Entwicklung und klinische Anwendung neuartiger retroviraler Vektorsysteme fuer die somatische Gentherapie Abschlussbericht

Retroviral vectors for the singular expression of the receptor molecule LNGFR on hematopoietic cells for gene marking and also vectors for the co-expression of LNGFR and HSV TK were developed within this subproject at Boehringer Mannheim GmbH. GMP production and toxicology studies were performed, clinical protocols were established. The planned gene marking study was not performed as no new data in comparison to the literature were expected at that time. The LNGF/TK vectors for the induction of GvL effects in leukemia/lymphoma patients with allogeneic T cells and the treatment of GvHD induced by the allogeneic cells were fully developed for clinical use. Crucial data were produced for the establishment of the production of retroviral vectors in cells growing in suspension. Approaches utilizing the mdr-1 coding vectors for the protection of stem cells were not pursued because of considerable doubts with respect to safety and scientific rationale. Substances have been separated from fungi by organic solvent extraction. In the case of biologic activity, isolation was carried out mainly by fermentation and preparative chromatography. Purity and molecular structure were determined by spectroscopic methods. Numerous drugs have been found which exhibit pharmacological activity and are of interests in the fields of allergy, immunology, thrombocyte aggregation, inotropy and vascular relaxation, disordered fat-metabolism and as antiretrovitale substances. By immunization of sheeps high-sensitive polyclonal cocaine antibodies were obtained. A corresponding development with mice resulted in considerably lower sensitivity. Using the anti-cocaine antibodies, a reference analytical system in micro titer plate scale (MTP-ELISA) for gas phase analysis has been designed with a detection limit of 500 fg. For cocaine and cannabinoide concentration by adsorption from the gas phase newly developed hydrophilic cellulose materials were applied. Preliminary tests have been carried out with luminescence detection and carrier - immobilized antibodies.
