    Variation in Environmental Parameters in Research and Aquaculture: Effects on Behaviour, Physiology and Cell Biology of Teleost Fish

    Over the last few years the increasing use of fish as animal models in scientific research and the increased fish breeding for human consumption have stressed the need for more knowledge on the effect of variations in environmental parameters on fish biology and on the welfare of specimens used both in research and aquaculture contexts. Experimental evidence shows that environmental variations can affect fish biology at various levels, from the molecular to that of the population, sometimes in a different way depending on the species considered. In order to achieve reproducible results in experiments involving fish it is necessary to set and maintain all environmental parameters constant at the optimal value to guarantee the wellness of the animal. The effects of the variation in environmental parameters on the behaviour, physiology and cell biology of teleosts are here discussed in order to provide useful information for research based on fish models

    Heat pipe fatigue test specimen: Metallurgical evaluation

    An innovative creep/fatigue test was run to simulate the temperature, mechanical load, and sodium corrosion conditions expected in a heat pipe designed to supply thermal energy to a Stirling cycle power converter. A sodium-charged Inconel 718 heat pipe with a Nickel 200 screen wick was operated for 1090 hr at temperatures between 950 K (1250 F) and 1050 K (1430 F) while being subjected to creep and fatigue loads in a servo-hydraulic testing machine. After testing, the heat pipe was sectioned and examined using optical microscopy, scanning electron microscopy, and electron microprobe analysis with wavelength dispersive x-ray spectroscopy. The analysis concentrated on evaluating topographic, microstructural, and chemical changes in the sodium exposed surfaces of the heat pipe wall and wick. Surface changes in the evaporator, condenser, and adiabatic sections of the heat pipe were examined in an effort to correlate the changes with the expected sodium environment in the heat pipe.     Increase in environmental temperature affects exploratory behaviour, anxiety and social preference in Danio rerio

    The aim of this work is to investigate the effect of a temperature increase on the behaviour of adult zebrafish (Danio rerio) maintained for 21 days at 34 °C (treatment) and 26 °C (control). The temperatures chosen are within the vital range of zebrafish and correspond to temperatures that this species encounters in the natural environment. Previous results showed that the same treatment affects the brain proteome and the behaviour of adult zebrafish by producing alterations in the proteins involved in neurotransmitter release and synaptic function and impairing fish exploratory behaviour. In this study, we have investigated the performance of treated and control zebrafish during environmental exploration by using four behavioural tests (novel tank diving, light and dark preference, social preference and mirror biting) that are paradigms for assessing the state of anxiety, boldness, social preference and aggressive behaviour, respectively. The results showed that heat treatment reduces anxiety and increases the boldness of zebrafish, which spent more time in potentially dangerous areas of the tank such as the top and the uncovered bright area and at a distance from the social group, thus decreasing protection for the zebrafish. These data suggest that the increase in ambient temperature may compromise zebrafish survival rate in the natural environment

    Feeding behaviour of larval European sea bass (Dicentrarchus labrax L.) in relation to temperature and prey density

    The feeding behaviour of larval European sea bass (Dicentrarchus labrax, L.) was analysed in relation to temperature and prey density under controlled laboratory conditions with the aim to assess the ability of larval fish to change the feeding tactic as a response to environmental changes. Larvae were acclimated for 20 days at three different temperatures (19, 22 and 26°C), and their feeding behaviour was then video-recorded in experimental trials, at two prey densities, consisting of swarms of 400/l and 1440/l Artemia nauplii. Results showed that there was a significant effect of the interaction between temperature and prey density on the proportion of swimming activity that was reduced at the high temperature-high prey density combination. This suggested a switching in the larval feeding behaviour from an active to an ambush tactic, when the temperature reached 26°C and the prey density was 1440 /l Artemia nauplii. These results are consistent with the current literature on fish larval behaviour in showing that the foraging tactic can be modulated by the interaction of different abiotic and biotic factors characterising the rearing environment

    Doxycycline increases neurogenesis and reduces microglia in the adult hippocampus.

    Adult hippocampal neurogenesis results in the continuous formation of new neurons and is a process of brain plasticity involved in learning and memory. Although inducible-reversible transgenic mouse models are increasingly being used to investigate adult neurogenesis, transgene control requires the administration of an activator, doxycycline (Dox), with unknown effects on adult neurogenesis. Here, we tested the effect of Dox administration on adult neurogenesis in vivo. We found that 4 weeks of Dox treatment at doses commonly used for gene expression control, resulted in increased neurogenesis. Furthermore, the dendrites of new neurons displayed increased spine density. Concomitantly, Iba1-expressing microglia was reduced by Dox treatment. These results indicate that Dox treatment may interfere with parameters of relevance for the use of inducible transgenic mice in studies of adult neurogenesis or brain inflammation

    Paroxysmal eye–head movements in Glut1 deficiency syndrome

    Objective:To describe a characteristic paroxysmal eye–head movement disorder that occurs in infants with Glut1 deficiency syndrome (Glut1 DS).Methods:We retrospectively reviewed the medical charts of 101 patients with Glut1 DS to obtain clinical data about episodic abnormal eye movements and analyzed video recordings of 18 eye movement episodes from 10 patients.Results:A documented history of paroxysmal abnormal eye movements was found in 32/101 patients (32%), and a detailed description was available in 18 patients, presented here. Episodes started before age 6 months in 15/18 patients (83%), and preceded the onset of seizures in 10/16 patients (63%) who experienced both types of episodes. Eye movement episodes resolved, with or without treatment, by 6 years of age in 7/8 patients with documented long-term course. Episodes were brief (usually &lt;5 minutes). Video analysis revealed that the eye movements were rapid, multidirectional, and often accompanied by a head movement in the same direction. Eye movements were separated by clear intervals of fixation, usually ranging from 200 to 800 ms. The movements were consistent with eye–head gaze saccades. These movements can be distinguished from opsoclonus by the presence of a clear intermovement fixation interval and the association of a same-direction head movement.Conclusions:Paroxysmal eye–head movements, for which we suggest the term aberrant gaze saccades, are an early symptom of Glut1 DS in infancy. Recognition of the episodes will facilitate prompt diagnosis of this treatable neurodevelopmental disorder.</jats:sec

    Does it do what it says on the tin? – Evaluation of an established UK University Employability Enhancement Award.

    University employability awards, in the UK particularly, aim to to assist students to develop career related skills and attributes and thus increase their potential to achieve graduate level employment. Self-report quantitative and qualitative data were collected via questionnaires, interviews and focus groups at intervals from two cohorts (N = 212) of a well-established career development and employability award at a large civic UK university. Findings indicated increases in confidence and aspiration, and in the ability to articulate and apply skills and abilities; also that the award may convey similar benefits to work experience. Survey data from award completers indicated that they had changed their career related behaviour, and students who have completed the award show a consistent small increase in their level of graduate employment when compared to the graduate employment figure for the university as a whole in the UK university destinations data. Employability related values, attitudes and behaviour may all change as a result of award experience. Reported changes implied a sense of improved resourcefulness. It is hypothesised that the award may enhance student employability somewhat via development of psychosocial resources, producing a shift in the student’s perception of self and identity
