    Raman-based spectrophenotyping of the most important cells of the immune system

    INTRODUCTION: Human peripheral blood mononuclear cells (PBMCs) are a heterogeneous population of cells that includes T and B lymphocytes. The total number of lymphocytes and their percentage in the blood can be a marker for the diagnosis of several human diseases. Currently, cytometric methods are widely used to distinguish subtypes of leukocytes and quantify their number. These techniques use cell immunophenotyping, which is limited by the number of fluorochrome-labeled antibodies that can be applied simultaneously. OBJECTIVE: B and T lymphocytes were isolated from peripheral blood obtained from healthy human donors. METHODS: The immunomagnetic negative selection was used for the enrichment of B and T cells fractions, and their purity was assessed by flow cytometry. Isolated cells were fixed with 0.5% glutaraldehyde and measured using confocal Raman imaging. K-means cluster analysis, principal component analysis and partial least squares discriminant methods were applied for the identification of spectroscopic markers to distinguish B and T cells. HPLC was the reference method for identifying carotene in T cells. RESULTS: Reliable discrimination between T and B lymphocytes based on their spectral profile has been demonstrated using label-free Raman imaging and chemometric analysis. The presence of carotene in T lymphocytes (in addition to the previously reported in plasma) was confirmed and for the first time unequivocally identified as β-carotene. In addition, the molecular features of the lymphocytes nuclei were found to support the discriminant analysis. It has been shown that although the presence of carotenoids in T cells depends on individual donor variability, the reliable differentiation between lymphocytes is possible based on Raman spectra collected from individual cells. CONCLUSIONS: This proves the potential of Raman spectroscopy in clinical diagnostics to automatically differentiate between cells that are an important component of our immune system

    Experimental study of smog microphysical and optical vertical structure in the Silesian Beskids, Poland

    This study presents the vertical profiles of aerosol optical and microphysical properties obtained from cable car and ground-based measurements in the Silesian Beskids, Poland. The data were collected during a measurement campaign between 25 February and March 11, 2018. An AE-51 micro-aethalometer and PMS7003 and OPC-N2 optical particle counters were mounted on the cable car and used to measure the profiles of equivalent of black carbon (eBC) concentration and aerosol size distribution. In situ measurements of the optical properties of the aerosols were obtained using an AE-31 aethalometer and photoacoustic devices. A prototype lidar was used to determine the planetary boundary layer (PBL) height and the aerosol layers. In the middle phase of the study (1–6 March 2018), significant night-time temperature inversions were observed. During the inversion period, the parameters describing the amount of aerosols in the air increased significantly. The concentration of eBC exceeded the level of 15 μg/m3 several times, with an average level of 5.39 ± 4.42 μg/m3. Conversely, the results obtained in the first and third phases of the experiment were at the level of the aerosol background, being 1.45 ± 0.88 μg/m3 and 0.90 ± 0.95 μg/m3, respectively. Significant differences were also observed in the vertical profiles of PM10 mass and eBC concentration. In the middle phase of the study, the profiles showed a significant reduction in the concentration of pollutants with height, while in the first and third phases, there were slight variations with height

    Methane production by fermentation is a complex biochemical process, in which micromolecular organic substances are broken down by anaerobic bacteria into simple stabilized chemicals—mainly methane CH4 and carbon dioxide CO2. The organic matter of the slurry consists mainly of fats, proteins and carbohydrates. As a result of biochemical changes in the process of anaerobic decomposition, some of this matter is mineralized to simple chemical compounds. Cattle and pig husbandry offers enormous potential for useable biogas plant substrates. As a result of the constantly increasing amounts of animal husbandry products, and increasingly stringent environmental protection requirements aimed at reusing natural fertilizers, it is necessary to look for alternative processing methods. The need for efficiency in obtaining biogas from substrates (e.g., manure) was met by the laboratory stand presented in this article, for which the Polish patent No. 232200 was obtained. The new technology also allows leaching of the organic liquid, e.g., from manure, and subjecting it to methane fermentation. The solution allows the individual elements of the technological line that determine the fermentation process to be tested under laboratory conditions. It also allows testing of the substrates in terms of fermentation, to determine their physical and chemical characteristics, and then to characterize the fermentation process in terms of the quality and quantity of the resulting biogas and the quality of post-fermentation residues. Compressing biogas for local distribution was also proposed. As part of the research, using a laboratory stand, the organic matter was leached from manure, for the purpose of biogas production. In addition, the biogas yield from manure at varying degrees of maturity was assessed. The best properties in terms of biogas yield forecasting were demonstrated by manure composted for 4–8 weeks

In this research, an automeasuring hydraulic press, a thermo-camera and the Fluent ANSYS software were used. The results of laboratory simulations, and the effects of experiments conducted on shale rocks to determine permanent changes in compressive strength, are presented. As both frozen rocks and rocks returned to room temperature show diminished compressive strength. It is suggested that prior freezing of rocks can increase the efficiency of fracturing

    Development of innovative technological solutions in animal production should be associated with reduction of greenhouse gases, ammonia emission, and with rational disposal of natural fertilizers. The presented solutions and concepts of biogas acquisition and its disposal as well as a disposal of digestate mass place a need to improve the technological process before the science. The main aim of the presented solutions for the use of methane fermentation of a natural fertiliser in agricultural conditions is their utilization and energy acquisition, in particular for households and farms. When considering the issue of methane fermentation, one should include all favourable fertilization, energy aspects and firstly, the ecological ones. During the methane fermentation, substrates are stabilized due to removal of a large amount of carbon. The only elements that are removed from the system are evolving gases: CH4, CO2 and H2S. During the discussed process, the entire nitrogen is preserved in the organic or ammonia form.Rozwój innowacyjnych rozwiązań technologii w produkcji zwierzęcej powinien być skojarzony z ograniczeniem emisji gazów cieplarnianych, a także racjonalnym zagospodarowaniem nawozów naturalnych na cele nawozowe i energetyczne. Przedstawione rozwiązania i koncepcje pozyskania biogazu oraz jego zagospodarowania, a także zagospodarowania masy pofermentacyjnej stawiają przed nauką potrzebę doskonalenia procesu technologicznego. Głównym celem przedstawionych rozwiązań zastosowania fermentacji metanowej nawozu naturalnego w warunkach rolnictwa jest ich utylizacja, jak również pozyskanie energii, zwłaszcza dla gospodarstw rodzinnych i farmerskich. Rozpatrując zagadnienie fermentacji metanowej, należy uwzględnić wszystkie korzystne aspekty nawozowe, energetyczne, a przede wszystkim ekologiczne. Podczas fermentacji metanowej substraty podlegają stabilizacji w wyniku usunięcia dużej ilości węgla. Jedynymi usuwanymi z systemu składnikami są wydzielające się gazy: CH4, CO2 i H2S.     Modern livestock facilities necessary in the production of milk, meat or other animal products should be constructed with environmental protection in mind, while ensuring high quality of production and animal welfare. The high level of mechanization in modern dairy farms, including automated and robotic processes, allows obtaining high quality raw material (e.g. milk), and significantly increasing labor and production efficiency. In addition, the use of photovoltaic (PV) panels, heat recovery from milk and obtaining biogas from the manure fermentation process, contributes to large energy savings on the farm. Excess of natural fertilizers, which are an animal byproduct, can be used as a substrate for methane fermentation. The presented examples of obtaining renewable energy allow improving the economic efficiency of animal production. They also ensure appropriate environmental conditions through the innovative management of natural fertilizers.Nowoczesne obiekty inwentarskie niezbędne w procesie produkcji mleka, mięsa lub innych produktów, powinny uwzględniać uwarunkowania związane z ochroną środowiska przy zapewnieniu wysokiej jakości produkcji oraz uwzględnieniu dobrostanu zwierząt.     Przeprowadzona analiza stanu przetwórstwa zbóż i produkcji pasz na potrzeby zwierząt gospodarskich w regionie Kirowa (Rosja) wykazała wysoką efektywność wykorzystania pasz zbożowych (ziarno rozdrabniane, suche i mokre) w diecie zwierząt. Opracowano technologię frakcyjną gniecenia mokrego ziarna paszowego w punktach przetwarzania ziarna po zbiorach i obliczono skuteczność jej zastosowania zamiast tradycyjnej technologii suszenia, sortowania i rozdrabniania ziarna paszowego po zbiorze, co wykazało, że roczny efekt nowej technologii frakcyjnej wynosi 2 059 840 rubli. Opracowano schemat konstrukcyjno-technologiczny dwustopniowego gniotownika ziarna, przeprowadzono teoretyczne badania jego procesu roboczego, które umożliwiły określenie racjonalnych parametrów projektowych elementów roboczych. Zgodnie z opracowanym schematem wyprodukowano gniotownik ziarna PZD-3, przeprowadzono jego wstępne testy, które wykazały zgodność deklarowanych parametrów z wymaganiami. Na podstawie wyników testu instytucja państwowa zaleciła kontynuację testowania dwustopniowego gniotownika PZD-3 po wyeliminowaniu stwierdzonych braków. W procesie rekonstrukcji kompleksu do czyszczenia i suszenia ziarna KZS-20Sh zastosowano technologię frakcyjną zgniatania ziarna paszowego i dwustopniowy gniotownik ziarna PZD-3. Zastosowanie tej technologii przetwarzania ziarna znacznie zmniejsza zużycie energii do suszenia (frakcja ziarna paszowego nie jest suszona, ale jest stosowana jako pasza dla zwierząt w formie zgniecionej), wydajność kompleksu zależy od frakcji ziarna.An analysis of the state of post-harvest grain processing and feed production for livestock needs in the Kirov region (RF), which showed the high efficiency of the use of grain feed in animal diet – crushed grain, dry and wet. The fractional technology for flattening (crushing) wet feed grain at the post-harvest grain processing points was developed, and the effectiveness of its application instead of the traditional technology for post-harvest drying, sorting and crushing of feed grain was calculated, which showed that the annual effect of the new fractional technology is 2 059 840 rubles. For the technology of grain crushing, a constructive-technological scheme of a two-stage grain crusher was developed, theoretical studies of its working process were carried out, which made it possible to determine the rational design parameters of the working elements. According to the developed scheme, the PZD-3 grain crusher was manufactured, its preliminary tests were carried out, which showed the compliance of the claimed parameters with the requirements. Based on the test results of the federal institution recommended continuing the testing of the PZD-3 two-stages crusher after eliminating the identified deficiencies. The fractional technology for crushing feed grain and the two-stage crusher for grain PZD-3 as part of this technology were used in the reconstruction of the grain cleaning and drying complex KZS-20Sh. The use of this technology of processing of grain significantly reduces the energy consumption for drying (the feed grain fraction is not dried, but is used for animal feed in crushed form), the productivity of the complex depends in proportion to the allocated feed fraction
