    Preliminary evaluation of probiotic properties of Lactobacillus strains isolated from Sardinian dairy products

    Twenty-three Lactobacillus strains of dairy origin were evaluated for some functional properties relevant to their use as probiotics. A preliminary subtractive screening based on the abilities to inhibit the growth of microbial pathogens and hydrolyze conjugated bile salts was applied, and six strains were selected for further characterization including survival under gastrointestinal environmental conditions, adhesion to gut epithelial tissue, enzymatic activity, and some safety properties. All selected strains maintained elevated cell numbers under conditions simulating passage through the human gastrointestinal tract, well comparable to the values obtained for the probiotic strain Lactobacillus rhamnosus GG, and were able to adhere to Caco-2 cells to various extents (from 3 to 20%). All strains exhibited high aminopeptidase, and absent or very low proteolytic and strong β-galactosidase activities; none was found to be haemolytic or to produce biogenic amines and all were susceptible to tetracycline, chloramphenicol, erythromycin, ampicillin, and amoxicillin/clavulanic acid. Our results indicate that the Lactobacillus strains analyzed could be considered appropriate probiotic candidates, due to resistance to GIT simulated conditions, antimicrobial activity, adhesion to Caco-2 cell-line, and absence of undesirable properties. They could be used as adjunct cultures for contributing to the quality and health related functional properties of dairy products

    Is the Sardinian Blue Zone the New Shangri-La for mental health? Evidence on depressive symptoms and its correlates in late adult life span

    Background: An area of extraordinary longevity (i.e., Sardinian Blue Zone) characterized by a very high prevalence of long-lived successful agers has been validated in Sardinia, an Italian island located in the Mediterranean Sea. Aims: This study was primarily aimed at examining whether dietary habits (intake of vegetables and fruit, animal-derived proteins, and carbohydrates-rich food), time spent on hobbies, subjective physical health, and socio-cultural context (Sardinian Blue Zone vs. another Sardinian rural area) predicted self-reported depressive symptoms in older adults recruited in the Sardinian Blue Zone and another Sardinian rural area not being characterized by a higher prevalence of long-lived individuals. Methods: Three hundred and eighteen community-dwellers, age 65 years and older, 188 females and 130 males (Mage = 79.1 years, SD = 6.9 years) were recruited from the Sardinian Blue Zone and another Sardinian rural area. Each participant individually completed a battery of instruments to assess lifestyle, food habits, perceived physical health, and depressive symptoms through the CES-D inventory. Results: Significant associations were found between depressive signs, perceived physical health, time spent gardening, proteins, and carbohydrates intake, respectively. Approximately 17% of the variance in the CES-D condition was predicted by socio-cultural context, perceived physical health, and gardening. Participants recruited in the Sardinian Blue Zone spent more time gardening and self-reported better physical health. Conclusions: current results suggest that a socio-cultural context where people age well (i.e., the Sardinian Blue Zone), and a healthy and physically active lifestyle are crucial for promoting well-being in late adulthood

    A combined non-invasive approach to the study of a mosaic model: First laboratory experimental results

    This paper presents first laboratory results of a combined approach carried out by the use of three different portable non-invasive electromagnetic methods: Digital holographic speckle pattern interferometry (DHSPI), stimulated infrared thermography (SIRT) and holographic subsurface radar (HSR), proposed for the analysis of a custom-built wall mosaic model. The model reproduces a series of defects (e.g., cracks, voids, detachments), simulating common deteriorated, restored or reshuffled areas in wall mosaics. DHSPI and SIRT, already well known in the field of non-destructive (NDT) methods, are full-field contactless techniques, providing complementary information on the subsurface hidden discontinuities. The use of DHSPI, based on optical imaging and interferometry, provides remote control and visualization of surface micro-deformation after induced thermal stress, while the use of SIRT allows visualization of thermal energy diffusion in the surface upon the induced thermal stress. DHSPI and SIRT data are complemented by the use of HSR, a contact method that provides localized information about the distribution of contrasts in dielectric permittivity and related possible anomalies. The experimental results, made by the combined use of these methods to the identification of the known anomalies in the mosaic model, are presented and discussed here as a contribution in the development of an efficient non-invasive approach to the in-situ subsurface analysis of ancient wall mosaics

    La urbanización actual es un fenómeno de naturaleza indudablemente compleja, en ella puede reconocerse cierta homogenización formal y funcional de territorios debido a los modos de crecimiento propios de procesos globales de fragmentación y segregación socio espacial. Cada ciudad comparte características con otras ciudades, sin embargo, manifiesta particularidades propias que deben ser interpretadas desde la mirada urbano-arquitectónica del Nuevo Urbanismo. Esto significa centrarse en las relaciones espaciales-sociales que conforman el espacio público. En el contexto de esta problemática global-local se pone a consideración un estudio de la las periferias -internas y externas- que forman parte de las nuevas geografías urbanas de la Ciudad de San Juan, Argentina, propio de los procesos contemporáneos. En él se expone la estrategia proyectual central para la articulación urbanística y social, con el propósito de contrarrestar el fenómeno de periferización y desigualdad socio-espacial, dado por el proceso de ocupación no sustentable de la planta urbana.Current urbanization is undoubtedly a phenomenon of complex nature, in it certain formal and functional homogenization of territories are recognized due to growth modes brought on by global fragmentation processes and socio-spatial segregations. Each city shares characteristics with other cities, however, each one manifests its own characteristics that must be interpreted from the urban- architectural look of New Urbanism. This means focusing on socio-spatial relations that make up the public space. In the context of this global-local problematic a study is set to consider internal and external peripheries that are part of the new urban geographies of the City of San Juan, Argentina, due to contemporary processes. In it the central design strategy for the urban and social articulation is exposed, in order to counteract the phenomenon of peripheralization and socio-spatial inequality, given by the process of unsustainable occupation of the urban plant

    Olive Oil Phenolic Compounds’ Activity against Age-Associated Cognitive Decline: Clinical and Experimental Evidence

    Epidemiological studies have shown that consuming olive oil rich in phenolic bioactive compounds is associated with a lower risk of neurodegenerative diseases and better cognitive performance in aged populations. Since oxidative stress is a common hallmark of age-related cognitive decline, incorporating exogenous antioxidants could have beneficial effects on brain aging. In this review, we firstly summarize and critically discuss the current preclinical evidence and the potential neuroprotective mechanisms. Existing studies indicate that olive oil phenolic compounds can modulate and counteract oxidative stress and neuroinflammation, two relevant pathways linked to the onset and progression of neurodegenerative processes. Secondly, we summarize the current clinical evidence. In contrast to preclinical studies, there is no direct evidence in humans of the bioactivity of olive oil phenolic compounds. Instead, we have summarized current findings regarding nutritional interventions supplemented with olive oil on cognition. A growing body of research indicates that high consumption of olive oil phenolic compounds is associated with better preservation of cognitive performance, conferring an additional benefit, independent of the dietary pattern. In conclusion, the consumption of olive oil rich in phenolic bioactive compounds has potential neuroprotective effects. Further research is needed to understand the underlying mechanisms and potential clinical applications

    Cardiovascular and Metabolic Benefits of Extra Virgin Olive Oil Phenolic Compounds: Mechanistic Insights from In Vivo Studies

    Extra virgin olive oil (EVOO) represents a significant source of monounsaturated fatty acids (MUFA) and vitamin E, but it is also considered a functional food, due to the content of peculiar bioactive molecules, such as phenolic compounds, being able to modulate various processes related to aging and the most common metabolic and degenerative diseases. A lot of experimental research has focused on some of these components, but in most cases, the studies were performed in vitro testing compounds at non-physiological concentrations and achieving results that cannot easily be translated in vivo. Recent clinical studies demonstrated that in vivo these compounds are able to regulate physiological functions and prevent several pathological events including metabolic and cardiovascular diseases (CVDs), which represent the main causes of death worldwide. This review aims to sum up the major evidence on the beneficial effects of EVOO phenolic compounds in vivo on these pathologies, describing and evaluating the efficacy in relation to the mechanisms of diseases of the whole phenolic fraction and some of its specific components

    Ferulic acid metabolites attenuate lps-induced inflammatory response in enterocyte-like cells

    Ferulic acid (FA) is a polyphenol pertaining to the class of hydroxycinnamic acids present in numerous foods of a plant origin. Its dietary consumption leads to the formation of several phase I and II metabolites in vivo, which represent the largest amount of ferulates in the circulation and in the intestine in comparison with FA itself. In this work, we evaluated their efficacy against the proinflam-matory effects induced by lipopolysaccharide (LPS) in intestinal Caco-2 cell monolayers, as well as the mechanisms underlying their protective action. LPS-induced overexpression of proinflammatory enzymes such as inducible nitric oxide synthase (iNOS) and the consequent hyperproduction of nitric oxide (NO) and cyclic guanosine monophosphate (cGMP) were limited by physiological relevant concentrations (1 µM) of FA, its derivatives isoferulic acid (IFA) and dihydroferulic acid (DHFA), and their glucuronidated and sulfated metabolites, which acted upstream by limiting the activation of MAPK p38 and ERK and of Akt kinase, thus decreasing the nuclear factor kappa-light-chain-enhancer of activated B cells (NF-kB) translocation into the nucleus. Furthermore, the compounds were found to promote the expression of Nrf2, which may have contributed to the downregulation of NF-kB activity. The overall data show that phase I/II metabolites retain the efficacy of their dietary free form in contrasting inflammatory response

    First Italian validation of the “satisfaction with simulation experience” scale (Sse) for the evaluation of the learning experience through simulation

    Background and aim of the work. Training in simulation through “mannequins” is increasingly widespread among nursing students. In the Italian context, however, there are no tools that measure the degree of student satisfaction after clinical training through simulation. The aim of the study is to provide a first validation in Italian of the Satisfaction with simulation experience” (SSE) scale, a tool already validated in several languages. Methods. After obtaining the author’s consent, the SSE was subjected to forward and backward translation. The content validity was assessed by 5 training experts by calculating the Content Validity Index by Item and by Scale (I-CVI and S-CVI); the face validity was tested on 4 nursing students who had participated in a simulation experience. Subsequently, the SSE was administered to 10 nursing students with testretest after 7 days in order to evaluate the reliability by calculating the reliability coefficient (r) and Cronbach’s α. Results. The author approved the final version of the SSE translated into Italian: I-CVI values>0.80 and S-CVI was 0.94. r is 0.88 and the α of the scale is 0.713. Conclusions. The detected values of I-CVI and S-CVI are satisfactory, demonstrating the validity of the content of the SSE-ITA. The test-retest showed “optimal” reliability and the α was considered acceptable given the little deviation from the original (0.776). Although the results demonstrate satisfactory values, this is a first validation and other studies with larger samples are needed. (www.actabiomedica.it)
