BACKGROUND Rice transcription regulator OsWRKY13 influences the functioning of more than 500 genes in multiple signalling pathways, with roles in disease resistance, redox homeostasis, abiotic stress responses, and development. RESULTS To determine the putative transcriptional regulation mechanism of OsWRKY13, the putative cis-acting elements of OsWRKY13-influenced genes were analyzed using the whole genome expression profiling of OsWRKY13-activated plants generated with the Affymetrix GeneChip Rice Genome Array. At least 39 transcription factor genes were influenced by OsWRKY13, and 30 of them were downregulated. The promoters of OsWRKY13-upregulated genes were overrepresented with W-boxes for WRKY protein binding, whereas the promoters of OsWRKY13-downregulated genes were enriched with cis-elements putatively for binding of MYB and AP2/EREBP types of transcription factors. Consistent with the distinctive distribution of these cis-elements in up- and downregulated genes, nine WRKY genes were influenced by OsWRKY13 and the promoters of five of them were bound by OsWRKY13 in vitro; all seven differentially expressed AP2/EREBP genes and six of the seven differentially expressed MYB genes were suppressed by in OsWRKY13-activated plants. A subset of OsWRKY13-influenced WRKY genes were involved in host-pathogen interactions. CONCLUSION These results suggest that OsWRKY13-mediated signalling pathways are partitioned by different transcription factors. WRKY proteins may play important roles in the monitoring of OsWRKY13-upregulated genes and genes involved in pathogen-induced defence responses, whereas MYB and AP2/EREBP proteins may contribute most to the control of OsWRKY13-downregulated genes. This work was supported by grants from the National Program of High Technology Development of China, the National Program on the Development of Basic Research in China, and the National Natural Science Foundation of China

Optimal Sensor Location Design for Reliable Fault Detection in Presence of False Alarms

    Get PDF
To improve fault detection reliability, sensor location should be designed according to an optimization criterion with constraints imposed by issues of detectability and identifiability. Reliability requires the minimization of undetectability and false alarm probability due to random factors on sensor readings, which is not only related with sensor readings but also affected by fault propagation. This paper introduces the reliability criteria expression based on the missed/false alarm probability of each sensor and system topology or connectivity derived from the directed graph. The algorithm for the optimization problem is presented as a heuristic procedure. Finally, a boiler system is illustrated using the proposed method

Protective Effects of Hydrogen against Low-Dose Long-Term Radiation-Induced Damage to the Behavioral Performances, Hematopoietic System, Genital System, and Splenic Lymphocytes in Mice

Molecular hydrogen (H2) has been previously reported playing an important role in ameliorating damage caused by acute radiation. In this study, we investigated the effects of H2 on the alterations induced by low-dose long-term radiation (LDLTR). All the mice in hydrogen-treated or radiation-only groups received 0.1 Gy, 0.5 Gy, 1.0 Gy, and 2.0 Gy whole-body gamma radiation, respectively. After the last time of radiation exposure, all the mice were employed for the determination of the body mass (BM) observation, forced swim test (FST), the open field test (OFT), the chromosome aberration (CA), the peripheral blood cells parameters analysis, the sperm abnormality (SA), the lymphocyte transformation test (LTT), and the histopathological studies. And significant differences between the treatment group and the radiation-only groups were observed, showing that H2 could diminish the detriment induced by LDLTR and suggesting the protective efficacy of H2 in multiple systems in mice against LDLTR

Progress in Root Cause and Fault Propagation Analysis of Large-Scale Industrial Processes

In large-scale industrial processes, a fault can easily propagate between process units due to the interconnections of material and information flows. Thus the problem of fault detection and isolation for these processes is more concerned about the root cause and fault propagation before applying quantitative methods in local models. Process topology and causality, as the key features of the process description, need to be captured from process knowledge and process data. The modelling methods from these two aspects are overviewed in this paper. From process knowledge, structural equation modelling, various causal graphs, rule-based models, and ontological models are summarized. From process data, cross-correlation analysis, Granger causality and its extensions, frequency domain methods, information-theoretical methods, and Bayesian nets are introduced. Based on these models, inference methods are discussed to find root causes and fault propagation paths under abnormal situations. Some future work is proposed in the end

SIGNED DIRECTED GRAPH BASED MODELING AND ITS VALIDATION FROM PROCESS KNOWLEDGE AND PROCESS DATA

This paper is concerned with the fusion of information from process data and process connectivity and its subsequent use in fault diagnosis and process hazard assessment. The Signed Directed Graph (SDG), as a graphical model for capturing process topology and connectivity to show the causal relationships between process variables by material and information paths, has been widely used in root cause and hazard propagation analysis. An SDG is usually built based on process knowledge as described by piping and instrumentation diagrams. This is a complex and experience-dependent task, and therefore the resulting SDG should be validated by process data before being used for analysis. This paper introduces two validation methods. One is based on cross-correlation analysis of process data with assumed time delays, while the other is based on transfer entropy, where the correlation coefficient between two variables or the information transfer from one variable to another can be computed to validate the corresponding paths in SDGs. In addition to this, the relationship captured by data-based methods should also be validated by process knowledge to confirm its causality. This knowledge can be realized by checking the reachability or the influence of one variable on another based on the corresponding SDG which is the basis of causality. A case study of an industrial process is presented to illustrate the application of the proposed methods
