    Cancer stem cells in prostate cancer: implications for targeted therapy

    Prostate cancer (PCa) is the most frequently diagnosed cancer in men and the second most common cause of cancer-related mortality among men in the developed world. Conventional anti-PCa therapies include surgery, radiation, hormonal ablation, and chemotherapy. Despite increasing efforts, these therapies are not effective for patients with advanced and/or metastatic disease. In most cases, cancer therapies fail due to an incomplete depletion of tumor cells, resulting in tumor relapse. The cancer stem cell (CSC) hypothesis is an emerging model that explains many of the molecular characteristics of oncological disease as well as the tendency of cancers to relapse, metastasize, and develop resistance to conventional therapies. CSCs are a reservoir of cancer cells that exhibit properties of self-renewal and the ability to reestablish the heterogeneous tumor cell population. The existence of PCa stem cells offers a theoretical explanation for many uncertainties regarding PCa and also for treatment resistance and disease progression once clinical cure is achieved. Therapies targeting CSCs might therefore lead to more effective cancer treatments, divergent from a traditional anti-proliferative approach, based on tumor bulk reduction accompanied by CSC-specific inhibition. Here, we focus on reviewing the historical perspective as well as concepts regarding stem cells and CSCs in PCa. In addition, we will report possible strategies and new clinical approaches that address the CSC-based concept of tumorigenesis in PCa. (C) 2017 S. Karger AG, Base

    Carcinoma de células escamosas com localização ocular em ovinos

    Os carcinomas de células escamosas (CCE) são tumores malignos, com origem nos queratinócitos, descritos em todos os animais domésticos1,2,4. Nos ovinos a sua incidência é muito rara1, podendo desenvolver-se no ouvido (localização mais frequente), plano nasal, região perianal, vulva, cauda e olho5. Os carcinomas de células escamosas do olho têm origem no epitélio da conjuntiva do limbus e pálpebras, incluindo a terceira pálpebra, chegando também a envolver a córnea 5,6. Esta neoplasia parece ter uma etiologia multifactorial, destacando-se como factores predisponentes raças com ausência de pigmentação cutânea periocular e habitat em áreas geográficas com exposição prolongada à luz solar, nomeadamente às radiações ultravioletas2,3. Há autores que atribuem também alguma importância à infecção pelo vírus da papilomatose5. Em ovinos o CCE foi assinalado em Marrocos, Austrália, Sul de França e Sul de Espanha2. Em Portugal não encontrámos dados publicados assinalando a sua presença nesta espécie. No presente trabalho descrevem-se dois casos de CCE, com localização ocular, em dois ovinos de refugo, do sexo feminino, adultos, de raça cruzada de Merino Branco, criados em extensivo na Herdade do Monte da Igreja, Aldeia do Freixo, Redondo

    Application of eurocodes 0 and 1 - comparison with sia 260 and 261 swiss standards

    The structural safety is one of the most important issues in construction. In order to ensure this safety there is a need to provide the appropriate carrying capacity to the structure. One of the initial stages of the design process is the quantification and combination of the different actions. Eurocodes 0 and 1 (EN 1990, EN 1991) are concerned by defining the basis of structural design and the actions that should normally be considered in the design of buildings and civil engineering works such as imposed load, wind and snow loads, and have brought a consistency of methodologies for structural design across European Union. Switzerland is one of the few countries in Europe which is not included in European Union, and that continues to follow internal standards, the SIA - Swiss Society of Engineers and Architects. Current investigation involved the comparative study of the requirements for safety and serviceability limit states, load combination format, and partial safety factors as well as the main actions to be considered for the structural design and verification. Both codes of practice are analyzed and compared for the different actions. An automatic calculation computer program was also developed during this study in order to estimate the values of the different actions. Finally, a real building was modelled so as to validate the above mentioned program and to analyze the main differences between European and Swiss codes.info:eu-repo/semantics/publishedVersio
