Identification of Ion Transport Compartments in Turtle Urinary Bladder
To identify the turtle urinary bladder cells involved in Na and Cl absorption and Hand HCO3 secretion cellular electrolyte concentrations and uptake of Br and Solutrast were determined using electron microprobe analysis. Whereas inhibition of transepithelial Na transport by ouabain (reversion of short circuit current) led to a pronounced K-Na exchange in granular, and most of the basal cells, surface CA-cells and some basal cells were ouabain insensitive. Surface CA-cells could be divided into a large Cl-rich and a small Cl-poor population. Since the ouabain-induced K-Na exchange could be completely prevented by blocking passive luminal Na entry by amiloride, granular and most of the basal cells seem to form a syncytial Na transport compartment. Luminal uptake of Br only occurred in Cl-poor surface CA-cells, indicating the sole responsibility of these cells for electrogenic and electroneutral Cl absorption and HCO3 secretion.
Serosal Br was taken up into all cell types. Whereas H secretion and serosal Br uptake into all cell types could be inhibited by 4-isothiocyano-4\u27-acetamido-2,2\u27-disulfonic stilbene (SITS), blockade of H secretion by lowering luminal pH to 4.5 diminished Br uptake only in Cl-rich surface CA-cells. Theses results indicate: a) Only Cl-rich surface CA-cells have a serosal anion exchanger involved in H secretion and b) granular and basal cells also possess a serosal anion exchanger, possibly responsible for cellular pH regulation.
Luminal endocytosis of the I-containing Solutrast was observed in apical regions of Cl-rich surface CA-cells after inhibition of H secretion, but not under steady-state conditions, indicating a transport related but not a constitutive endo-exocytosis
Computationally efficient methods for modelling laser wakefield acceleration in the blowout regime
Electron self-injection and acceleration until dephasing in the blowout
regime is studied for a set of initial conditions typical of recent experiments
with 100 terawatt-class lasers. Two different approaches to computationally
efficient, fully explicit, three-dimensional particle-in-cell modelling are
examined. First, the Cartesian code VORPAL using a perfect-dispersion
electromagnetic solver precisely describes the laser pulse and bubble dynamics,
taking advantage of coarser resolution in the propagation direction, with a
proportionally larger time step. Using third-order splines for macroparticles
helps suppress the sampling noise while keeping the usage of computational
resources modest. The second way to reduce the simulation load is using
reduced-geometry codes. In our case, the quasi-cylindrical code CALDER-CIRC
uses decomposition of fields and currents into a set of poloidal modes, while
the macroparticles move in the Cartesian 3D space. Cylindrical symmetry of the
interaction allows using just two modes, reducing the computational load to
roughly that of a planar Cartesian simulation while preserving the 3D nature of
the interaction. This significant economy of resources allows using fine
resolution in the direction of propagation and a small time step, making
numerical dispersion vanishingly small, together with a large number of
particles per cell, enabling good particle statistics. Quantitative agreement
of the two simulations indicates that they are free of numerical artefacts.
Both approaches thus retrieve physically correct evolution of the plasma
bubble, recovering the intrinsic connection of electron self-injection to the
nonlinear optical evolution of the driver
Effect of zinc supplementation on respiratory tract infections in children with cystic fibrosis
Zinc (Zn) has significant anti-oxidant and anti-inflammatory activity. Zn deficiency can occur in subsets of patients with cystic fibrosis (CF) especially those with malabsorption and impaired growth. Although supplemental Zn has significantly reduced infections in various disorders, its efficacy has not been thoroughly investigated in CF. We performed a double blind placebo controlled pilot study to investigate the effect of daily 30 mg elemental Zn for 1 year on the rate of respiratory tract infections (RTIs), use of antibiotics and plasma cytokines in 26 children with CF (ages 7–18 years). Plasma Zn, Cu, inflammatory cytokines and ex vivo generation of IL-2 were measured at baseline and at the end of the study. The number of days of oral antibiotics was lower in Zn treated patients compared to placebo ( P = 0.05). However, compared to placebo, the effect of Zn was greater in patients who exhibited low plasma Zn at baseline ( P = 0.02) than those who had plasma Zn levels identical to normal subjects ( P = 0.55). Zn supplementation was marginally effective in reducing percentage increase in plasma IL-6 and IL-8 while increasing the percentage change in ex vivo generation of IL-2 in isolated mononuclear cell. In conclusion, oral intake of 30 mg/day of Zn reduced the number of days of oral antibiotics used to treat RTIs in children with CF. A higher daily Zn dose may be needed to decrease RTIs and modify immune responses. Electron Microprobe Analysis of Electrolytes in Whole Cultured Epithelial Cells
Microprobe analysis was used to determine electrolyte contents in whole epithelial sheets of A6 cells and to investigate the most critical points of this method. Analysis of dextran standard sections of different thickness revealed that low accelerating voltages of about 10 kV are best suited for whole freeze-dried cells on thick supports, since 5 μm thick sections are not penetrated by 10 kV electrons. Washing of A6 cells for 10 sec with distilled water led to cell swelling of about 40%, but the molar concentration ratios and the concentrations per dry weight (dw) were not altered. Washing for 60 sec with distilled water caused a further increase in cell volume (120%) and loss of cellular K and Cl (90 mmol/kg dw). Washing with isotonic NH4-acetate led to a loss of cell Cl already after 10 sec.
To characterize the Na transport compartment, A6 cells cultured on permeable supports were washed for 5 sec with distilled water, freeze-dried, and analyzed. Inhibition of transepithelial Na transport by ouabain increased Na/P from 0.15±0.07 to 0.75±0.03 and Cl/P from 0.21±0.001 to 0.38±0.003 while KIP decreased from 0.83±0.08 to 0.32±0.03. The changes in cell Na and K contents can be explained by K/Na exchange; the increase in Cl content indicates some cell swelling. Since the ouabain-induced changes could be prevented by apical amiloride, the apical membrane provides the most important pathway for Na entry in A6 cells
On a generalised model for time-dependent variance with long-term memory
The ARCH process (R. F. Engle, 1982) constitutes a paradigmatic generator of
stochastic time series with time-dependent variance like it appears on a wide
broad of systems besides economics in which ARCH was born. Although the ARCH
process captures the so-called "volatility clustering" and the asymptotic
power-law probability density distribution of the random variable, it is not
capable to reproduce further statistical properties of many of these time
series such as: the strong persistence of the instantaneous variance
characterised by large values of the Hurst exponent (H > 0.8), and asymptotic
power-law decay of the absolute values self-correlation function. By means of
considering an effective return obtained from a correlation of past returns
that has a q-exponential form we are able to fix the limitations of the
original model. Moreover, this improvement can be obtained through the correct
choice of a sole additional parameter, . The assessment of its validity
and usefulness is made by mimicking daily fluctuations of SP500 financial

Measurement protocols and determinants of peak oxygen consumption in adults with Down syndrome:a systematic review
This systematic review aimed to provide an overview of test protocols used to measure peak oxygen uptake (VO2peak) in adults with Down syndrome (DS) and to determine how generalisable the outcomes are for the entire population of adults with DS by describing the sample characteristics of these studies and their impact on VO2peak. A literature search (PROSPERO CRD42022309560) was performed (18 July 2023) using the following databases: PubMed, CINAHL, APA PsycINFO, Web of Science, Embase and SPORTDiscus. For articles to be included, they had to be peer-reviewed pubications, reporting VO2peak or VO2max for individuals with DS separately, with a sample of n ≥ 5 and a mean age ≥18 years. Systematic reviews and meta-analyses were excluded but their reference lists were searched for additional papers to include. Studies were evaluated for risk of bias following the guidelines of Kmet et al. The results were summarised with frequency statistics. Forty-three studies were included in this systematic review. Sample sizes of included adults with DS ranged from n = 4–226, with a total of n = 1498 adults with DS being included. Most studies (29/43) used the same standardised maximal exercise treadmill protocol to measure VO2peak in adults with DS, and 33 out of 43 studies used at least one objective criterion to determine a valid maximal effort. Participants were predominantly male, under 40 years old, and overweight or obese. Additionally, the diversity of study samples was lacking or not reported. The most widely used, standardised, maximal exercise test treadmill protocol is recommended for future use in research and practice, including objective criteria to determine valid maximal effort. The current study samples are not representative of the population of adults with DS in terms of sex, age and diverse backgrounds and therefore likely overestimate VO2peak of this population.</p
Hard Photodisintegration of a Proton Pair
We present a study of high energy photodisintegration of proton-pairs through
the gamma + 3He -> p+p+n channel. Photon energies from 0.8 to 4.7 GeV were used
in kinematics corresponding to a proton pair with high relative momentum and a
neutron nearly at rest. The s-11 scaling of the cross section, as predicted by
the constituent counting rule for two nucleon photodisintegration, was observed
for the first time. The onset of the scaling is at a higher energy and the
cross section is significantly lower than for deuteron (pn pair)
photodisintegration. For photon energies below the scaling region, the scaled
cross section was found to present a strong energy-dependent structure not

Immunolocalization of dually phosphorylated MAPKs in dividing root meristem cells of Vicia faba, Pisum sativum, Lupinus luteus and Lycopersicon esculentum

constitutive nuclear localization; however, not all
studied plant species show co-localization of activated
MAPKs to mitotic microtubules.
Abstract The mitogen-activated protein kinase (MAPK)
signaling pathway is involved not only in the cellular
response to biotic and abiotic stress but also in the regulation
of cell cycle and plant development. The role of
MAPKs in the formation of a mitotic spindle has been
widely studied and the MAPK signaling pathway was
found to be indispensable for the unperturbed course of cell
division. Here we show cellular localization of activated
MAPKs (dually phosphorylated at their TXY motifs) in
both interphase and mitotic root meristem cells of Lupinus
luteus, Pisum sativum, Vicia faba (Fabaceae) and Lycopersicon esculentum (Solanaceae). Nuclear localization
of activated MAPKs has been found in all species. Colocalization
of these kinases to mitotic microtubules was
most evident in L. esculentum, while only about 50 % of
mitotic cells in the root meristems of P. sativum and V.
faba displayed activated MAPKs localized to microtubules
during mitosis. Unexpectedly, no evident immunofluorescence
signals at spindle microtubules and phragmoplast
were noted in L. luteus. Considering immunocytochemical
analyses and studies on the impact of FR180204 (an
inhibitor of animal ERK1/2) on mitotic cells, we hypothesize
that MAPKs may not play prominent role in the
regulation of microtubule dynamics in all plant species
Large scale quantum simulations: C_60 impacts on a semiconducting surface
We present tight binding molecular dynamics simulations of C_60 collisions on
the reconstructed diamond(111) surface, carried out with an O(N) method and
with cells containing 1140 atoms. The results of our simulations are in very
good agreement with experiments performed under the same impact conditions.
Furthermore our calculations provide a detailed characterization of the
microscopic processes occuring during the collision, and allow the
identification of three impact regimes, as a function of the fullerene incident
energy. Finally, the study of the reactivity between the cluster and the
surface gives insight into the deposition mechanisms of C_60 on semiconducting
substrates
