    Substituted diaryl diselenides: Cytotoxic and apoptotic effect in human colon adenocarcinoma cells

    AbstractAimsTo investigate the effects and study the underlying cell death mechanisms of diaryl diselenides, including: diphenyl diselenide (C6H5Se)2; 4-chlorodiphenyl diselenide (4-ClC6H4Se)2; 3-(trifluoromethyl)-diphenyl diselenide (3-CF3C6H4Se)2 and 4-methoxydiphenyl diselenide (4-MeOC6H4Se)2, on the human colon adenocarcinoma cell line HT-29.Main methodsThe viability of HT-29 cells after exposure to the diaryl diselenides and its substituted structures was based on the MTT assay. To verify if cell death was mediated throughout apoptosis mechanisms, flow cytometry and real-time PCR (qPCR) analyses were conducted.Key findingsThe MTT assay and flow cytometry analyses showed that (3-CF3C6H4Se)2 and (4-MeOC6H4Se)2 induced cytotoxicity through apoptosis mechanisms in HT-29 cells. qPCR revealed there was an up-regulation of pro-apoptotic (Bax, casapase-9, caspase-8, apoptosis-inducing factor (AIF) and Endonuclease G (EndoG)) and cell-cycle arrest genes (p53 and p21) and down-regulation of anti-apoptotic (Bcl-2 and survivin) and Myc genes.SignificanceThese results demonstrate that (3-CF3C6H4Se)₂ and (4-MeOC6H4Se)2 have the potential to induce apoptosis in HT-29 cells through the activation of caspase-dependent and independent pathways and through cell-cycle arrest

    Tratamento térmico em sementes de cenoura

    This study aimed at erradicating seed-borne fungi on carrot, Daucus carota L., seeds, cv. Brasília, by thermotherapy (dry heat) without any damage to its viability. The study was carried out in 1994/95, at Faculdade de Agronomia Eliseu Maciel, Federal University of Pelotas, RS, Brazil. Four seed lots, two harvested in 1993/94 and two harvested in 1994/95, were heated at 70°C for periods of: 0, 3, 6, 9, 12 and 15 days. The following parameters were used to evaluate the seeds: seed health test (blotter-test), standard germination test and vigor tests (cold test and accelerated aging test). The results showed that: dry heat at 70°C for 15 days eliminated seed-borne fungi of high quality carrot seeds, without reducing germination and vigor; high quality carrot seeds can be dehydrated to 1% moisture content without damage to it; and the seed quality or age affects its performance in heat treatments.O objetivo do presente estudo foi desenvolver um método para erradicar fungos de sementes de cenoura, Daucus carota L., da cultivar Brasília, por termoterapia (calor seco), sem prejuízo da viabilidade. O trabalho foi desenvolvido em 1994 e 1995, no Campus da Faculdade de Agronomia Eliseu Maciel, da Universidade Federal de Pelotas, RS. Quatro lotes de sementes de cenoura, dois da safra 93/94 e dois da safra 94/95 foram submetidos a diferentes tempos de exposição ao calor: 0, 3, 6, 9, 12 e 15 dias, em estufa com circulação de ar a 70°C. Os parâmetros utilizados para avaliação foram: teste de sanidade (blotter-test), teste de germinação padrão e testes de vigor (teste de frio e teste de envelhecimento acelerado). Os resultados obtidos indicam que a exposição ao calor seco, com temperatura de 70°C, durante 15 dias controla todos os fungos presentes em sementes de cenoura de alta qualidade fisiológica, sem prejuízo da germinação e do vigor; que sementes de cenoura com alta qualidade fisiológica podem ter seu grau de umidade reduzido a níveis próximos a 1,0% sem prejuízo da germinação e do vigor; e que a qualidade fisiológica e/ou idade das sementes influencia seu desempenho na termoterapia

    Nano‐/microfiber scaffold for tissue engineering: Physical and biological properties

    Alginate hydrogel (AH) has intrinsic physical and biological limitations that hinder its broader application in tissue engineering. We hypothesized that the inclusion of nanofibers in the hydrogel and the use of a biotemplate that mimics nature would enhance the translational potential of alginate hydrogels. In this study, we have shown a method to obtain nano‐/microfibers of titanium (nfTD) and hydroxyapatite (nfHY) using cotton as a biotemplate. These fibers were incorporated in the alginate hydrogel and the mechanical characteristics and biological response to these reinforced materials were evaluated. We observed that these nanofibers resembled the structure of natural collagen and did not mediate cell toxicity. The incorporation of nfTD or nfHY to the AH has not increased the viscosity of the hydrogel. Therefore, this is a feasible method to produce a scaffold with improved physical characteristics, while at the same time generating an enhanced environment for cell adhesion and proliferation. © 2012 Wiley Periodicals, Inc.     Perfil genético das linhagens de touros leiteiros Holandês e Jersey disponíveis no Brasil

    The Holstein and Jersey dairy cattle breeds are among the most prevalent in Brazil. To evaluate differences among the PTAs (predicted transmitting abilities) of these breeds and their lines, data were collected on 385 Holstein bulls and 82 Jersey sires with semen commercially available from nine Brazilian companies. Three different sire lines for each breed were found. The general linear models method was used for the comparison among lines and between breeds. The two most prevalent lines of Holstein breed presented higher average PTAs for milk yield (1,061.04 pounds and 975.32 pounds) and lower PTAs for percentage of milk solids (from -0.05% to -0.0003%) (P<0.05). These results indicate the supply of sires that mostly increase the milk yield in the Brazilian market. The Jersey breed presented a higher inbreeding coefficient (6.62%) than the Holstein breed (5.14%) (P<0.05). Although the Holstein breed presented higher PTAs for milk yield and lower PTAs for percentage of milk solids (P<0.05), the productive life of the Jersey breed (1.17 months) was longer than it was in the Holstein breed (0.40 months) (P<0.05). We identified the existence of an important variability of the available genetic profiles, what allows the Brazilian producers to choose the most adequate semen for their production system. It is necessary to consider the genetic profiles of sires' lines offered for artificial insemination in Brazil to understand and direct the genetic pattern of Brazilian dairy cattle

    Grau de multicolinearidade e variáveis envolvidas na dependência linear em modelos aditivo-dominantes

    The objective of this work was to assess the degree of multicollinearity and to identify the variables involved in linear dependence relations in additive-dominant models. Data of birth weight (n=141,567), yearling weight (n=58,124), and scrotal circumference (n=20,371) of Montana Tropical composite cattle were used. Diagnosis of multicollinearity was based on the variance inflation factor (VIF) and on the evaluation of the condition indexes and eigenvalues from the correlation matrix among explanatory variables. The first model studied (RM) included the fixed effect of dam age class at calving and the covariates associated to the direct and maternal additive and non-additive effects. The second model (R) included all the effects of the RM model except the maternal additive effects. Multicollinearity was detected in both models for all traits considered, with VIF values of 1.03 - 70.20 for RM and 1.03 - 60.70 for R. Collinearity increased with the increase of variables in the model and the decrease in the number of observations, and it was classified as weak, with condition index values between 10.00 and 26.77. In general, the variables associated with additive and non-additive effects were involved in multicollinearity, partially due to the natural connection between these covariables as fractions of the biological types in breed composition

    Can glutathione be a biomarker for suicide risk in women 18 months postpartum?

    Background: Suicide risk is prominent among the problems affecting populations, mainly due to the broad family, psychosocial and economic impact. Most individuals at suicidal risk have some mental disorder. There is considerable evidence that psychiatric disorders are accompanied by the activation of neuro-immune and neuro-oxidative pathways. The aim of the study is to evaluate the serum levels of oxidative stress biomarkers in women at risk of suicide after 18 months of postpartum. Methods: This is a case-control study, nested within a cohort study. From this cohort, 45 women [15 without mood disorders and 30 with mood disorders (Major depression and Bipolar disorder)] were selected at 18 months postpartum, the depression and suicide risk were assessed using the MiniInternational Neuropsychiatric Interview Plus (MINI-Plus) instrument, module A and C, respectively. Blood was collected and stored for later analysis of the reactive species (DCFH), superoxide dismutase (SOD), and glutathione reduced (GSH). For data analysis, the SPSS program was used. To compare the nominal covariates with the outcome GSH levels, the Student’s t-test or analysis of variance (ANOVA) was used. Spearman’s correlation was performed for analysis between the quantitative covariates and the outcome. To analyze the interaction between the factors, multiple linear regression was performed. Bonferroni analysis was used as an additional/secondary result to visualize differences in glutathione levels according to risk severity. After the adjusted analysis, p-values < 0.05 were considered statistically significant. Results: The percentage of suicide risk observed in our sample of women at 18 months postpartum was 24.4% (n = 11). After adjusting for the independent variables, only the presence of suicide risk remained associated with the outcome (β = 0.173; p = 0.007), low levels of GSH at 18 months after postpartum. Likewise, we verified the difference in GSH levels according to the degree of suicide risk, observing a significant association between the differences in glutathione means in the group of women with moderate to high risk compared to the reference group (no suicide risk) (p = 0.009). Conclusion: Our findings suggest that GSH may be a potential biomarker or etiologic factor in women at moderate to high risk of suicide
