    Moderate exercise increases affinity of large very low density lipoproteins for hydrolysis by lipoprotein lipase

    Context: Postprandial triglyceride (TG) concentration is independently associated with cardiovascular disease risk. Exercise reduces postprandial TG concentrations but the mechanisms responsible are unclear. Objective: To determine the effects of exercise on affinity of chylomicrons, large very low density lipoproteins (VLDL1) and smaller VLDL (VLDL2) for lipoprotein lipase (LPL) mediated TG hydrolysis. Design: Within-participant cross-over study. Setting: A University metabolic investigation unit. Participants: Ten overweight/obese men. Interventions: Participants undertook two oral fat tolerance tests, separated by 7–14 days, in which they had blood taken fasting and for 4 hours after a high-fat mixed meal. On the afternoon before one test, they performed a 90-minute treadmill walk at 50% maximal oxygen uptake (EX); no exercise was performed before the control test (CON). Main outcome measures: Circulating TG-rich lipoprotein concentrations; affinity of chylomicrons, VLDL1, VLDL2 for LPL-mediated TG hydrolysis. Results: Exercise significantly reduced fasting VLDL1-TG concentration (CON: 0.49(0.33–0.72) mmol.l−1, EX: 0.36(0.22–0.59) mmol.l−1, [geometric means (95% confidence interval)]; p=0.04). Time-averaged postprandial chylomicron-TG (CON: 0.55±0.10 mmol.l−1, EX: 0.39±0.08 mmol.l−1, [mean±SEM], p=0.03) and VLDL1-TG (CON: 0.85±0.13 mmol.l−1, EX: 0.66±0.10 mmol.l−1, p=0.01) concentrations were both lower in EX than CON. Affinity of VLDL1 for LPL-mediated TG hydrolysis increased by 2.2(1.3–3.7) fold (geometric mean (95% confidence interval)) (p=0.02) in the fasted state and 2.6(1.8–2.6) fold (p=0.001) postprandially. Affinity of chylomicrons and VLDL2 was not significantly different between trials. Conclusions: Exercise increases affinity of VLDL1 for LPL-mediated TG hydrolysis both fasting and postprandially. This mechanism is likely to contribute to exercise's TG-lowering effect

    Xylanase and xylo- oligosaccharide prebiotic improve the growth performance and concentration of potentially prebiotic oligosaccharides in the ileum of broiler chickens

    The objective of this study was to investigate the effect of supplementing broiler diets with xylanase or xylo- oligosaccharide (XOS) on growth performance, the concentration of non-starch polysaccharide (NSP) hydrolysis products in the ileum and concentration of short chain fatty acids (SCFA) in the caeca of broiler chickens. In total, 500 male Ross 308 broilers were used in this 29-day (d) study. The treatments were organised into a 2×2 plus 1 factorial arrangement consisting of two additives (xylanase or XOS) at two levels (low or high) plus a control treatment with no additives. This gave five treatments with 100 bird in each treatment group. The diets were slightly deficient in protein by 20 g/kg and energy by 1 MJ/kg. On d 14 and 28, two birds per pen were euthanised, the caeca content collected and analysed for short chain fatty acid (SCFA) concentration. On d 29, six birds per pen were euthanised and ileal digesta were collected and analysed for the concentration of NSP fractions. On d 14, caecal acetic acid, iso-butyric acid, iso-valeric acid, n-valeric acid and total SCFA concentrations were significantly greater (P≤0.05) when diets were supplemented with XOS compared with xylanase. Ileal concentration of arabinose, galactose and glucuronic acid (GlucA2) were significantly greater (P≤0.05) in the insoluble NSP fraction when diets were supplemented with a high level of xylanase, compared with the control treatment. Ileal concentration of fructose was significantly greater (P≤0.05) in the water soluble NSP when a high level of xylanase or low level of XOS were included in the diet compared with the control. It was concluded that xylanase and XOS had similar effects on NSP concentration and SCFA in the caeca, although there was little effect on performance. This observation demonstrated further benefits of xylanase supplementation in wheat-based broiler diets beyond digesta viscosity reduction and the release of extra nutrients

    Wheat sample affects growth performance and the apparent metabolisable energy value for broiler chickens

    The aim of this study was to examine chemical composition, quality characteristics, apparent metabolisable energy (AME) and nutrient utilisation of wheat samples currently available to the UK poultry industry and their effect on broiler growth performance. Seventeen current UK wheat samples were used to formulate 17 diets, all of which included 670 g/kg of each wheat sample and 330 g/kg of a balancer feed. Eight hundred (800), day old male Ross 308 broilers were allocated randomly to 160 raised floor pens. Each diet was replicated eight times, fed ad libitum from 0 to 21d age in a randomised complete block design. Excreta were quantitatively collected during the last three days for AME determination. The content of protein, ash and gross energy (GE) ranged from 97 to 143 g/kg DM, 12.8 to 19.6 g/kg DM and 17.81 to 18.24 MJ/kg DM, respectively. The amount of starch and total non-starch polysaccharides (NSP) ranged from 671 to 728 and 80.1 to 98.2 g/kg DM, respectively. The quality characteristics of wheat samples were in the expected range. There were differences (P<0.05) in AME and AMEn of wheat samples. The AME of the wheat had a maximum range of 1.13 MJ/kg DM between samples. Dry matter retention (DMR) and fat digestibility (FD) were significantly different (P<0.05) between wheat samples. The daily feed intake (FI) and weight gain (WG) of broilers fed two wheat samples were significantly (P<0.05) lower as compared to other samples and their low FI and WG were not related to their chemical composition and quality characteristics. The ash content of wheat samples was negatively associated with AMEn (r = − 0.489, P<0.05). The coefficient of FD was positively related to AMEn (r = 0.552, P<0.05). Chemical composition and quality characteristics of the wheat did not relate (P>0.05) to FI and WG of broilers. There was no relationship between growth performance of broilers and AMEn of the wheat samples

    The effects of supplementary bacterial phytase on dietary energy and total tract amino acid digestibility when fed to young chickens

    1. Four diets were offered to broiler chickens from 7 to 17 d of age; these included a phosphorus-adequate positive control (PC) (4·7 g/kg available P), a sub-optimal P negative control (NC, 2·5 g/kg available P) with (500 and 12500 FTU/kg) and without phytase. Dietary apparent metabolisable energy (AME), dietary net energy for production (NEp), the efficiency of AME retention (Kre), heat production and total tract amino acid digestibility coefficients were determined. The determination of NEp involved a comparative slaughter technique in which growing chickens were fed the experimental diets ad libitum. 2. Feed intake, weight gain and feed conversion efficiency increased significantly in a dose dependent manner in response to dietary phytase activity. Overall, the NEp of the phytase supplemented diets significantly improved by approximately 15·6% compared with the negative control, while dietary AME was unaffected. Although phytase did not affect AME, the large increase in the NEp demonstrated that dietary phytases improves energy utilisation, i.e. diverting more energy, not accounted for in the AME procedure, for production. This is largely a result of the stimulatory effect that phytase has on feed intake rather than on digestibility of the diet. 3. Overall, the diet supplemented with 12500 FTU had 6·4% significant improvement in total tract digestibility coefficients of the total amino acids compared with the negative control. With regard to individual amino acids, the impact of phytase was far more pronounced for threonine, an important component of the gastrointestinal mucin, than for other amino acids. 4. Dietary NEp was more highly correlated with performance criteria than dietary AME and seems to be a more sensitive way to evaluate broiler response to phytase supplementation

    Sequence-dependent structure/function relationships of catalytic peptide-enabled gold nanoparticles generated under ambient synthetic conditions

    YesPeptide-enabled nanoparticle (NP) synthesis routes can create and/or assemble functional nanomaterials under environmentally friendly conditions, with properties dictated by complex interactions at the biotic/abiotic interface. Manipulation of this interface through sequence modification can provide the capability for material properties to be tailored to create enhanced materials for energy, catalysis, and sensing applications. Fully realizing the potential of these materials requires a comprehensive understanding of sequence-dependent structure/function relationships that is presently lacking. In this work, the atomic-scale structures of a series of peptide-capped Au NPs are determined using a combination of atomic pair distribution function analysis of high-energy X-ray diffraction data and advanced molecular dynamics (MD) simulations. The Au NPs produced with different peptide sequences exhibit varying degrees of catalytic activity for the exemplar reaction 4-nitrophenol reduction. The experimentally derived atomic-scale NP configurations reveal sequence-dependent differences in structural order at the NP surface. Replica exchange with solute-tempering MD simulations are then used to predict the morphology of the peptide overlayer on these Au NPs and identify factors determining the structure/catalytic properties relationship. We show that the amount of exposed Au surface, the underlying surface structural disorder, and the interaction strength of the peptide with the Au surface all influence catalytic performance. A simplified computational prediction of catalytic performance is developed that can potentially serve as a screening tool for future studies.     Influence of the in vivo method and basal dietary ingredients employed in the determination of the amino acid digestibility of wheat distillers dried grains with solubles in broilers

    As distillers dried grains with solubles (DDGS) become increasingly available, it is important to determine their nutritional value for precise feed formulation. The accurate determination of digestibility is crucial, and it is known that the methods used will affect the values obtained. An experiment was designed to determine and compare the standardized ileal digestibility (SID) of amino acids from wheat DDGS using a semisynthetic diet and a difference method using 4 further diets based on corn, wheat, corn DDGS, and wheat DDGS. Eighty 1-d-old male broilers were fed a commercial starter diet until d 21. Between d 21 and 23, they were fed test diets in order to adapt to those diets before the trial took place between d 24 and 27. The trial period took place between d 24 and 27. Feed intake was measured, excreta collected, and at d 27, all birds were culled and ileal digesta was collected for the determination of apparent ileal digestibility and SID of amino acids. Values determined were similar to those reported elsewhere in the literature, although SID values for lysine were particularly low, being 0.26, 0.27, or 0.32, measured in semisynthetic, corn, or wheat diet backgrounds, respectively. It appeared that diet type employed was influential in the values obtained. The SID values for methionine, cysteine, methionine plus cysteine, and arginine were significantly lower (P < 0.05) when measured in semisynthetic diet backgrounds than wheat- or corn-based diets. It appears that dextrose and possibly purified starch have a detrimental impact on the broiler digestive tract. This may affect all digestibility methodologies in which such a diet base is used

    Effect of age on the relationship between metabolizable energy and digestible energy for broiler chickens

    A total of 960 male Ross 308 chicks (day-old) were used to investigate the effect of age on the relationship between metabolizable energy (ME) and digestible energy (DE) for broiler chickens. Bird growth variables, nitrogen retention (NR), nitrogen digestibility (ND), as well as the relative weight of liver, pancreas, and the gastrointestinal tract were determined. Practical diets that compared 2 cereals (corn and wheat) and exogenous xylanase (0 or 16,000 BXU/kg) were evaluated at 5 ages (7, 14, 21, 28, and 35 D) in a 2 × 2 × 5 factorial arrangement of treatments with 8 replicates per treatment and started with 30 birds per replicate. A randomized block ANOVA analysis of repeated measures was performed, and a 2 × 2 × 5 factorial structure was used to investigate the 2 dietary treatment factors (cereal type and the presence of xylanase) within the 5 bird ages (7, 14, 21, 28, and 35 D), and their interactions. Apparent metabolizable energy (AME) increased linearly from 7 until 28 D of age, but (P 0.05) from 14 to 35 D of age. The AME: DE ratio was lowest (P 0.05) differences thereafter. Cereal type and xylanase supplementation did not (P > 0.05) change the ME: DE ratio. The results indicate that determining ME before 14 D of age may give absolute values that are lower than would be obtained with older birds. ME values that are determined on older broiler chickens may overestimate the energy availability of practical feeds used in broiler starter feeds

    The effect of dietary calcium inclusion on broiler gastrointestinal pH: quantification and method optimization

    There is little consensus as to the most appropriate methodology for the measurement of gastrointestinal pH in chickens. An experiment was conducted to establish the optimum sampling method for the determination of broiler digesta pH in birds fed differing levels of dietary calcium. Ross 308 broilers (n = 60) were fed one of two experimental diets, one containing 0.8% monocalcium phosphate and 2% limestone and one containing 0.4% monocalcium phosphate and 1% limestone. Four factors were investigated to determine the most appropriate method of measuring broiler gastrointestinal digesta pH: removal from the tract, prolonged air exposure, altering the temperature of the assay, and controlling the water content of the digesta. The conditions were assessed at bird ages from 7 to 42 d post hatch. Dietary Ca content had no significant effect on in situ pH, but it contributed towards variance in ex situ pH of both gizzard and duodenum digesta

    Broiler chicken response to xylanase and fermentable xylooligosaccharide supplementation

    A study was conducted to determine the effect of dietary fiber (DF), xylanase (XYL), xylooligosaccharides (XOS), and a combination of XYL and xylooligosaccharides (STBIO) on chicken growth performance, N-corrected apparent metabolizable energy (AMEn), and nutrient availability, characteristics of the gastrointestinal tract (GIT), and cecal content of short-chain fatty acids (SCFA). A 35-day experiment was performed on 1,920 as hatched Ross 308 broiler chicks, reared in 96 pens and fed ad libitum. Experimental diets were split into 2 phases: starter (0−21 d) and finisher (22−35 d). There were 2 basal diets, first contained 54% maize and in the second, 5% of the maize was replaced by wheat bran as DF. The diets were split into 4 batches: one of them was used as a control, and each of the others were supplemented either with XYL or XOS or with the STBIO. Each diet was fed to 12 pens following randomization. The data were analyzed in GenStat (20th edition) by ANOVA using a 2 £ 4 factorial design. The addition of STBIO improved feed conversion ratio (FCR) and increased weight gain (WG) from 21 to 35 d and from 0 to 35 d (P < 0.05). The inclusion of DF had a negative effect on N and fat retention coefficients at 35 d as well as AMEn and dry matter retention at 21 and 35 d. At 21 d, neutral detergent fiber (NDF) retention was increased when xylanase and STBIO were added to the diet (P < 0.001) and at d 35 the highest retention was noted when the diet was supplemented with DF and XYL or STBIO (P = 0.001). There was no dietary effect on jejunum histomorphometry (P > 0.05). The addition of DF increased the concentration of cecal SCFA in particular valeric and propionic acid at 35-day-old birds (P < 0.05). It can be concluded that addition of STBIO in diet could provide benefits in terms of fiber degradation, WG, and feed efficiency

    Scattering of Noncommutative Waves and Solitons in a Supersymmetric Chiral Model in 2+1 Dimensions

    Interactions of noncommutative waves and solitons in 2+1 dimensions can be analyzed exactly for a supersymmetric and integrable U(n) chiral model extending the Ward model. Using the Moyal-deformed dressing method in an antichiral superspace, we construct explicit time-dependent solutions of its noncommutative field equations by iteratively solving linear equations. The approach is illustrated by presenting scattering configurations for two noncommutative U(2) plane waves and for two noncommutative U(2) solitons as well as by producing a noncommutative U(1) two-soliton bound state.Comment: 1+13 pages; v2: reference added, version published in JHE
