    Design and Operation of Oil Discharge Systems and Characteristics of Oil Used in the Baffin Island Oil Spill Project

    As part of the Baffin Island Oil Spill (BIOS) Project, two experimental oil discharges were made into bays at Cape Hatt at the northern end of Baffin Island. The objective was to allow the comparison of the nearshore fate and effects of an untreated surface oil slick and oil chemically dispersed into the water column. Weathered Lagomedio crude oil (15 cu m) was discharged onto the water surface in one bay, and most of the slick became stranded on the intertidal zone under the influence of an onshore wind and ebb tide. The oil thickness averaged about 1 mm on the beach face. The same volume and type of oil premixed with Corexit 9527 in a ratio of 10:1 was pumped into a second bay through a perforated diffuser pipe lying on the bottom sediments. The cloud of chemically dispersed oil contacted the bottom sediments and benthic organisms in the second bay and an adjacent third bay.     N-Terminal Phosphorylation Sites of Herpes Simplex Virus 1 ICP0 Differentially Regulate Its Activities and Enhance Viral Replication

    This is the publisher's version, also available electronically from http://jvi.asm.org/content/87/4/2109The herpes simplex virus 1 (HSV-1) infected cell protein 0 (ICP0) is an immediate-early phosphoprotein that transactivates viral gene expression. Evidence suggests that phosphorylation regulates the functions of ICP0, and three regions (termed regions I, II, and III) in the protein are known to be phosphorylated. Mutation of the putative phosphorylation sites within region I, termed Phos 1, which lies in the N-terminal portion of ICP0, impairs the E3 ubiquitin (Ub) ligase and ND10-disrupting activities of ICP0 in cell culture and diminishes viral replication. To identify the specific phosphorylation site(s) or residues responsible for the phenotypes observed with Phos 1, individual residues within region I were mutated to alanine (S224A, T226A, T231A, and T232A) and one double mutant S224A/T226A was constructed. Tissue culture studies demonstrated that the S224A, S224A/T226A, T231A, and T232A mutants were unable to dissociate the cellular protein PML from ND10 and that the S224/T226A mutant was defective in its ability to dissociate the cellular protein Sp100 from ND10. Additionally, the transactivation activity of ICP0 was impaired in the S224A and S224A/T226A mutants. The S224A and S224A/T226A mutant forms were more stable than wild-type ICP0, suggesting that their ability to autoubiquitinate was limited. Moreover, one ICP0 ubiquitination target, USP-7, was also more stable after infection with these two mutants. Lastly, the replication of the S224A and S224A/T226A mutant viruses was reduced in cell culture and in vivo. Overall, our data suggest that specific phosphorylation sites within region I differentially regulate the activities of ICP0, which are required for efficient viral replication

    Susceptibility of Northern Minnesota Lakes to Acid Deposition Impacts

    ABSTRACT-lake chemistry surveys indicate a large number of lakes with acid neutralizing capability (ANC) below 200 μeq/L occur in northeast Minnesota where shallow soils over bedrock and exposed rock outcrops predominate, and in moraine areas having rolling to steep topography in north-central and east-central Minnesota. In the Boundary Waters area, lake chemistry is strongly associated with bedrock geology. lakes with ANC \u3c100 μeq/L are associated with granite, basalt, and gabbro formations, while lakes with ANC of 100- 200 μeq/L are associated with slate and greenstone formations. In the rest of the state where soils are deep, landform, soil type, and lake hydrology determine lake chemistry. Most low ANC lakes are found in terminal moraine areas. These lakes are generally small ( \u3c40 ha in area), have limited groundwater inflow, and typically classed as precipitation-dominated seepage lakes. Higher ANC lakes (\u3e400 μeq/L) are often associated with agricultural and residential land uses. Relationships found between ANC and bedrock geology, and between ANC and landform and soils, provided the basis for mapping the distribution of low ANC surface waters in Minnesota. Empirical and process models used to evaluate the actual susceptibility of low ANC lakes in the Upper Midwest to acid deposition impacts and indicated precipitation pH 4.6-4.7 is a threshold level for lake acidification.     Identification of salivary mucin MUC7 binding proteins from Streptococcus gordonii

    <p>Abstract</p> <p>Background</p> <p>The salivary mucin MUC7 (previously known as MG2) can adhere to various strains of streptococci that are primary colonizers and predominant microorganisms of the oral cavity. Although there is a growing interest in interaction between oral pathogens and salivary mucins, studies reporting the specific binding sites on the bacteria are rather limited. Identification and characterization of the specific interacting proteins on the bacterial cell surface, termed adhesins, are crucial to further understand host-pathogen interactions.</p> <p>Results</p> <p>We demonstrate here, using purified MUC7 to overlay blots of SDS-extracts of <it>Streptococcus gordonii </it>cell surface proteins, 4 MUC7-binding bands, with apparent molecular masses of 62, 78, 84 and 133 kDa from the <it>Streptococcus gordonii </it>strain, PK488. Putative adhesins were identified by in-gel digestion and subsequent nanoLC-tandem mass spectrometry analysis of resultant peptides. The 62 kDa and 84 kDa bands were identified as elongation factor (EF) Tu and EF-G respectively. The 78 kDa band was a <it>hppA </it>gene product; the 74 kDa oligopeptide-binding lipoprotein. The 133 kDa band contained two proteins; alpha enolase and DNA-directed RNA polymerase, beta' subunit. Some of these proteins, for example alpha enolase are expected to be intracellular, however, flow cytometric analysis confirmed its location on the bacterial surface.</p> <p>Conclusion</p> <p>Our data demonstrated that <it>S. gordonii </it>expressed a number of putative MUC7 recognizing proteins and these contribute to MUC7 mucin binding of this streptococcal strain.</p

    Paradoxical Facilitation of Working Memory after Basolateral Amygdala Damage

    Working memory is a vital cognitive capacity without which meaningful thinking and logical reasoning would be impossible. Working memory is integrally dependent upon prefrontal cortex and it has been suggested that voluntary control of working memory, enabling sustained emotion inhibition, was the crucial step in the evolution of modern humans. Consistent with this, recent fMRI studies suggest that working memory performance depends upon the capacity of prefrontal cortex to suppress bottom-up amygdala signals during emotional arousal. However fMRI is not well-suited to definitively resolve questions of causality. Moreover, the amygdala is neither structurally or functionally homogenous and fMRI studies do not resolve which amygdala sub-regions interfere with working memory. Lesion studies on the other hand can contribute unique causal evidence on aspects of brain-behaviour phenomena fMRI cannot “see”. To address these questions we investigated working memory performance in three adult female subjects with bilateral basolateral amygdala calcification consequent to Urbach-Wiethe Disease and ten healthy controls. Amygdala lesion extent and functionality was determined by structural and functional MRI methods. Working memory performance was assessed using the Wechsler Adult Intelligence Scale-III digit span forward task. State and trait anxiety measures to control for possible emotional differences between patient and control groups were administered. Structural MRI showed bilateral selective basolateral amygdala damage in the three Urbach-Wiethe Disease subjects and fMRI confirmed intact functionality in the remaining amygdala sub-regions. The three Urbach-Wiethe Disease subjects showed significant working memory facilitation relative to controls. Control measures showed no group anxiety differences. Results are provisionally interpreted in terms of a ‘cooperation through competition’ networks model that may account for the observed paradoxical functional facilitation effect

    Cystic fibrosis: An inherited disease affecting mucin-producing organs

    Our current understanding of cystic fibrosis (CF) has revealed that the biophysical properties of mucus play a considerable role in the pathogenesis of the disease in view of the fact that most mucus-producing organs are affected in CF patients. In this review, we discuss the potential causal relationship between altered cystic fibrosis transmembrane conductance regulator (CFTR) function and the production of mucus with abnormal biophysical properties in the intestine and lungs, highlighting what has been learned from cell cultures and animal models that mimic CF pathogenesis. A similar cascade of events, including mucus obstruction, infection and inflammation, is common to all epithelia affected by impaired surface hydration. Hence, the main structural components of mucus, namely the polymeric, gel-forming mucins, are critical to the onset of the disease. Defective CFTR leads to epithelial surface dehydration, altered pH/electrolyte composition and mucin concentration. Further, it can influence mucin transition from the intracellular to extracellular environment, potentially resulting in aberrant mucus gel formation. While defective HCO3− production has long been identified as a feature of CF, it has only recently been considered as a key player in the transition phase of mucins. We conclude by examining the influence of mucins on the biophysical properties of CF sputum and discuss existing and novel therapies aimed at removing mucus from the lungs
