    Biogeochemical significance and biomarker potential of novel glycerolipids and respiratory quinones in the marine environment

    Microorganisms catalyze processes which shape the chemistry of the biosphere, drive global element cycles, and ultimately influence the composition of EarthĂą s surface and interior on geological timescales. Microbial lipids provide crucial information about the distribution, activity, and adaptation of microbial communities in both modern and ancient environments. In this thesis, advanced chromatographic methods were developed that have significantly expanded the analytical window for microbial lipidomics in complex environmental sample matrices and have paved the way for the discovery of novel microbial biomarkers. Application of the novel protocols to samples from microbial cultures, the hypersaline Discovery Basin and the stratified water column and sediments of the Black Sea revealed an exceptional diversity of lipids. The combination of respiratory quinone and core and intact polar lipid analysis provided detailed information about microbial community composition and biogeochemical processes the microbes are mediating

    Detection of Diatomic Carbon in 2I/Borisov

    2I/Borisov is the first-ever observed interstellar comet (and the second detected interstellar object (ISO)). It was discovered on 2019 August 30 and has a heliocentric orbital eccentricity of ~3.35, corresponding to a hyperbolic orbit that is unbound to the Sun. Given that it is an ISO, it is of interest to compare its properties—such as composition and activity—with the comets in our solar system. This study reports low-resolution optical spectra of 2I/Borisov. The spectra were obtained by the MDM Observatory Hiltner 2.4 m telescope/Ohio State Multi-Object Spectrograph (on 2019 October 31.5 and November 4.5, UT). The wavelength coverage spanned from 3700 to 9200 Å. The dust continuum reflectance spectra of 2I/Borisov show that the spectral slope is steeper in the blue end of the spectrum (compared to the red). The spectra of 2I/Borisov clearly show CN emission at 3880 Å, as well as C2 emission at both 4750 and 5150 Å. Using a Haser model to covert the observed fluxes into estimates for the molecular production rates, we find Q(CN) = 2.4 ± 0.2 × 10ÂČ⁎ s⁻Âč, and Q(C₂) = (5.5 ± 0.4) × 10ÂČÂł s⁻Âč at the heliocentric distance of 2.145 au. Our Q(CN) estimate is consistent with contemporaneous observations, and the Q(C₂) estimate is generally below the upper limits of previous studies. We derived the ratio Q(C₂)/Q(CN) = 0.2 ± 0.1, which indicates that 2I/Borisov is depleted in carbon-chain species, but is not empty. This feature is not rare for the comets in our solar system, especially in the class of Jupiter-family comets

    Unusual butane- and pentanetriol-based tetraether lipids in Methanomassiliicoccus luminyensis, a representative of the seventh order of methanogens

The definitive version was published in Applied and Environmental Microbiology 82 (2016): 4505-4516, doi:10.1128/AEM.00772-16.A new clade of archaea has recently been proposed to constitute the seventh methanogenic order, the Methanomassiliicoccales, which is related to the Thermoplasmatales and the uncultivated archaeal clades deep-sea hydrothermal vent Euryarchaeota group 2 and marine group II Euryarchaeota but only distantly related to other methanogens. In this study, we investigated the membrane lipid composition of Methanomassiliicoccus luminyensis, the sole cultured representative of this seventh order. The lipid inventory of M. luminyensis comprises a unique assemblage of novel lipids as well as lipids otherwise typical for thermophilic, methanogenic, or halophilic archaea. For instance, glycerol sesterpanyl-phytanyl diether core lipids found mainly in halophilic archaea were detected, and so were compounds bearing either heptose or methoxylated glycosidic head groups, neither of which have been reported so far for other archaea. The absence of quinones or methanophenazines is consistent with a biochemistry of methanogenesis different from that of the methanophenazine-containing methylotrophic methanogens. The most distinctive characteristic of the membrane lipid composition of M. luminyensis, however, is the presence of tetraether lipids in which one glycerol backbone is replaced by either butane- or pentanetriol, i.e., lipids recently discovered in marine sediments. Butanetriol dibiphytanyl glycerol tetraether (BDGT) constitutes the most abundant core lipid type (>50% relative abundance) in M. luminyensis. We have thus identified a source for these unusual orphan lipids.     Microarray and pathway analysis reveals decreased CDC25A and increased CDC42 associated with slow growth of BCL2 overexpressing immortalized breast cell line

    Bcl-2 is an anti-apoptotic protein that is frequently overex-pressed in cancer cells but its role in carcinogenesis is not clear. We are interested in how Bcl-2 expression affects non-cancerous breast cells and its role in the cell cycle. We prepared an MCF10A breast epithelial cell line that stably overexpressed Bcl-2. We analyzed the cells by flow cytometry after synchronization, and used cDNA microarrays with quantitative reverse-transcription PCR (qRTPCR) to determine differences in gene expression. The microarray data was subjected to two pathway analysis tools, parametric analysis of gene set enrichment (PAGE) and ingenuity pathway analysis (IPA), and western analysis was carried out to determine the correlation between mRNA and protein levels. The MCF10A/Bcl-2 cells exhibited a slow-growth phenotype compared to control MCF10A/Neo cells that we attributed to a slowing of the G1-S cell cycle transition. A total of 363 genes were differentially expressed by at least two-fold, 307 upregulated and 56 downregulated. PAGE identified 22 significantly changed gene sets. The highest ranked network of genes identified by IPA contained 24 genes. Genes that were chosen for further analysis were confirmed by qRT-PCR, however, the western analysis did not always confirm differential expression of the proteins. Downregulation of the phosphatase CDC25A could solely be responsible for the slow growth pheno-type in MCF10A/Bcl-2 cells. Increased levels of GTPase Cdc42 could be adding to this effect. PAGE and IPA are valuable tools for microarray analysis, but protein expression results do not always follow mRNA expression results. Originally published Cell Cycle, Vol. 7, No. 19, Oct. 200
