Growth, seed development and genetic analysis in wild type and Def mutant of Pisum sativum L
<p>Abstract</p> <p>Background</p> <p>The <it>def </it>mutant pea (<it>Pisum sativum </it>L) showed non-abscission of seeds from the funicule. Here we present data on seed development and growth pattern and their relationship in predicting this particular trait in wild type and mutant lines as well as the inheritance pattern of the <it>def </it>allele in F<sub>2 </sub>and F<sub>3 </sub>populations.</p> <p>Findings</p> <p>Pod length and seed fresh weight increase with fruit maturity and this may affect the abscission event in pea seeds. However, the seed position in either the distal and proximal ends of the pod did not show any difference. The growth factors of seed fresh weight (FW), width of funicles (WFN), seed width (SW) and seed height (SH) were highly correlated and their relationships were determined in both wild type and <it>def </it>mutant peas. The coefficient of determination <it>R</it><sup>2 </sup>values for the relationship between WFN and FW, SW and SH and their various interactions were higher for the <it>def </it>dwarf type. Stepwise multiple regression analysis showed that variation of WFN was associated with SH and SW. Pearson's chi square analysis revealed that the inheritance and segregation of the <it>Def </it>locus in 3:1 ratio was significant in two F<sub>2 </sub>populations. Structural analysis of the F3 population was used to confirm the inheritance status of the <it>Def </it>locus in F<sub>2 </sub>heterozygote plants.</p> <p>Conclusions</p> <p>This study investigated the inheritance of the presence or absence of the <it>Def </it>allele, controlling the presence of an abscission zone (AZ) or an abscission-less zone (ALZ) forming in wild type and mutant lines respectively. The single major gene (<it>Def</it>) controlling this phenotype was monogenic and <it>def </it>mutants were characterized and controlled by the homozygous recessive <it>def </it>allele that showed no palisade layers in the hilum region of the seed coat.</p

Comparable outcomes among trial and nontrial participants in a clinical trial of antibiotics for childhood pneumonia: a retrospective cohort study.
OBJECTIVES: We compared characteristics and outcomes of children enrolled in a randomized controlled trial (RCT) comparing oral amoxicillin and benzyl penicillin for the treatment of chest indrawing pneumonia vs. children who received routine care to determine the external validity of the trial results. STUDY DESIGN AND SETTING: A retrospective cohort study was conducted among children aged 2-59 months admitted in six Kenyan hospitals. Data for nontrial participants were extracted from inpatient records upon conclusion of the RCT. Mortality among trial vs. nontrial participants was compared in multivariate models. RESULTS: A total of 1,709 children were included, of whom 527 were enrolled in the RCT and 1,182 received routine care. History of a wheeze was more common among trial participants (35.4% vs. 11.2%; PÂ <Â 0.01), while dehydration was more common among nontrial participants (8.6% vs. 5.9%; PÂ =Â 0.05). Other patient characteristics were balanced between the two groups. Among those with available outcome data, 14/1,140 (1.2%) nontrial participants died compared to 4/527 (0.8%) enrolled in the trial (adjusted odds ratio, 0.7; 95% confidence interval: 0.2-2.1). CONCLUSION: Patient characteristics were similar, and mortality was low among trial and nontrial participants. These findings support the revised World Health Organization treatment recommendations for chest indrawing pneumonia
Risk factors for death among children aged 5-14 years hospitalised with pneumonia: a retrospective cohort study in Kenya.
INTRODUCTION: There were almost 1âmillion deaths in children aged between 5 and 14 years in 2017, and pneumonia accounted for 11%. However, there are no validated guidelines for pneumonia management in older children and data to support their development are limited. We sought to understand risk factors for mortality among children aged 5-14 years hospitalised with pneumonia in district-level health facilities in Kenya. METHODS: We did a retrospective cohort study using data collected from an established clinical information network of 13 hospitals. We reviewed records for children aged 5-14 years admitted with pneumonia between 1 March 2014 and 28 February 2018. Individual clinical signs were examined for association with inpatient mortality using logistic regression. We used existing WHO criteria (intended for under 5s) to define levels of severity and examined their performance in identifying those at increased risk of death. RESULTS: 1832 children were diagnosed with pneumonia and 145 (7.9%) died. Severe pallor was strongly associated with mortality (adjusted OR (aOR) 8.06, 95%âCI 4.72 to 13.75) as were reduced consciousness, mild/moderate pallor, central cyanosis and older age (>9 years) (aOR >2). Comorbidities HIV and severe acute malnutrition were also associated with death (aOR 2.31, 95%âCI 1.39 to 3.84 and aOR 1.89, 95%âCI 1.12 to 3.21, respectively). The presence of clinical characteristics used by WHO to define severe pneumonia was associated with death in univariate analysis (OR 2.69). However, this combination of clinical characteristics was poor in discriminating those at risk of death (sensitivity: 0.56, specificity: 0.68, and area under the curve: 0.62). CONCLUSION: Children >5 years have high inpatient pneumonia mortality. These findings also suggest that the WHO criteria for classification of severity for children under 5 years do not appear to be a valid tool for risk assessment in this older age group, indicating the urgent need for evidence-based clinical guidelines for this neglected population

Structural and Biophysical Characterization of Staphylococcus Aureus SaMazF Shows Conservation of Functional Dynamics
The Staphylococcus aureus genome contains three toxin-antitoxin modules, including one mazEF module, SamazEF. Using an on-column separation protocol we are able to obtain large amounts of wild-type SaMazF toxin. The protein is well-folded and highly resistant against thermal unfolding but aggregates at elevated temperatures. Crystallographic and nuclear magnetic resonance (NMR) solution studies show a well-defined dimer. Differences in structure and dynamics between the X-ray and NMR structural ensembles are found in three loop regions, two of which undergo motions that are of functional relevance. The same segments also show functionally relevant dynamics in the distantly related CcdB family despite divergence of function. NMR chemical shift mapping and analysis of residue conservation in the MazF family suggests a conserved mode for the inhibition of MazF by MazE

Vavilovia (Vavilovia formosa (Stev.) Fed.) belongs to the tribe Fabeae along with peas, vetchlings, vetches and lentils. It prefers high mountain areas in Armenia, Azerbaijan, Georgia, Iran, Iraq, Lebanon, Russia, Syria and Turkey. A true success in the ex situ conservation has recently been achieved, within the display plot Flora and Vegetation of Armenia in the Yerevan Botanic Garden. The hybridization between vavilovia and other Fabeae was done in the N. I. Vavilov Institute of Plant Industry with F1 seeds and F1 plants that did not produce the next generation. The recent molecular research showed that vavilovia belongs to a Lathyrus-Pisum-Vavilovia clade with a clearly distinct status.Vavilovija (Vavilovia formosa (Stev.) Fed.) pripada tribusu Fabeae, zajedno sa graĆĄkovima, grahorima, grahoricama i soÄivima. Vavilovija raste u visokoplaninskim oblastima Jermenije, AzerbejdĆŸana, Gruzije, Iraka, Irana, Libana, Rusije, Sirije i Turske. Pravi uspeh u ex situ konzervaciji postignut je nedavno u okviru izloĆŸbenog polja 'Flora i vegetacija Jermenije' u botaniÄkoj baĆĄti u Jerevanu. Hibridizacija izmeÄu vavilovije i graĆĄka izvrĆĄena je u institutu Vavilov, sa F1 semenima i F1 biljkama koje nisu uspele da daju sledeÄe pokoljenje. SkoraĆĄnja molekularna istraĆŸivanja pokazala su da vavilovija pripada grupi Lathyrus - Pisum - Vavilovia sa jasno izraĆŸenim posebnim statusom
<em>Tendril-less</em> regulates tendril formation in pea leaves
Tendrils are contact-sensitive, filamentous organs that permit climbing plants to tether to their taller neighbors. Tendrilled legume species are grown as field crops, where the tendrils contribute to the physical support of the crop prior to harvest. The homeotic tendril-less (tl) mutation in garden pea (Pisum sativum), identified almost a century ago, transforms tendrils into leaflets. In this study, we used a systematic marker screen of fast neutronâgenerated tl deletion mutants to identify Tl as a Class I homeodomain leucine zipper (HDZIP) transcription factor. We confirmed the tendril-less phenotype as loss of function by targeting induced local lesions in genomes (TILLING) in garden pea and by analysis of the tendril-less phenotype of the t mutant in sweet pea (Lathyrus odoratus). The conversion of tendrils into leaflets in both mutants demonstrates that the pea tendril is a modified leaflet, inhibited from completing laminar development by Tl. We provide evidence to show that lamina inhibition requires Unifoliata/LEAFY-mediated Tl expression in organs emerging in the distal region of the leaf primordium. Phylogenetic analyses show that Tl is an unusual Class I HDZIP protein and that tendrils evolved either once or twice in Papilionoid legumes. We suggest that tendrils arose in the Fabeae clade of Papilionoid legumes through acquisition of the Tl gene
