    Nonlinear optical properties of a channel waveguide produced with crosslinkable ferroelectric liquid crystals

    A binary mixture of ferroelectric liquid crystals (FLCs) was used for the design of a channel waveguide. The FLCs possess two important functionalities: a chromophore with a high hyperpolarizability β\beta and photoreactive groups. The smectic liquid crystal is aligned in layers parallel to the glass plates in a sandwich geometry. This alignment offers several advantages, such as that moderate electric fields are sufficient to achieve a high degree of polar order. The arrangement was then permanently fixed by photopolymerization which yielded a polar network possessing a high thermal and mechanical stability which did not show any sign of degradation within the monitored period of several months. The linear and nonlinear optical properties have been measured and all four independent components of the nonlinear susceptibility tensor dˉ\bar d have been determined. The off-resonant dd-coefficients are remarkably high and comparable to those of the best known inorganic materials. The alignment led to an inherent channel waveguide for p-polarized light without additional preparation steps. The photopolymerization did not induce scattering sites in the waveguide and the normalized losses were less than 2 dB/cm.     Idiopathic hypertrophic pachymeningitis presenting with occipital neuralgia

    Background: Although occipital neuralgia is usually caused by degenerative arthropathy, nearly 20 other aetiologies may lead to this condition.Methods: We present the first case report of hypertrophic pachymeningitis revealed by isolated occipital neuralgia.Results and conclusions: Idiopathic hypertrophic pachymeningitis is a plausible cause of occipital neuralgia and may present without cranial-nerve palsy. There is no consensus on the treatment for idiopathic hypertrophic pachymeningitis, but the usual approach is to start corticotherapy and then to add immunosuppressants. When occipital neuralgia is not clinically isolated or when a first-line treatment fails, another disease diagnosis should be considered.     Data filtering in the readout of the CMS Electromagnetic Calorimeter

    For an efficient data taking, the Electromagnetic Calorimeter (ECAL) data of the CMS experiment must be limited to 10\% of the full event size (1MB). Other requirements limit the average data size to 2kB per data acquisition link. These conditions imply a reduction factor of close to twenty on the data collected. The data filtering in the readout of the ECAL detector is discussed. Test beam data are used to study the digital filtering applied in the readout channels and a full detector simulation allows to estimate the energy thresholds to achieve the desired data suppression factor

    BACKGROUND: Nasopharyngeal carcinomas (NPC) are consistently associated with the Epstein-Barr virus (EBV). Their malignant epithelial cells contain the viral genome and express several antigenic viral proteins. However, the mechanisms of immune escape in NPCs are still poorly understood. EBV-transformed B-cells have been reported to release exosomes carrying the EBV-encoded latent membrane protein 1 (LMP1) which has T-cell inhibitory activity. Although this report suggested that NPC cells could also produce exosomes carrying immunosuppressive proteins, this hypothesis has remained so far untested. METHODS: Malignant epithelial cells derived from NPC xenografts – LMP1-positive (C15) or negative (C17) – were used to prepare conditioned culture medium. Various microparticles and vesicles released in the culture medium were collected and fractionated by differential centrifugation. Exosomes collected in the last centrifugation step were further purified by immunomagnetic capture on beads carrying antibody directed to HLA class II molecules. Purified exosomes were visualized by electron microscopy and analysed by western blotting. The T-cell inhibitory activities of recombinant LMP1 and galectin 9 were assessed on peripheral blood mononuclear cells activated by CD3/CD28 cross-linking. RESULTS: HLA-class II-positive exosomes purified from C15 and C17 cell supernatants were containing either LMP1 and galectin 9 (C15) or galectin 9 only (C17). Recombinant LMP1 induced a strong inhibition of T-cell proliferation (IC50 = 0.17 nM). In contrast recombinant galectin 9 had a weaker inhibitory effect (IC50 = 46 nM) with no synergy with LMP1. CONCLUSION: This study provides the proof of concept that NPC cells can release HLA class-II positive exosomes containing galectin 9 and/or LMP1. It confirms that the LMP1 molecule has intrinsic T-cell inhibitory activity. These findings will encourage investigations of tumor exosomes in the blood of NPC patients and assessment of their effects on various types of target cells

    miR-31 is consistently inactivated in EBV-associated nasopharyngeal carcinoma and contributes to its tumorigenesis

    BACKGROUND: As a distinctive type of head and neck cancers, nasopharyngeal carcinoma (NPC) is genesis from the clonal Epstein-Barr virus (EBV)-infected nasopharyngeal epithelial cells accumulated with multiple genetic lesions. Among the recurrent genetic alterations defined, loss of 9p21.3 is the most frequent early event in the tumorigenesis of EBV-associated NPC. In addition to the reported CDKN2A/p16, herein, we elucidated the role of a miRNA, miR-31 within this 9p21.3 region as NPC-associated tumor suppressor. METHODS: The expression and promoter methylation of miR-31 were assessed in a panel of NPC tumor lines and primary tumors. Its in vitro and in vivo tumor suppression function was investigated through the ectopic expression of miR-31 in NPC cells. We also determined the miR-31 targeted genes and its involvement in the growth in NPC. RESULTS: Downregulation of miR-31 expression was detected in almost all NPC cell line, patient-derived xenografts (PDXs) and primary tumors. Both homozygous deletion and promoter hypermethylation were shown to be major mechanisms for miR-31 silencing in this cancer. Strikingly, loss of miR-31 was also obviously observed in the dysplastic lesions of nasopharynx. Restoration of miR-31 in C666-1 cells inhibited the cell proliferation, colony-forming and migratory capacities. Dramatic reduction of in vitro anchorage-independent growth and in vivo tumorigenic potential were demonstrated in the stable clones expressing miR-31. Furthermore, we proved that miR-31 suppressed the NPC cell growth via targeting FIH1 and MCM2. CONCLUSIONS: The findings provide strong evidence to support miR-31 as a new NPC-associated tumor suppressor on 9p21.3 region.
