t-ratio = -2.24, ļæ½ļæ½=.45, 95% CI=-.05-.95, p = .07), but the trajectory by preeclampsia increased over time (t-ratio = 3.153, ļæ½ļæ½=.65, 95%CI=.11-1.18, p = .002). Moreover, there was a significant interaction between the two (t-ratio = -2.24, ļæ½ļæ½=-1.244, 95%CI=.15-2.33, p = .02), such that the BMI of children born to mothers with both GDM and preeclampsia showed consistent increases over time. Conclusions: GDM and preeclampsia could be used as a marker for childhood obesity risk and the identification of a high-risk group, providing potential early intervention. These findings highlight the importance of managing obstetric complications, as an effective method of child obesity prevention

    Development of ground pipeline system for high-level scientific data products of the Hisaki satellite mission and its application to planetary space weather

    The Hisaki satellite is the first-ever space telescope mission dedicated to planetary sciences. Atmospheres and magnetospheres of our solar system planets are continuously monitored by the extreme ultraviolet (EUV) spectrometer onboard Hisaki. This paper describes a data pipeline system developed for processing high-level scientific and ancillary data products from the Hisaki mission. The telemetry data downlinked from the satellite are stored in a ground telemetry database, processed in the pipeline to imaging spectral data with a 1-min temporal resolution and ancillary data products, and then archived in a public database. The imaging spectra can be further reduced to higher-level data products for practical scientific use. For example, light curves of the power emitted from Jupiterā€™s aurora and plasma torus with a temporal resolution of 10-min can be reduced from the imaging spectral data; the reduced light curves reveal the transport processes of energy and mass in Jupiterā€™s magnetosphere and associated interplanetary solar wind conditions. Continuous monitoring with Hisaki will contribute considerably to our understanding of space weather relating to planets in our solar system

    Video-assisted thoracic surgery attenuates perioperative oxidative stress response in lung cancer patients: a preliminary study

    Objectives: Reactive oxygen species (ROS) function as key metabolites that can impair biological processes. The aim of this preliminary study was to assess the perioperative oxidative changes in lung cancer surgery.Methods: We measured the levels of blood hydroperoxides, a type of ROS, as an index of oxidative injury to cellular components, as well as the plasma ferric-reducing ability as an index of total antioxidant potential in 32 lung cancer patients. Hydroperoxides were measured by the levels of diacron reactive oxygen metabolites (d-ROMs). The antioxidant potential was determined by the biological antioxidant potential (BAP), which represents the levels of endogenous antioxidant enzymes.Results: Lung cancer patients had slightly affected oxidative stress before surgery. The d-ROM and BAP levels after surgery and were significantly decreased than before surgery (p< 0.001) and the levels recovered preoperatively at third postoperative day. The d-ROM level in video-assisted surgery group (n=17) was significantly decreased than those in thoracotomy group (n=15) at third and seventh post-operative day (p < 0.001, and < 0.02).Conclusions: Lung cancer patients had already exposed oxidative stress before surgery and surgical intervention also generates large amounts of ROS. Video-assisted thoracic surgery can reduce the ROS compared to the standard thoractomy

    Variation of Jupiter's Aurora Observed by Hisaki/EXCEED: 3. Volcanic Control of Jupiter's Aurora:Io's Volcanic Effect on Jovian Aurora

    Temporal variation of Jupiter's northern aurora during enhanced Io volcanic activity was detected using the EXCEED spectrometer on board the Hisaki Earth-orbiting planetary space telescope. It was found that in association with reported Io volcanic events in early 2015, auroral power and estimated field-aligned currents were enhanced during day of year 40ā€“120. Furthermore, the far ultraviolet color ratio decreased during the event, indicating a decrease of auroral electron mean energy and total acceleration by <30%. During the episode of enhanced Io volcanic activity, Jupiter's magnetosphere contains more source current via increased suprathermal plasma density by up to 42%; therefore, it would have required correspondingly less electron acceleration to maintain the enhanced field-aligned current and corotation enforcement current. Sporadic large enhancements in auroral emission detected more frequently during the active period could have been contributed by nonadiabatic magnetospheric energization

    Amino acid residue doublet propensity in the proteinā€“RNA interface and its application to RNA interface prediction

    Proteinā€“RNA interactions play essential roles in a number of regulatory mechanisms for gene expression such as RNA splicing, transport, translation and post-transcriptional control. As the number of available proteinā€“RNA complex 3D structures has increased, it is now possible to statistically examine proteinā€“RNA interactions based on 3D structures. We performed computational analyses of 86 representative proteinā€“RNA complexes retrieved from the Protein Data Bank. Interface residue propensity, a measure of the relative importance of different amino acid residues in the RNA interface, was calculated for each amino acid residue type (residue singlet interface propensity). In addition to the residue singlet propensity, we introduce a new residue-based propensity, which gives a measure of residue pairing preferences in the RNA interface of a protein (residue doublet interface propensity). The residue doublet interface propensity contains much more information than the sum of two singlet propensities alone. The prediction of the RNA interface using the two types of propensities plus a position-specific multiple sequence profile can achieve a specificity of about 80%. The prediction method was then applied to the 3D structure of two mRNA export factors, TAP (Mex67) and UAP56 (Sub2). The prediction enables us to point out candidate RNA interfaces, part of which are consistent with previous experimental studies and may contribute to elucidation of atomic mechanisms of mRNA export

    Separated Transcriptomes of Male Gametophyte and Tapetum in Rice: Validity of a Laser Microdissection (LM) Microarray

    In flowering plants, the male gametophyte, the pollen, develops in the anther. Complex patterns of gene expression in both the gametophytic and sporophytic tissues of the anther regulate this process. The gene expression profiles of the microspore/pollen and the sporophytic tapetum are of particular interest. In this study, a microarray technique combined with laser microdissection (44K LM-microarray) was developed and used to characterize separately the transcriptomes of the microspore/pollen and tapetum in rice. Expression profiles of 11 known tapetum specific-genes were consistent with previous reports. Based on their spatial and temporal expression patterns, 140 genes which had been previously defined as anther specific were further classified as male gametophyte specific (71 genes, 51%), tapetum-specific (seven genes, 5%) or expressed in both male gametophyte and tapetum (62 genes, 44%). These results indicate that the 44K LM-microarray is a reliable tool to analyze the gene expression profiles of two important cell types in the anther, the microspore/pollen and tapetum

    Recruitment of Young Medical Apprentices (RYOMA) Project: A Comprehensive Surgical Education Program at a Local Academic Institute in Japan

    Objectives The number of young surgeons in Japan has significantly decreased in recent years, which may lead to future problems in the medical field. Therefore, comprehensive training programs for young surgeons are needed. Design Retrospective study Setting We developed a specific education program called the "Recruitment of Young Medical Apprentices" (RYOMA) project. Participants We performed this project between January 2008 and August 2013 on fourth- to sixth-year medical students and internship doctors. The RYOMA project included step-by-step surgical education programs on open and scopic procedures as dry, wet, and animal laboratory training. Our goal was to increase the number of young and specialist surgeons. Results Based on an interview questionnaire answered by 90 medical students, most young students were interested in surgical training and several chose to become surgeons in the future. The most positive opinions regarding the field of surgery were the impressive results achieved with surgery, whereas negative opinions included the difficulty of the surgical skill, physical concerns related to difficult work environments, and the severity of surgical procedures. The present program has begun to resolve negative opinions through adequate training or simulations. Of the 19 medical students and internship doctors who attended the RYOMA project in 2008, 17 trainees (90%) were satisfied with this special surgical program and 16 (88%) showed interest in becoming surgeons. The number of participants considering the field of surgery increased between 2008 and 2013. Of 23 participants, 19 (83%) had a positive opinion of the program after the training. Conclusions Gaining experience in surgical training from an early stage in medical school and step-by-step authorized education by teaching staff are important for recruiting students and increasing the number of young surgeons
