    Delivery of short interfering ribonucleic acid-complexed magnetic nanoparticles in an oscillating field occurs via caveolae-mediated endocytosis

    Gene delivery technologies to introduce foreign genes into highly differentiated mammalian cells have improved significantly over the last few decades. Relatively new techniques such as magnetic nanoparticle-based gene transfection technology are showing great promise in terms of its high transfection efficiency and wide-ranging research applications. We have developed a novel gene delivery technique, which uses magnetic nanoparticles moving under the influence of an oscillating magnetic array. Herein we successfully introduced short interfering RNA (siRNA) against green fluorescent protein (GFP) or actin into stably-transfected GFP-HeLa cells or wild-type HeLa and rat aortic smooth muscle cells, respectively. This gene silencing technique occurred in a dose- and cell density- dependent manner, as reflected using fluorescence intensity and adhesion assays. Furthermore, using endocytosis inhibitors, we established that these magnetic nanoparticle-nucleic acid complexes, moving across the cell surface under the influence of an oscillating magnet array, enters into the cells via the caveolae-mediated endocytic pathway

    Web Application to Investigate Butler County Overdose Death Data

    Background: Drug overdose deaths, specifically opioid-related deaths, are a public health crisis in the United States with high incidence observed in many Midwestern states, including Ohio. Butler County, Ohio, has the third highest opioid-related death rate in the state. Information on overdose deaths, collected by the county coroner, can serve as a data source for analysis of this public health concern. Given this access, stakeholders can investigate trends in their community for their idiosyncratic interest.Methods: A web application was developed, using the R Shiny package, to visualize and explore the characteris-tics of all overdose deaths in Butler County between 2013 and 2018. Demographics of the decedents, drugs found in the decedentsā€™ postmortem toxicology analyses, annual trends in overdose deaths, and the location of these cases can be examined.Results: The web application provides a graphical user interface that allows a user to request specific analyses and summaries. ā€œWho is dying from opioid overdoses?,ā€ ā€œWhat drugs, including opioids, are found in people dying from drug overdoses?,ā€ and ā€œHas the number of opioid involved deaths increased in a specific community over time?ā€ are examples of questions that can be explored using this application.Conclusion: This application empowers both the public and local policymakers to investigate the impact of overdose deaths on their communities. Understanding characteristics of the epidemic is an important first step to addressing this problem. The expansion of this application to include other counties in Ohio could be truly beneficial to communities that need it

    Using internet search queries for infectious disease surveillance: screening diseases for suitability

    Background: Internet-based surveillance systems provide a novel approach to monitoring infectious diseases. Surveillance systems built on internet data are economically, logistically and epidemiologically appealing and have shown significant promise. The potential for these systems has increased with increased internet availability and shifts in health-related information seeking behaviour. This approach to monitoring infectious diseases has, however, only been applied to single or small groups of select diseases. This study aims to systematically investigate the potential for developing surveillance and early warning systems using internet search data, for a wide range of infectious diseases. Methods: Official notifications for 64 infectious diseases in Australia were downloaded and correlated with frequencies for 164 internet search terms for the period 2009ā€“13 using Spearmanā€™s rank correlations. Time series cross correlations were performed to assess the potential for search terms to be used in construction of early warning systems. Results: Notifications for 17 infectious diseases (26.6%) were found to be significantly correlated with a selected search term. The use of internet metrics as a means of surveillance has not previously been described for 12 (70.6%) of these diseases. The majority of diseases identified were vaccine-preventable, vector-borne or sexually transmissible; cross correlations, however, indicated that vector-borne and vaccine preventable diseases are best suited for development of early warning systems. Conclusions: The findings of this study suggest that internet-based surveillance systems have broader applicability to monitoring infectious diseases than has previously been recognised. Furthermore, internet-based surveillance systems have a potential role in forecasting emerging infectious disease events, especially for vaccine-preventable and vector-borne diseases.     Identification of an immunodominant CD4+ T cell epitope in the VP6 protein of rotavirus following intranasal immunization of BALB/c mice

    AbstractThe only lymphocytes required for protection against fecal rotavirus shedding after intranasal immunization of BALB/c (H-2d) mice with a chimeric rotavirus VP6 protein (MBPāˆ·VP6) and the mucosal adjuvant LT(R192G) are CD4+ T cells. The purpose of this study was to identify CD4+ T cell epitopes within VP6 that might be responsible for this protection. To make this determination, spleen cells obtained from BALB/c mice following intranasal immunization with MBPāˆ·VP6/LT(R192G) were stimulated in vitro with either MBPāˆ·VP6 or overlapping VP6 peptides containing ā‰¤30 amino acids (AA). The numbers of memory (CD44high) CD4+ T cells stimulated to produce TH1 and TH17 cytokines (IFNĪ³ and IL-17), as well as the quantities of these cytokines released into the cell supernatants, were then measured relative to those produced in mock-stimulated cells from the same animals. One epitope expected to be found was the VP6 14-mer AA289ā€“302, previously identified as a CD4+ T cell epitope in H-2d mice. This was not observed but instead the only VP6 epitope identified was AA242ā€“259, the dominant CD4+ T cell epitope previously reported after oral, live rotavirus immunization

    Prostatic trypsin-like kallikrein-related peptidases (KLKs) and other prostate-expressed tryptic proteinases as regulators of signalling via proteinase-activated receptors (PARs)

    The prostate is a site of high expression of serine proteinases including members of the kallikrein-related peptidase (KLK) family, as well as other secreted and membrane-anchored serine proteinases. It has been known for some time that members of this enzyme family elicit cellular responses by acting directly on cells. More recently, it has been recognised that for serine proteinases with specificity for cleavage after arginine and lysine residues (trypsin-like or tryptic enzymes) these cellular responses are often mediated by cleavage of members of the proteinase-activated receptor (PAR) family - a four member sub-family of G protein-coupled receptors. Here, we review the expression of PARs in prostate, the ability of prostatic trypsin-like KLKs and other prostate-expressed tryptic enzymes to cleave PARs, as well as the prostate cancer-associated consequences of PAR activation. In addition, we explore the dysregulation of trypsin-like serine proteinase activity through the loss of normal inhibitory mechanisms and potential interactions between these dysregulated enzymes leading to aberrant PAR activation, intracellular signalling and cancer-promoting cellular changes
