Directional outlyingness applied to distances between genomic words

    Get PDF
The detection of outlier curves/images is crucial in many areas, such as environmental, meteorological, medical, or economic contexts. In the functional framework, outlying observations are not only those that contain atypically high or low values, but also curves that present a different shape or pattern from the rest of the curves in the sample. In this short paper, we mention some recent methods for outlier detection in functional data and apply a recently proposed measure, the directional outlyingness, and the functional outlier map to detect words with outlying distance distribution in the human genome.publishe

FAPESP - FUNDAÇÃO DE AMPARO À PESQUISA DO ESTADO DE SÃO PAULOCNPQ - CONSELHO NACIONAL DE DESENVOLVIMENTO CIENTÍFICO E TECNOLÓGICOThe pharmacokinetics and the local toxicity of commercial and liposome-encapsulated mepivacaine formulations injected intra-orally in rats were studied. Animals were divided in groups (n=4-6) and treated with 0.1 mL of the formulations: 2% mepivacaine with 1:100,000 epinephrine (MVC(2%EPI)), 3% mepivacaine (MVC(3%)), and 2% liposome-encapsulated mepivacaine (MVC(LUV)). The results showed that the 2% liposome-encapsulated mepivacaine reduced C(max), prolonged AUC(0-infinity) and t(1/2) compared with 3% plain and 2% vasoconstritor-associated mepivacaine, after intraoral injection. In addition, it was also observed that liposomal mepivacaine might protect the tissue against local inflammation evoked by plain or vasoconstrictors-associated mepivacaine, giving supporting evidence for its safety and possible clinical use in dentistry1726876FAPESP - FUNDAÇÃO DE AMPARO À PESQUISA DO ESTADO DE SÃO PAULOCNPQ - CONSELHO NACIONAL DE DESENVOLVIMENTO CIENTÍFICO E TECNOLÓGICOFAPESP - FUNDAÇÃO DE AMPARO À PESQUISA DO ESTADO DE SÃO PAULOCNPQ - CONSELHO NACIONAL DE DESENVOLVIMENTO CIENTÍFICO E TECNOLÓGICOFAPESP [Proc 06/00121-9]2006/00121-9sem informaçã

Modelling the tumour microenvironment in long-term microencapsulated 3D co-cultures recapitulates phenotypic features of disease progression

3D cell tumour models are generated mainly in non-scalable culture systems, using bioactive scaffolds. Many of these models fail to reflect the complex tumour microenvironment and do not allow long-term monitoring of tumour progression. To overcome these limitations, we have combined alginate microencapsulation with agitation-based culture systems, to recapitulate and monitor key aspects of the tumour microenvironment and disease progression. Aggregates of MCF-7 breast cancer cells were microencapsulated in alginate, either alone or in combination with human fibroblasts, then cultured for 15 days. In co-cultures, the fibroblasts arranged themselves around the tumour aggregates creating distinct epithelial and stromal compartments. The presence of fibroblasts resulted in secretion of pro-inflammatory cytokines and deposition of collagen in the stromal compartment. Tumour cells established cell–cell contacts and polarised around small lumina in the interior of the aggregates. Over the culture period, there was a reduction in oestrogen receptor and membranous E-cadherin alongside loss of cell polarity, increased collective cell migration and enhanced angiogenic potential in co-cultures. These phenotypic alterations, typical of advanced stages of cancer, were not observed in the mono-cultures of MCF-7 cells. The proposed model system constitutes a new tool to study tumour-stroma crosstalk, disease progression and drug resistance mechanisms

ABSTRACT: This study compared the health status of Curraleiro cattle and other breeds (Nellore, Girolando, Guzera, and Caracu) for toxoplasmosis and neosporosis screening against anti-Neospora caninum and anti-Toxoplasma gondii antibodies, respectively. We used 119 female bovines of Curraleiro, Guzera, Nellore, Caracu, and Girolando breeds from four farms in Goias and Tocantins states and in the border of Goias, Minas Gerais, and Bahia states. All the farms had seropositive bovines for N. caninum. The number of seropositive Curraleiro cattle was significantly higher than the number of Guzera and Nellore cattle. Also, antibodies against toxoplasmosis were found in animals from all properties. There was no significant difference between the number of seropositive Curraleiro cattle and the breeds Nellore, Girolando, and Guzerá; however, it was significantly higher than the number of seropositive animals of Caracu breed

The aims of this study were to characterize the macrofauna associated with the articulated calcareous algae occurring on a hydrodynamic gradient off the Vitória metropolitan area, Espírito Santo State, Brazil, and to describe its temporal distribution. Physicochemical variables were measured in situ, and samples of sediment, water and phytal were taken from three sites along the coast in August and November 2005 (winter and spring, respectively) and in February and May 2006 (summer and fall, respectively). Twenty random samples were taken from the rocky surface in the lower midlittoral zone at each site, each season. Six species of articulated algae harboring 9,651 animal specimens distributed among 189 taxa were collected. The dominant taxonomic groups were Mollusca, Polychaeta and Crustacea. The main patterns revealed by statistical analysis related to differences in the abundance and richness at the sample sites, as well as to the correlation between the physicochemical variables and community distance matrices, indicating the importance of the physicochemical variables in the differentiation of the communities. Results also indicated that the variable which most influenced the physicochemical and community properties was the hydrodynamic character of the sites: a high level of hydrodynamism resulting in the calcareous algae´s sheltering a lower abundance and diversity of animal species than a lower level one.

--- - Label: BACKGROUND NlmCategory: BACKGROUND content: The presence of Plasmodium vivax malaria parasites in the human bone marrow (BM) is still controversial. However, recent data from a clinical case and experimental infections in splenectomized nonhuman primates unequivocally demonstrated the presence of parasites in this tissue. - Label: METHODS NlmCategory: METHODS content: In the current study, we analyzed BM aspirates of 7 patients during the acute attack and 42 days after drug treatment. RNA extracted from CD71+ cell suspensions was used for sequencing and transcriptomic analysis. - Label: RESULTS NlmCategory: RESULTS content: We demonstrated the presence of parasites in all patients during acute infections. To provide further insights, we purified CD71+ BM cells and demonstrated dyserythropoiesis and inefficient erythropoiesis in all patients. In addition, RNA sequencing from 3 patients showed that genes related to erythroid maturation were down-regulated during acute infections, whereas immune response genes were up-regulated. - Label: CONCLUSIONS NlmCategory: CONCLUSIONS content: This study thus shows that during P. vivax infections, parasites are always present in the BM and that such infections induced dyserythropoiesis and ineffective erythropoiesis. Moreover, infections induce transcriptional changes associated with such altered erythropoietic response, thus highlighting the importance of this hidden niche during natural infections
